7-Acetoxy-cochinchinone I.

ABSTRACT: The title compound {systematic name: 12-[(2E)-3,7-dimethyl-2,6-octa-dien-yl]-5,8-dihydr-oxy-2,2-dimethyl-2H,6H-pyrano[3,2-b]xanthen-6-one}, C(30)H(32)O(6), has four fused rings (A/B/C/D) and the xanthone ring system (A/B/C) is essentially planar, with dihedral angles of 1.85?(13) and 2.47?(13)°, respectively, between rings A and B, and between rings B and C. The chromene ring D is in a sofa form. The geranyl side chain is axially attached to ring C with an (-)-synclinal conformation. The 3-methyl-2-butenyl terminal of the geranyl side chain is disordered with the site-occupancy ratio of 0.513?(5):0.487?(5). The acet-oxy group is attached axially to ring A with an (+)-synclinal conformation. An intra-molecular O-H?O hydrogen bond involving the carbonyl and hydroxyl groups generates an S(6) ring motif. In the crystal, weak C-H?O and C-H?? inter-actions, and ?-? inter-actions with centroid-centroid distances of 3.6562?(16) and 3.6565?(16)?Å are observed.

Project description:THE TITLE COMPOUND [SYSTEMATIC NAME: (1S,3aS,4aR,4bS,5S,6R,6aR,10aR,10bR,12aS)-5,6-bis-(acet-yloxy)-1-(3-fur-yl)-1,5,6,6a,7,10a,10b,11,12,12a-deca-hydro-4b,7,7,10a,12a-penta-methyl-oxireno[c]phenanthro[1,2-d]pyran-3,8(3aH,4bH)-dione], C(30)H(36)O(9), is a limonoid-type triterpene isolated from Aglaia elaeagnoidea (A. Juss.) Benth. (Meliaceae) from Queensland, northern Australia. It contains the gedunin core of four trans-fused six-membered rings with an oxirane ring annelated to the fourth ring. A terminal 3-furyl unity and two acet-oxy groups in a mutual cis-disposition supplement the mol-ecule. A comparison between the gedunin cores of the title compound, the parent compound gedunin, and three further gedunin derivatives revealed considerable variations in their conformation stemming from the conformational lability of the first screw-boat ring and the third twist-boat ring. A sensitive measure for the third ring is one C-C-C-C torsion angle, which is 14.2?(2)° in the title compound, but varies in other cases from ca 20 to ca -40°. In the crystalline state, 6?-acetoxy-gedunin shows ten comparatively weak C-H?O inter-actions, with H?O distances in the range of 2.33-2.69?Å.

Project description:A study of cyclopropanations of oxy-substituted ethenes with ethyl α-diazoaroyl acetates is described. Whereas trimethylsilyl vinyl ether and ethyl vinyl ether gave dihydrofuran products, stable cyclopropanes were isolated when vinyl acetate was used. Yields ranged from 0-87%, depending on the nature and position of the aryl ring substituent.

Identification of adducts formed by reaction of N-acetoxy-3,5-dimethylaniline with DNA.

Project description:Aromatic amines constitute one of the most extensively studied classes of chemical carcinogens. Although monocyclic aromatic amines are generally regarded as weak carcinogens, a recent epidemiologic study of bladder cancer found that the arylamine 3,5-dimethylaniline (3,5-DMA) may play a significant role in the etiology of this disease in man. Investigations using experimental animals also strongly suggested that DNA adducts-of indeterminate structure-formed by 3,5-DMA might account for its presumptive activity. The present study was undertaken to determine the structures of the major DNA adducts formed in vitro by the known, and possibly carcinogenic, N-hydroxylated metabolite. Calf thymus DNA (ct-DNA) was modified by reaction with N-acetoxy-3,5-dimethylaniline (N-AcO-3,5-DMA). After enzymatic hydrolysis of DNA to individual 2'-deoxyribonucleosides, adduct profiles were determined using HPLC/MS. 3,5-DMA formed four major DNA adducts, one to 2'-deoxyguanosine (dG), two to 2'-deoxyadenosine (dA), and one to 2'-deoxycytidine (dC). Reactions of N-AcO-3,5-DMA with dG, dA, and dC produced the same adducts as reaction with ct-DNA with very similar profiles. Adducts were isolated chromatographically and unambiguously characterized as N-(deoxyguanosin-8-yl)-3,5-dimethylaniline (dG-C8-3,5-DMA), 4-(deoxyadenosin- N(6)-yl)-3,5-dimethylaniline (dA- N(6)-3,5-DMA), N-(deoxyadenosin-8-yl)-3,5-dimethylaniline (dA-C8-3,5-DMA), and N-(deoxycytidin-5-yl)-3,5-dimethylaniline (dC-C5-3,5-DMA) by high-resolution mass spectra (HR-MS) and NMR spectroscopy including (1)H NMR, (13)C NMR, and two-dimensional NMR. This report includes the first detailed description of a dC adduct of an aromatic amine. The present results provide chemical support for a carcinogenic mechanism of action by 3,5-DMA based on N-hydroxylation and the intermediacy of a nitrenium ion in the formation of DNA adducts.

Enzymatic deprotection of the cephalosporin 3'-acetoxy group using Candida antarctica lipase B.

Project description:Cephalosporins remain one of the most important classes of antibiotics. A useful site for derivatization involves generation of and chemistry at the 3'-hydroxymethyl position. While 3'-acetoxymethyl-substituted cephalosporins are readily available, deacetylation to access the free 3'-hydroxymethyl group is problematic when the carboxylic acid is protected as an ester. Herein we report that this important transformation has been efficiently accomplished using Candida antarctica lipase B. Although this transformation is difficult to carry out using chemical methods, the enzymatic deacetylation has been successful on gram scale, when the cephalosporin is protected as either the benzhydryl or tert-butyl esters and on the corresponding sulfoxide and sulfone of the tert-butyl ester.

Synthesis of polymer-bound 4-acetoxy-3-phenylbenzaldehyde derivatives: applications in solid-phase organic synthesis.

Project description:Aminomethyl polystyrene resin was reacted with 4-(5'-formyl-2'-hydroxyphenyl)benzoic acid and 4-(5'-formyl-2'-hydroxyphenyl)phenyl propionic acid, respectively, in the presence of 1-hydroxybenzotriazole and 1,3-diisopropylcarbodiimide to yield polymer-bound benzaldehydes. The phenolic group in resins was acetylated with acetic anhydride to afford two polymer-bound 4-acetoxybenzaldehydes. The reductive amination of polymer-bound linkers by amines and sodium triacetoxyborohydride, followed by sulfonylation with arylsulfonyl chloride derivatives in the presence of pyridine and the cleavage with TFA/DCM/H2O, produced pure sulfonamides.
