Redetermination of 4-hydroxy-benzaldehyde.

ABSTRACT: This is a redetermination of the structure of the title compound, C(7)H(6)O(2), which was first reported by Iwasaki [Acta Cryst. (1977 ?), B33, 1646-1648]. The results are obtained with greater precision in the present study. Crystal packing is stabilized by inter-molecular O-H?O inter-actions between the hydroxyl and aldehyde groups which link the mol-ecules into chains in a zigzag pattern along the [110] plane of the unit cell.

Project description:The title compound (ethyl vanillin), C(9)H(10)O(3), an important food additive and flavouring agent approved by FAO/WHO, has a vanilla odor four times that of vanillin and shows anti--mutagenic activity. There are two mol-ecules in the asymmetric unit, each having a planar conformation and an intramolecular O-H?O bond. Mol-ecules are connected side-by-side, building infinite ribbons along cvia inter-molecular O-H?O hydrogen bonds between the carbonyl and hydroxyl groups. The ribbons are then packed into layers perpendicular to the a axis.

Project description:The mol-ecule of the title compound, [Sb(C(6)H(5))(3)(C(7)H(6)NO(2))(2)], is located on a twofold axis defined by the metal center and two C atoms of a coordinated phenyl group. The Sb center has a slightly distorted trigonal-bipyramidal geometry, with the axial positions occupied by the O atoms of symmetry-related 2-hydroxy-benzaldehyde oximate ligands. An intra-molecular O-H?N inter-action is present. The crystal structure is stabilized by C-H?O inter-actions.

Project description:The mol-ecule of the title compound, C(20)H(20)N(2)O(6), lies across a crystallographic inversion centre, the asymmetric unit comprising one half-mol-ecule. An intra-molecular O-H⋯N hydrogen bond generates a six-membered ring, producing an S(6) ring motif. Pairs of inter-molecular C-H⋯O hydrogen bonds link neighbouring mol-ecules into a layer with R(2) (2)(38) ring motif. The crystal structure is further stabilized by the inter-molecular C-H⋯π inter-actions.

Redetermination and absolute configuration of 6-hydroxy-salvinolone.

Project description:THE CRYSTAL STRUCTURE OF THE TITLE COMPOUND [SYSTEMATIC NAME: 5,6,10-trihydr-oxy-7-isopropyl-1,1,4a-trimethyl-2,3,4,4a-tetra-hydro-phenanthren-9(1H)-one], C(20)H(26)O(4), has been reported previously [Salae et al. (2009 ?). Acta Cryst. E65, o2379-o2380], but the absolute configuration could not be determined as there was no significant anomalous dispersion using data collected with Mo radiation. The absolute configuration has now been determined by refinement of the Flack parameter with data collected using Cu radiation. The absolute configuration at position 4a of the diterpenoid is (R)-methyl; other features of the mol-ecule and its crystal packing are similar to those previously described.

Modeling-Assisted Design of Thermostable Benzaldehyde Lyases from Rhodococcus erythropolis for Continuous Production of α-Hydroxy Ketones.

Project description:Enantiopure α-hydroxy ketones are important building blocks of active pharmaceutical ingredients (APIs), which can be produced by thiamine-diphosphate-dependent lyases, such as benzaldehyde lyase. Here we report the discovery of a novel thermostable benzaldehyde lyase from Rhodococcus erythropolis R138 (ReBAL). While the overall sequence identity to the only experimentally confirmed benzaldehyde lyase from Pseudomonas fluorescens Biovar I (PfBAL) was only 65 %, comparison of a structural model of ReBAL with the crystal structure of PfBAL revealed only four divergent amino acids in the substrate binding cavity. Based on rational design, we generated two ReBAL variants, which were characterized along with the wild-type enzyme in terms of their substrate spectrum, thermostability and biocatalytic performance in the presence of different co-solvents. We found that the new enzyme variants have a significantly higher thermostability (up to 22 °C increase in T50 ) and a different co-solvent-dependent activity. Using the most stable variant immobilized in packed-bed reactors via the SpyCatcher/SpyTag system, (R)-benzoin was synthesized from benzaldehyde over a period of seven days with a stable space-time-yield of 9.3 mmol ⋅ L-1 ⋅ d-1 . Our work expands the important class of benzaldehyde lyases and therefore contributes to the development of continuous biocatalytic processes for the production of α-hydroxy ketones and APIs.

Crystal structure of a new monoclinic polymorph of 2,4-di-hydroxy-benzaldehyde 4-methyl-thio-semi-carbazone.

Project description:The title compound, C9H11N3O2S, is a second monoclinic (P21/c) polymorph of the previously reported Cc form [Tan et al. (2008b ▶). Acta Cryst. E64, o2224]. The mol-ecule is non-planar, with the dihedral angle between the N3CS residue (r.m.s. deviation = 0.0816 Å) and the benzene ring being 21.36 (4)°. The conformation about the C=N bond [1.292 (2) Å] is E, the two N-bound H atoms are anti, and the inner hy-droxy O-bound and outer amide N-bound H atoms form intra-molecular hydrogen bonds to the imine N atom. Crucially, the H atom of the outer hy-droxy group is approximately syn to the H atom of the benzene C atom connecting the two C atoms bearing the hy-droxy substituents. This arrangement enables the formation of supra-molecular tubes aligned along [010] and sustained by N-H⋯O, O-H⋯S and N-H⋯S hydrogen bonds; the tubes pack with no specific inter-actions between them. While the mol-ecular structure in the Cc form is comparable, the H atom of the outer hy-droxy group is approximately anti, rather than syn. This different orientation leads to the formation a three-dimensional architecture based on N-H⋯O and O-H⋯S hydrogen bonds.

Project description:The crystal structure of the title compound (common name HEPES), C(8)H(18)N(2)O(4)S, has been redetermined at 100 K in order to properly elucidate the protonation state of the HEPES molecule. The piperazine ring has a chair conformation and one of the N atoms in the ring is protonated, which was not previously reported [Gao, Yin, Yang, & Xue (2004). Acta Cryst. E60, o1328-o1329]. The change of protonation state of the nitrogen atom significantly affects the intermolecular interactions in the HEPES crystal. The structure is stabilized by N-H⋯O and O-H⋯O hydrogen bonds and ionic inter-actions, as the title compound in solid state is a zwitterion. HEPES mol-ecules pack in layers that are held together by ionic and weak inter-actions, while a hydrogen-bonded network connects the layers.

Project description:The title Schiff base, C(11)H(14)ClNO(4), originally refined as a neutral mol-ecule [Chumakov, Antosyak, Mazus, Tsapkov & Samus (2000), Crystallogr. Rep.45, 945-950], is inter-preted as a zwitterionic compound. There are two independent zwitterions in the asymmetric unit. Five of the six hydr-oxy groups of the -CH(2)OH fragments are disordered. In one zwitterion, two are disordered over three sites [0.53 (1):0.33 (1):0.14 (1) and 0.65 (1):0.18 (1):0.17 (1)] and the third over two sites [0.66 (1):0.34 (1)]. In the second zwitterion, two are disordered over two sites [0.84 (1):0.16 (1) and 0.83 (1):0.17 (1)] and the third hydr-oxy group is ordered.
