Strongly Phosphorescent Iridium(III)-Porphyrins - New Oxygen Indicators with Tuneable Photophysical Properties and Functionalities.

ABSTRACT: Synthesis and characterization of four iridium(III)-octaethylporphyrins and a ?-extended iridium(III)-benzoporphyrin are presented. Strong room-temperature phosphorescence was observed for all of the complexes with quantum yields of up to 30 %. Axial ligands were introduced to tune the photophysical properties and the solubility. Complexes bearing lipophilic ligands such as pyridine or N-(n-butyl)imidazole were incorporated into polystyrene to obtain optical oxygen sensors. Covalent coupling of the dye is possible by introduction of ligands with binding domains (1-imidazoleacetic acid). This enabled preparation of a water-soluble oxygen probe (by staining bovine serum albumin) and a trace oxygen sensor (by coupling to amino-modified silica gel).

Tuning the Photophysical and Excited State Properties of Phosphorescent Iridium(III) Complexes by Polycyclic Unit Substitution.

Project description:Two novel N-embedded polycyclic units functionalized phosphorescent iridium(III) complexes (Ir-1 and Ir-2) with substituents in different positions have been prepared. Complex Ir-1 bearing the substituent at the 3-position shows a distinct blue shift single-peak emission (524 nm) with a higher luminescence efficiency (ΦPL=42 %) and shorter emission lifetime (τ=282 ns) by comparison with 4-position substitution based complex Ir-2 (ΦPL=23 %, τ=562 ns), which exhibits a dual-peak emission (564 nm and 602 nm), and phosphorescence color can be tuned from green to yellow. In addition, DFT calculations demonstrate that unusual ligand-to-metal charge transfer (3LMCT) excited state property can be found in Ir-2, which is in contrast to metal-to-ligand charge transfer (3MLCT) excited state character in Ir-1. This result can be attribute to strong electron-donating character and 4-position substitution effect of the unit.

Mitochondrial Dynamics Tracking with Two-Photon Phosphorescent Terpyridyl Iridium(III) Complexes.

Project description:Mitochondrial dynamics, including fission and fusion, control the morphology and function of mitochondria, and disruption of mitochondrial dynamics leads to Parkinson's disease, Alzheimer's disease, metabolic diseases, and cancers. Currently, many types of commercial mitochondria probes are available, but high excitation energy and low photo-stability render them unsuitable for tracking mitochondrial dynamics in living cells. Therefore, mitochondrial targeting agents that exhibit superior anti-photo-bleaching ability, deep tissue penetration and intrinsically high three-dimensional resolutions are urgently needed. Two-photon-excited compounds that use low-energy near-infrared excitation lasers have emerged as non-invasive tools for cell imaging. In this work, terpyridyl cyclometalated Ir(III) complexes (Ir1-Ir3) are demonstrated as one- and two-photon phosphorescent probes for real-time imaging and tracking of mitochondrial morphology changes in living cells.

Targeting cancer cell metabolism with mitochondria-immobilized phosphorescent cyclometalated iridium(iii) complexes.

Project description:Cancer cell metabolism is reprogrammed to sustain the high metabolic demands of cell proliferation. Recently, emerging studies have shown that mitochondrial metabolism is a potential target for cancer therapy. Herein, four mitochondria-targeted phosphorescent cyclometalated iridium(iii) complexes have been designed and synthesized. Complexes 2 and 4, containing reactive chloromethyl groups for mitochondrial fixation, show much higher cytotoxicity than complexes 1 and 3 without mitochondria-immobilization properties against the cancer cells screened. Further studies show that complexes 2 and 4 induce caspase-dependent apoptosis through mitochondrial damage, cellular ATP depletion, mitochondrial respiration inhibition and reactive oxygen species (ROS) elevation. The phosphorescence of complexes 2 and 4 can be utilized to monitor the perinuclear clustering of mitochondria in real time, which provides a reliable and convenient method for in situ monitoring of the therapeutic effect and gives hints for the investigation of anticancer mechanisms. Genome-wide transcriptional analysis shows that complex 2 exerts its anticancer activity through metabolism repression and multiple cell death signalling pathways. Our work provides a strategy for the construction of highly effective anticancer agents targeting mitochondrial metabolism through rational modification of phosphorescent iridium complexes.

