Facile Synthesis of Mutagen X (MX): 3-Chloro-4-(dichloromethyl)-5-hydroxy-5H-furan-2-one.

ABSTRACT: 3-Chloro-4-(dichloromethyl)-5-hydroxy-5H-furan-2-one (Mutagen X, MX) was synthesized in six steps from commercially-available and inexpensive starting materials (27% overall yield). This synthesis enables the preparation of MX analogs and does not require the use of chlorine gas, as do previously reported methods.

Project description:The title compound, C(16)H(12)ClNO(2), featuring a furan-2(5H)-one (?-butyrolactone) core, contains two mol-ecules (A and B) in the asymmetric unit, with different dihedral angles between the central ring and the pendant phenyl and chloro-benzene rings [43.33?(8) and 20.16?(8)°, respectively, for A, and 47.79?(8) and 13.87?(8)°, respectively, for B]. In the crystal, the A mol-ecules are linked into [001] chains by single C-H?O inter-actions. The B mol-ecules also form [001] chains, but their relative orientations in the chains are quite different to those of the A mol-ecules so that adjacent B mol-ecules are linked by two C-H?O hydrogen bonds. Finally, C-H?O inter-actions and aromatic ?-? stacking contacts [centroid-centroid separations = 3.754?(1) and 3.817?(1)?Å] link the chains into a two-dimensional array parallel to (010).

Project description:In the mol-ecule of the title compound, C(27)H(21)ClN(2)O(4), the dihedral angle between the furan rings is 67.00?(3)°. The chloro-phenyl ring is oriented at dihedral angles of 76.61?(3) and 69.36?(3)° with respect to the furan rings. An intra-molecular N-H?O inter-action results in the formation of an eight-membered ring with a twisted conformation. In the crystal structure, inter-molecular N-H?O and C-H?O inter-actions link the mol-ecules into a three-dimensional network, forming R(2) (2)(16) ring motifs. Three weak C-H?? inter-actions are also found.

Project description:The crystal structure and solid-state packing of 4-chloro-5H-1,2,3-dithiazol-5-one and two polymorphs of 4-chloro-5H-1,2,3-dithiazole-5-thione were analyzed and compared to structural data of similar systems. These five-membered S,N-rich heterocycles are planar with considerable bond localization. All three structures demonstrate tight solid-state packing without voids which is attributed to a rich network of short intermolecular electrostatic contacts. These include Sδ+…Nδ-, Sδ+…Oδ-, Sδ+…Clδ- and Sδ+…Sδ- interactions that are well within the sum of their van der Waals radii (∑VDW). B3LYP, BLYP, M06, mPW1PW, PBE and MP2 were employed to calculate their intramolecular geometrical parameters, the Fukui condensed functions to probe their reactivity, the bond order, Bird Index and NICS(1) to establish their aromaticity.
