Stenosternus Karsch, a possible link between Neotropical and Afrotropical Orphninae (Coleoptera, Scarabaeidae).

ABSTRACT: The monotypical orphnine genus Stenosternus Karsch is known from a single specimen of Stenosternus costatus collected on the São Tomé island (Gulf of Guinea). The holotype of Stenosternus costatus Karsch is re-examined and its characters are discussed and illustrated. Although the genus was implicitly placed by Paulian (1984) in the Old World tribe Orphnini Erichson, re-examination of the holotype of Stenosternus costatus shows that it has characters similar to those of the members of the New World tribe Aegidiini Paulian. Placement of Stenosternus in the Aegidiini is supported by the metepisternum widened posteriorly (forming posterior metepisternal lock for closed elytra) and a keel separating basal and anterolateral parts of propleurae. Relationships of Stenosternus with other orphnine taxa and possible ways of origin of São Toméan orphnine fauna are discussed.

Sampling Efficacy and Survival Rates of Labarrus pseudolividus (Coleoptera: Scarabaeidae) and Onthophagus taurus (Coleoptera: Scarabaeidae) Using Flotation and Sieve-Separation Methodology.

Project description:Understanding collection methodologies and their limitations are essential when targeting specific arthropods for use in habitat restoration, conservation, laboratory colony formation, or when holistically representing local populations using ecological surveys. For dung beetles, the most popular collection methodology is baited traps, followed by light traps and unbaited flight-intercept traps during diversity surveys. A less common collection method, flotation, is assumed to be laborious and messy, and so only a handful of papers exist on its refinement and strengths. Our purpose was threefold: First, we tested the recovery and survival rates of Labarrus (=Aphodius) pseudolividus (Balthasar) and Onthophagus taurus (Schreber) when floating beetle-seeded dung pats to determine potential collection and safety issues. We collected 72.4 and 78% of the seeded L. pseudolividus and O. taurus, respectively, with >95% survival rating. Second, we developed a flotation-sieving technique that enables users to rapidly collect and passively sort dung beetles with less time and effort. Specifically, we often collected 50-100 g of wild dung beetles within a couple of hours of gathering dung and sorted them in a couple more by allowing dung beetles to sort themselves by size within a series of sieves; Third, we reviewed flotation-based advantages and disadvantages in comparison to other methodologies.

Potential cooperations between odorant-binding proteins of the scarab beetle Holotrichia oblita Faldermann (Coleoptera: Scarabaeidae).

Project description:It was previously thought that the odorant binding proteins (OBPs) in the sensillum lymph might serve as carriers, which could carry lipophilic odorant molecules to olfactory receptors. In this study, two novel OBP genes of the scarab beetle Holotrichia oblita were screened using an antennal cDNA library. The full cDNA of HoblOBP3 and HoblOBP4 was cloned using reverse transcription PCR and rapid amplification of the cDNA ends. Homology modeling of both OBPs was performed using SWISS-MODEL on-line tools. Next, the two OBPs were expressed in Escherichia coli and purified using Ni ion affinity chromatography. The ligand-binding properties of HoblOBP3 and HoblOBP4 in 42 ligands respectively were measured using the fluorescence probe N-phenyl-naphthylamine (1-NPN). The results obtained from competitive binding assays demonstrated that HoblOBP4 showed a broader range of binding affinities to the test compounds, while HoblOBP3 displays more specific binding affinity. Furthermore, other OBPs and CSPs were expressed in Escherichia coli and purified using Ni ion affinity chromatography. Binding curves were measured for binary mixtures of OBPs and CSPs using 1-NPN, and the Scatchard plots exhibited "J"-like nonlinear correlation trends in some samples. In addition, competitive binding assays of the HoblOBP1 and HoblOBP2 mixtures and of the HoblOBP2 and HoblOBP4 mixtures with representative compounds unexpectedly demonstrated good affinity, which revealed extreme differences that were only obtained using the individual proteins. In the immunocytochemical analysis, colocalization of HoblOBP1 and HoblOBP2, and of HoblOBP2 and HoblOBP4, was detected in the sensilla basiconica and sensilla placodea, respectively. All of these results suggested that HoblOBP1 and HoblOBP2, as well as HoblOBP2 and HoblOBP4, may serve as heterodimers in the sensillum lymph.

A Cretaceous Chafer Beetle (Coleoptera: Scarabaeidae) with Exaggerated Hind Legs-Insight from Comparative Functional Morphology into a Possible Spring Movement.

Project description:The phenomenon of exaggerated morphological structures has fascinated people for centuries. Beetles of the family Scarabaeidae show many very diverse exaggerated characters, for example, a variety of horns, enlarged mandibles or elongated antennal lamellae. Here, we report a new Mesozoic scarab, Antiqusolidus maculatus gen. et sp. n. from the Lower Cretaceous Yixian Formation (~125 Ma), which has unusually robust and structured hind legs with greatly enlarged spurs and a unique elongated apical process. Based on simulations and finite element analyses, the function of these structures is hypothesized to support springing to aid movement and fighting. Based on available morphological characters, we performed phylogenetic analyses (maximum parsimony) of the main subfamilies and families of Scarabaeoidea. The results support the placement of Antiqusolidus gen. n. as a sister group of Rutelinae within the phytophagous lineage of pleurostict Scarabaeidae. Furthermore, the unusual delicate color marking patterns in the fossil specimens suggest that the new species might have been diurnal and potentially visited the leaves or flowers of Early Cretaceous plants. This morphological and functional study on this extinct scarab species provides new sights into exaggerated structures in Mesozoic insects.