Photophysical and Photobiological Properties of Dinuclear Iridium(III) Bis-tridentate Complexes.

Project description:A series of cationic dinuclear iridium(III) complexes (Ir1-Ir5) bearing terpyridine-capped fluorenyl bridging ligands and different polypyridyl or cyclometalating terminal tridentate ligands were synthesized, characterized, and evaluated for their photophysical and photobiological activities. The influence of the bridging and terminal ligands on the photophysical properties of the complexes was investigated by UV-vis absorption, emission, and transient absorption spectroscopy and simulated by TDDFT calculations. All of the complexes displayed strong bridging-ligand localized visible 1π,π* absorption and red- or near-infrared phosphorescence as well as broad triplet excited-state absorption across both visible and NIR wavelengths. These triplet states were assigned as predominantly 3π,π* for Ir1 (τ = 3.1 μs) and Ir4 (τ = 48 μs) and 3CT (charge transfer) for Ir2, Ir3, and Ir5 (τ = 1.7-2.7 μs). Complexes Ir1-Ir5 acted as in vitro photodynamic therapy (PDT) agents toward human SK-MEL-28 melanoma cells when activated with visible light, with submicromolar photocytotoxicity and phototherapeutic indices ranging from 20 to almost 300. The in vitro PDT effects with visible light did not correlate with singlet oxygen (1O2) quantum yields or DNA photocleaving capacity probed under cell-free conditions. All of the Ir(III) complexes phosphoresced brightly when associated with compromised cells (with or without light treatment) and exhibited photoactivated cellular uptake, highlighting the theranostic potential of this new class of Ir(III) complex photosensitizers.

Tunable phosphorescent NIR oxygen indicators based on mixed benzo- and naphthoporphyrin complexes.

Project description:A series of ?-extended phosphorescent palladium(II) and platinum(II) porphyrin complexes were synthesized, in which additional benzene rings are fused radially onto at least one of the four peripheral benzo groups. The photophysical properties of the metalloporphyrins palladium(II)-meso-tetra-(4-fluorophenyl)mononaphthotribenzoporphyrin (Pd1NF), cis-palladium(II)-meso-tetra-(4-fluorophenyl)dibenzodinaphthoporphyrin (Pd2NF), and palladium(II)-meso-tetra-(4-fluorophenyl)monobenzotrinaphthoporphyrin (Pd3NF) and the corresponding platinum(II) compounds (Pt1NF, cis-Pt2NF, Pt3NF) were investigated. The compounds under investigation absorb intensively in the near-infrared region (628-691 nm) and emit at room temperature at 815-882 nm. Phosphorescence quantum yields of the platinum(II) porphyrins range from 25 to 53% with luminescence decay times of 21 to 44 ?s in deoxygenated toluene solutions at room temperature. The corresponding palladium(II) complexes exhibit quantum yields in the range of 7 to 18% with lifetimes of 106 to 206 ?s. Density functional theory (DFT) calculations revealed nonplanar geometries for all complexes and corroborate the absorption characteristics. The subsequent ? extension of the porphyrin system leads to near-infrared absorbing oxygen indicators with tailor-made luminescence properties as well as tunable oxygen sensitivity.

A Phosphorescent Iridium(III) Complex-Modified Nanoprobe for Hypoxia Bioimaging Via Time-Resolved Luminescence Microscopy.

Project description:Oxygen plays a crucial role in many biological processes. Accurate monitoring of oxygen level is important for diagnosis and treatment of diseases. Autofluorescence is an unavoidable interference in luminescent bioimaging, so that an amount of research work has been devoted to reducing background autofluorescence. Herein, a phosphorescent iridium(III) complex-modified nanoprobe is developed, which can monitor oxygen concentration and also reduce autofluorescence under both downconversion and upconversion channels. The nanoprobe is designed based on the mesoporous silica coated lanthanide-doped upconversion nanoparticles, which contains oxygen-sensitive iridium(III) complex in the outer silica shell. To image intracellular hypoxia without the interferences of autofluorescence, time-resolved luminescent imaging technology and near-infrared light excitation, both of which can reduce autofluorescence effectively, are adopted in this work. Moreover, gradient O2 concentration can be detected clearly through confocal microscopy luminescence intensity imaging, phosphorescence lifetime imaging microscopy, and time-gated imaging, which is meaningful to oxygen sensing in tissues with nonuniform oxygen distribution.

