ABSTRACT: In the title compound, C(14)H(10)O(2), the seven-membered oxepine ring adopts a twist-boat conformation with a dihedral angle between the mean planes of the two fused benzene rings of 42.0?(1)°. In the crystal, mol-ecules are linked into chains propagating along the c axis by inter-molecular C-H?O hydrogen bonds and the chains are arranged in layers parallel to (100).

Project description:In the title compound, C(23)H(21)NO(5), the two benzene rings of the tricyclic unit are oriented at a dihedral angle of 37.5 (8)°. The 2,4-dimeth-oxy-anilino residue is oriented at a dihedral angle of 60.2 (8)° towards the phen-oxy ring. In the crystal, the central carbonyl O atom accepts two hydrogen bonds from the N-H and C-H groups. A further inter-molecular C-H⋯O inter-action involving one of the meth-oxy O atoms is also observed.

Project description:There are two independent mol-ecules (A and B) in the asymmetric unit of the title compound, C(19)H(23)NOS. In each mol-ecule, the seven-membered thiepine ring is bent into a slightly twisted V-shape. The dihedral angles between the mean planes of the two benzene rings fused to the thiepine ring are 75.7?(5) in mol-ecule A and 73.8?(4)° in mol-ecule B. In both mol-ecules, an intra-molecular O-H?N hydrogen bond occurs. In the crystal, weak inter-molecular C-H?O and C-H??-ring inter-actions are observed.

The 2,4-difluoro-anilino residue is oriented at a dihedral angle of 68.2 (1)° with respect to the phen-oxy ring. In the crystal, N-H⋯O hydrogen bonds between the amino group and the carbonyl O atom of the oxepinone ring link the mol-ecules into infinte chains along the c axis.

New short strategy for the synthesis of the dibenz[b,f]oxepin scaffold.

Project description:In this report a short and efficient synthesis of the dibenz[b,f]oxepin framework through intramolecular SNAr and McMurry reactions is described. The diaryl ethers required for the McMurry reaction have been obtained in good yields under microwave-assisted conditions of the reaction of salicylaldehydes with fluorobenzaldehydes without catalysts. Application of an intramolecular McMurry reaction to the synthesized diarylethers using TiCl4/Zn in THF gave the target dibenzo[b,f]oxepin system in 53%-55% yields.

Project description:The title compound, C(22)H(18)O(7), also known as laurentiquinone B, is a new anthraquinone which was isolated from Vismia laurentii, a Cameroonian medicinal plant. The asymmetric unit contains two independent mol-ecules. Each of them contains four fused rings, three of which are coplanar and typical of anthracene, while the heterocyclic rings adopt envelope conformations. Intra-molecular O-H⋯O hydrogen bonds result in the formation of two planar rings, which are also almost coplanar with the adjacent rings. In the crystal structure, inter-molecular O-H⋯O and C-H⋯O hydrogen bonds link the mol-ecules and a π-π contact is also present [centroid-centroid distance = 3.967 (3) Å].

The Discovery of Novel 10,11-Dihydro-5H-dibenz[b,f]azepine SIRT2 Inhibitors.

Project description:Isoform selective inhibitors of the sirtuins (NAD+-dependent histone deacetylases) should enable an in depth study of the molecular biology underpinning these targets and how they are deregulated in diseases such as cancer and neurodegeneration. Herein, we present the discovery of structurally novel SIRT2 inhibitors. Hit molecule 8 was discovered through the chemical synthesis and biological characterization of a small-molecule compound library based around the 10,11-dihydro-5H-dibenz[b,f]azepine scaffold. In vitro screening assays revealed compound 8 to have an IC50 of 18 μM against SIRT2 and to exhibit more than 30-fold selectivity compared to SIRT1. Cellular assays, performed on MCF-7 cells, confirmed the in vitro selectivity and showed hit 8 to have antiproliferative activity at a concentration of 30 μM. Computational studies were performed to predict the SIRT2 binding mode and to rationalise the observed selectivity.
