The Na atoms are disordered over three sites [occupancies 0.775?(4), 0.781?(6) 0.444?(6)].

Project description:The non-receptor tyrosine kinase Syk has a diverse range of biological functions, including a critical role in the intracellular signalling cascade for the surface immunoglobulin receptor on B lymphocytes, and the Fc receptor expressed on numerous immune effector cells. It is therefore seen as a potential therapeutic target in a variety of conditions, including autoimmune, allergic and malignant diseases. Fostamatinib disodium is the orally bioavailable prodrug of R406, a relatively selective small molecule inhibitor of Syk, that has accordingly shown activity in numerous cell types in vitro, and efficacy in a remarkable range of animal models in vivo, including rodent models of asthma, inflammatory arthritis, lupus, glomerulonephritis, diabetes and lymphoma. Success in these models has translated to phase II clinical trials in autoimmune thrombocytopenia, lymphoma and, most notably, rheumatoid arthritis, in which larger phase III trials are currently in progress. Whilst the diverse biological functions of Syk, coupled to the potential off-target effects of this kinase inhibitor are a source of possible toxicity, the available data thus far augurs well for future clinical use of Fostamatinib in a wide range of human diseases.

Project description:The asymmetric unit of the anhydrous title compound, 2Na(+)·C(4)H(4)O(6) (2-), contains two sodium cations and one tartrate anion. Each sodium ion is six coordinate, with bonding to six O atoms from both the carboxyl-ate and hydroxyl groups of the anion. A three-dimensional coordination network is formed with sodium ions stacking in layers along the c-axis direction. This network is supported by additional O-H?O hydrogen bonds.

Project description:The asymmetric unit of the title compound, Na(2)(MoO(2))(3)(As(2)O(7))(2), is composed of two cyclic MoAs(2)O(11) units and an MoO(6) corner-sharing octa-hedron. The anionic framework can be decomposed into two types of layers, viz. MoO(2)As(2)O(7) and Mo(2)As(2)O(14), which use mixed Mo-O-As and As-O-Mo bridges to achieve a new three-dimensional structure with two types of large channels in which the Na(+) cations are located. Two O atoms are disordered and are located in two positions close to their initial positions with occupancy ratios of 0.612?(17):0.388?(17) and 0.703?(12):0.298?(12).

Project description:The title compound {systematic name: poly[[?(7)-(2R)-2-amino-3-sulfonato-propano-ato]disodium]}, [Na(2)(C(3)H(5)NO(5)S)](n), was obtained through solvent-thermal reaction of l-cysteic acid and aqueous sodium hydroxide. The monomer consists of two Na(+) cations that are coordinated to the deprotonated amino acid. The latter acts as donor utilizing all available coordination sites, viz. the amino, the carboxyl-ate and the sulfonate residues, so producing a monomeric framework in which the two coordinated Na(+) ions have different coordination spheres and geometries. One of the Na(+) ions has an O(5) coordination sphere with a typical geometric arrangement, inter-mediate between trigonal-bipyramidal and square-pyramidal; all the O atoms from the amino acid (three from the sulfonate and two from the caboxylate residues) act as donors. The second Na(+) ion is tetracoordinated within an NO(3) coordination sphere. The Na(+) ion binds to the amino N atom, to one of the O atom of the carb-oxy-lic residue and to two O atoms of the sulfonate group in a distorted tetra-hedral arrangement. As the sulfonate O atoms bind to both Na(+) ions, a three-dimensional polymeric framework is obtained.

Project description:In the title compound, [FeNa(2)(SO(4))(2)(H(2)O)(4)], the Fe(2+) cation is situated on a centre of inversion and is hexa-coordinated by four O atoms from water mol-ecules and two O atoms from two sulfate anions in an octa-hedral geometry. The coordination environment of Na(+) comprises six O atoms in a more distorted octa-hedral arrangement, with Na-O distances between 2.368 (1) and 2.612 (1) Å. The structure contains an extensive three-dimensional network of O-H⋯O hydrogen bonds.

Project description:The crystal structure of disodium hydrogen citrate sesquihydrate, 2Na2 (+)·C6H6O7 (2-)·1.5H2O, has been solved and refined using laboratory X-ray single-crystal diffraction data, and optimized using density functional techniques. The asymmetric unit contains two independent hydrogen citrate anions, four sodium cations and three water molecules. The coordination polyhedra of the cations (three with a coordination number of six, one with seven) share edges to form isolated 8-rings. The un-ionized terminal carb-oxy-lic acid groups form very strong hydrogen bonds to non-coordinating O atoms, with O⋯O distances of 2.46 Å.