Phosphorescent cationic iridium(iii) complexes dynamically bound to cyclodextrin vesicles: applications in live cell imaging.

Project description:We report cationic Ir(iii) complexes functionalized with adamantyl groups designed to bind to β-cyclodextrin vesicles (CDV) with high affinity (K a = 1 × 106 M-1). The emission of the complexes is tuned by changing the nature of the cyclometalating ligands. The host-guest adduct of CDV and Ir(iii) complexes shows increased and significantly blue-shifted emission due to the lower mobility of the Ir(iii)-complexes residing in the less polar environment of the vesicle surface. Ir(iii)-decorated CDV are efficiently taken up by cells and can be used in live cell imaging. The CDV act as carriers to transport the phosphorescent complexes into cells where they selectively stain mitochondria.

Ratiometric Molecular Probes Based on Dual Emission of a Blue Fluorescent Coumarin and a Red Phosphorescent Cationic Iridium(III) Complex for Intracellular Oxygen Sensing.

Project description:Ratiometric molecular probes RP1 and RP2 consisting of a blue fluorescent coumarin and a red phosphorescent cationic iridium complex connected by a tetra- or octaproline linker, respectively, were designed and synthesized for sensing oxygen levels in living cells. These probes exhibited dual emission with good spectral separation in acetonitrile. The photorelaxation processes, including intramolecular energy transfer, were revealed by emission quantum yield and lifetime measurements. The ratios (R(I) = (I(p)/I(f))) between the phosphorescence (I(p)) and fluorescence (I(f)) intensities showed excellent oxygen responses; the ratio of R(I) under degassed and aerated conditions ( R(I)(0) was 20.3 and 19.6 for RP1 and RP2. The introduction of the cationic Ir (III) complex improved the cellular uptake efficiency compared to that of a neutral analogue with a tetraproline linker. The emission spectra of the ratiometric probes internalized into living HeLa or MCF-7 cells could be obtained using a conventional microplate reader. The complex RP2 with an octaproline linker provided ratios comparable to the ratiometric measurements obtained using a microplate reader: the ratio of the R(I)) value of RP2 under hypoxia (2.5% O2) to that under normoxia (21% O2) was 1.5 and 1.7 for HeLa and MCF-7 cells, respectively. Thus, the intracellular oxygen levels of MCF-7 cells could be imaged by ratiometric emission measurements using the complex RP2.

Cyclometalated Iridium(III) Complexes as AIE Phosphorescent Probes for Real-Time Monitoring of Mitophagy in Living Cells.

Project description:Mitophagy, which is a special autophagy that removes damaging mitochondria to maintain sufficient healthy mitochondria, provides an alternative path for addressing dysfunctional mitochondria and avoiding cellular death. In the present study, by coupling the triphenylamine group with 2-phenylimidazo[4,5-f][1,10]phenanthroline derivatives, we synthesized five Ir(III) complexes with an AIE property that are expected to fulfill requirements for real-time monitoring of mitophagy. Ir1-Ir5 were exploited to image mitochondria with a short incubation time by confocal microscopy and inductive coupled plasma-mass spectrometry (ICP-MS). Due to aggregation-induced emission (AIE), Ir1-Ir5 exhibited excellent photostability compared to MitoTracker Green (MTG). Moreover, Ir1-Ir5 manifested satisfactory photostability in the mitochondrial physiological pH range. In addition, the uptake mechanism of Ir1 was investigated using confocal microscopy and flow cytometry analysis. Finally, using both Ir1 and LysoTracker Green, we were able to achieve real-time monitoring of mitophagy.
