β-Silyloxy Allylboronate Esters through an Aldehyde Borylation/Homologation Sequence.

Project description:The areas of carbonyl borylation and the homologation of carbon-boron bonds have provided a number of fruitful methods in organic synthesis. Combining these approaches, the homologation of α-oxyboronate esters, provides pathways to access complex organoboronate esters stereoselectively. To this end, the homologation of α-silyloxyboronate esters with lithiated allyl chlorides to form β-silyloxy allylboronate esters is reported. Direct oxidation of the homologation products provides β-silyloxy allyl alcohols in good yield. The homologation provides a range of allylic alcohols, albeit with low diastereoselectivity.

Project description:Advances on the use of the 2-pinacolacetal-tripropylphosphonium salt DualPhos as a general reagent for the two-carbon aldehyde to alkenal homologation and a chemoselective iron (III) chloride mediated deprotection are described. The strategy allows isolation of the latent alkenal intermediates or direct hydrolysis to (E)-alkenals. The robust chemical stability of the latent alkenals is demonstrated in a total synthesis of the macrolactone phomolide G.

Project description:m-Iodosylbenzoic acid performs iodinations of arenes in the presence of iodine at room temperature in acetonitrile. Separation of pure products is conveniently achieved by scavenging any aryl iodide by ion exchange with IRA-900 (hydroxide form). The reduced form of the reagent, m-iodobenzoic acid, can be easily recovered from the ion exchange resin or from the basic aqueous solution by simple acidification with HCl.

Tandem aldehyde-alkyne-amine coupling/cycloisomerization: A new synthesis of coumarins.

Project description:Cu-catalyzed A(3) coupling of ethoxyacetylene, pyrrolidine and salicylaldehydes led to a concomitant cycloisomerization followed by hydrolysis of the resultant vinyl ether to afford coumarins in a cascade process. The reaction proceeded through exclusive 6-endo-dig cyclization and is compatible with halo and keto groups giving coumarins in good to moderate yields.

Revised Mechanism for a Ruthenium-Catalyzed Coupling of Aldehyde and Terminal Alkyne.

Project description:Ruthenium catalysts have been found to be of great use for many kinds of reactions. Understanding the details of the catalytic cycle allows to not only rationalize experimental results but also to improve upon reactions. Herein, we present a detailed computational study of a ruthenium-catalyzed coupling between a terminal alkyne and an aldehyde. The reaction under examination facilitates novel access to olefins with the concurrent loss of a single carbon as carbon monoxide. The reaction was first developed in 2009, but the tentative mechanism initially proposed was proven to be contradictory to some experimental data obtained since then. Using a combination of computational investigations and isotope-labeling experiments, several potential mechanisms have been studied. In contrast to the [2+2] cycloaddition mechanism suggested for similar catalysts, we propose a new consensus pathway that proceeds through the formation of a ruthenium-vinylidene complex that undergoes an aldol-type reaction with the aldehyde to yield the product olefins. Computational insights into the influence of different reagents used to optimize reaction conditions and the intricacies of decarbonylation of a Ru-CO complex affecting catalyst turnover are highlighted.

Dimer involvement and origin of crossover in nickel-catalyzed aldehyde-alkyne reductive couplings.

Project description:The mechanism of nickel(0)-catalyzed reductive coupling of aldehydes and alkynes has been studied. Extensive double-labeling crossover studies have been conducted. While previous studies illustrated that phosphine- and N-heterocyclic carbene-derived catalysts exhibited differing behavior, the origin of these effects has now been evaluated in detail. Many variables, including ligand class, sterics of the ligand and alkyne, temperature, and ring size being formed in intramolecular versions, all influence the extent of crossover observed. A computational evaluation of these effects suggests that dimerization of a key metallacyclic intermediate provides the origin of crossover. Protocols that proceed with crossover are typically less efficient than those without crossover given the thermodynamic stability and low reactivity of the dimeric metallacycles involved in crossover pathways.

Mechanism and transition-state structures for nickel-catalyzed reductive alkyne-aldehyde coupling reactions.

Project description:The mechanism of nickel-catalyzed reductive alkyne-aldehyde coupling reactions has been investigated using density functional theory. The preferred mechanism involves oxidative cyclization to form the nickeladihydrofuran intermediate followed by transmetalation and reductive elimination. The rate- and selectivity-determining oxidative cyclization transition state is analyzed in detail. The d --> pi*(perpendicular) back-donation stabilizes the transition state and leads to higher reactivity for alkynes than alkenes. Strong Lewis acids accelerate the couplings with both alkynes and alkenes by coordinating with the aldehyde oxygen in the transition state.

Fluorosomes: a convenient new reagent to detect and block multivalent and complex receptor-ligand interactions.

Project description:We describe for the first time fluorescent virus-like particles decorated with biologically active mono- and multisubunit immune receptors of choice and the basic application of such fluorosomes (FSs) to visualize and target immune receptor-ligand interactions. For that purpose, human embryonic kidney (HEK)-293 cells were stably transfected with Moloney murine leukemia virus (MoMLV) matrix protein (MA) GFP fusion constructs. To produce FSs, interleukins (ILs), IL-receptors (IL-Rs), and costimulatory molecules were fused to the glycosyl phosphatidyl inositol anchor acceptor sequence of CD16b and coexpressed along with MoMLV group-specific antigen-polymerase (gag-pol) in MA::GFP(+) HEK-293 cells. We show that IL-2 decorated but not control-decorated FSs specifically identify normal and malignant IL-2 receptor-positive (IL-2R(+)) lymphocytes by flow cytometry. In addition to cytokines and costimulatory molecules, FSs were also successfully decorated with the heterotrimeric IL-2Rs, allowing identification of IL-2(+) target cells. Specificity of binding was proven by complete inhibition with nonlabeled, soluble ligands. Moreover, IL-2R FSs efficiently neutralized soluble IL-2 and thus induced unresponsiveness of T cells receiving full activation stimuli via T-cell antigen receptor and CD28. FSs are technically simple, multivalent tools for assessing and blocking mono- and multisubunit immune receptor-ligand interactions with natural constituents in a plasma membrane context.