Carbenoxolone Disodium Treatment for Canine Pituitary-Dependent Hyperadrenocorticism.

Project description:Pituitary-dependent hyperadrenocorticism (PDH) is mainly caused by pituitary corticotroph tumors in dogs. A characteristic feature of corticotroph tumors is their resistance to negative feedback by glucocorticoids. In some animal species, including dogs, the aberrant expression of 11?-hydroxysteroid dehydrogenase (11HSD), a cortisol metabolic enzyme, is observed in corticotroph tumors. We previously reported that carbenoxolone (CBX), an inhibitor of 11HSD, suppressed ACTH secretion from the pituitary gland, and decreased cortisol concentrations in healthy dogs. Therefore, the aim of this study was to investigate the therapeutic effects of CBX on dogs with PDH. Six dogs with PDH were treated with 60 to 80 mg/kg/day of CBX for 6 weeks, followed by trilostane, which is a commonly used agent for canine PDH. CBX treatment led to a gradual decrease in both basal and in corticotropic releasing hormone (CRH)-stimulated plasma ACTH concentrations and CRH-stimulated serum cortisol concentrations, without side effects. However, basal and stimulated ACTH and cortisol concentrations remained higher than those of healthy dogs, and clinical symptoms such as polydipsia and polyuria were not ameliorated. After a 2-week wash-out interval, trilostane was administered for 2 weeks. Although basal plasma ACTH concentrations were higher after trilostane treatment than CBX treatment, polydipsia and polyuria resolved in all six dogs. The reason for the lack of improvement in polydipsia and polyuria with CBX treatment is unclear. Other mechanisms, in addition to a partial decrease in ACTH secretion, are likely to be involved. In conclusion, this is the first study to report the in vivo effects of CBX in dogs with PDH. The findings suggest that CBX inhibits ACTH secretion from canine pituitary tumors, resulting in lower cortisol concentrations.

Project description:In the title compound, 2Na(+)·C(9)H(3)NO(8) (2-)·3H(2)O, the asymmetric unit consists of two Na(+) cations, one dihydrogen pyridine-2,3,5,6-tetra-carboxyl-ate dianion (H(2)pdtc(2-)) and three water mol-ecules coordinated to the Na(+) cations. The configuration of the anion is stabilized by intramolecular O-H?O hydrogen bonding between vicinal carboxylate/carboxy groups. The Na(+) cations are bridged by the H(2)pdtc(2-) dianions, generating layers extending infinitely in sheets parallel to (001), and further pillared by the water mol-ecule linkers to build up a three-dimensional framework.

Calcium disodium hexa-thio-diphosphate(IV) octa-hydrate.

Project description:Single crystals of the title compound, CaNa(2)(P(2)S(6))·8H(2)O, were obtained by adding calcium hydroxide to an aqueous solution of Na(4)(P(2)S(6))·6H(2)O. The structure is isotypic with that of its strontium analogue and consists of one Ca(2+) cation, two Na(+) cations, one-half of a centrosymmetric (P(2)S(6))(4-) anion with staggered confirmation and four water mol-ecules in the asymmetric unit. The crystal structure can be described as being built up from layers of cations and anions extending parallel to (101). Within a layer, each CaO(8) polyhedron is connected via edge-sharing to two NaO(4)S(2) octa-hedra and to one NaO(2)S(4) octa-edron. The NaO(4)S(2) octa-hedra are, in turn, linked with two (P(2)S(6))(4-) anions through common corners. Various O-H?S hydrogen-bonding inter-actions lead to cohesion of adjacent layers. The Ca(2+) and one Na(+) cation are situated on a twofold rotation axis and the second Na(+) cation is situated on an inversion centre.

Project description:The asymmetric unit of the title compound, [Na(2)(C(8)H(8)O(8))](n), contains one Na(+) ion and half of a 2,3-bis(carboxymethyl)butanedioate (H(2)BTC(2-)) dianion, which lies on a center of symmetry. The dianion exhibits a μ(10)-bridging mode. Each Na atom lies in a NaO(6) octa-hedron defined by six O atoms from five dianions. Adjacent NaO(6) octa-hedra share a common O-O edge, generating a biocta-hedron; adjacent biocta-hedra are O-O edge-connected to one another, building up a chain along [001]. The chains are connected by adjacent H(2)BTC(2-) anions into a three-dimensional framework. The structure is further stabilized by O-H⋯O hydrogen bonds.