Synopsis of the cyclocephaline scarab beetles (Coleoptera, Scarabaeidae, Dynastinae).

Project description:The cyclocephaline scarabs (Scarabaeidae: Dynastinae: Cyclocephalini) are a speciose tribe of beetles that include species that are ecologically and economically important as pollinators and pests of agriculture and turf. We provide an overview and synopsis of the 14 genera of Cyclocephalini that includes information on: 1) the taxonomic and nomenclatural history of the group; 2) diagnosis and identification of immature life-stages; 3) economic importance in agroecosystems; 4) natural enemies of these beetles; 5) use as food by humans; 6) the importance of adults as pollination mutualists; 7) fossil cyclocephalines and the evolution of the group; 8) generic-level identification of adults. We provide an expanded identification key to genera of world Cyclocephalini and diagnoses for each genus. Character illustrations and generic-level distribution maps are provided along with discussions on the relationships of the tribe's genera.

Comparative cytogenetics of three species of Dichotomius (Coleoptera, Scarabaeidae).

Project description:Meiotic and mitotic chromosomes of Dichotomius nisus, D. semisquamosus and D. sericeus were analyzed after conventional staining, C-banding and silver nitrate staining. In addition, Dichotomius nisus and D. semisquamosus chromosomes were also analyzed after fluorescent in situ hybridization (FISH) with an rDNA probe. The species analyzed had an asymmetrical karyotype with 2n = 18 and meta-submetacentric chromosomes. The sex determination mechanism was of the Xy(p) type in D. nisus and D. semisquamosus and of the Xy (r) type in D. sericeus. C-banding revealed the presence of pericentromeric blocks of constitutive heterochromatin (CH) in all the chromosomes of the three species. After silver staining, the nucleolar organizer regions (NORs) were located in autosomes of D. semisquamosus and D. sericeus and in the sexual bivalent of D. nisus. FISH with an rDNA probe confirmed NORs location in D. semisquamosus and in D. nisus. Our results suggest that chromosome inversions and fusions occurred during the evolution of the group.

New Coleoptera records from New Brunswick, Canada: Geotrupidae and Scarabaeidae.

Project description:Two species of Geotrupidae, Geotrupes splendidus splendidus (Fabricius) and Odonteus liebecki (Wallis), are newly reported for New Brunswick, Canada. Twelve species of Scarabaeidae are added to the faunal list of the province, including Aegialia criddlei Brown, Caelius humeralis (Brown), Dialytellus dialytoides (Fall), Diapterna omissa (LeConte), Diapterna pinguis (Haldeman), Planolinoides aenictus (Cooper and Gordon), Stenotothorax badipes (Melsheimer), and Ataenius strigatus (Say), which are also newly recorded for the Maritime provinces. Collection data, habitat data, and distribution maps are presented for each species.

Mitochondrial phylogenomics reveals deep relationships of scarab beetles (Coleoptera, Scarabaeidae).

Project description:In this study, we newly sequenced the complete mitochondrial genomes (mitogenomes) of two phytophagous scarab beetles, and investigated the deep level relationships within Scarabaeidae combined with other published beetle mitogenome sequences. The complete mitogenomes of Dicronocephalus adamsi Pascoe (Cetoniinae) and Amphimallon sp. (Melolonthinae) are 15,563 bp and 17,433 bp in size, respectively. Both mitogenomes have the typical set of 37 genes (13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes) and an A+T-rich region, with the same gene arrangement found in the majority of beetles. The secondary structures for ribosomal RNA genes (rrnL and rrnS) were inferred by comparative analysis method. Results from phylogenetic analyses provide support for major lineages and current classification of Scarabaeidae. Amino acid data recovered Scarabaeidae as monophyletic. The Scarabaeidae was split into two clades. One clade contained the subfamilies Scarabaeinae and Aphodiinae. The other major clade contained the subfamilies Dynastinae, Rutelinae, Cetoniinae, Melolonthinae and Sericini. The monophyly of Scarabaeinae, Aphodiinae, Dynastinae, Cetoniinae and Sericini were strongly supported. The Scarabaeinae was the sister group of Aphodiinae. The Cetoniinae was sister to the Dynastinae + Rutelinae clade. The Melolonthinae was a non-monophyletic group. The removal of fast-evolving sites from nucleotide dataset using a pattern sorting method (OV-sorting) supported the family Scarabaeidae as a monophyletic group. At the tribe level, the Onthophagini was non-monophyletic with respect to Oniticellini. Ateuchini was sister to a large clade comprising the tribes Onthophagini, Oniticellini and Onitini. Eurysternini was a sister group of the Phanaeini + Ateuchini clade.

A possible genetic link between MTHFR genotype and smoking behavior.

Project description:BACKGROUND: Hyperhomocysteinemia is an independent risk factor for stroke and other vascular events. The variant methylenetetrahydrofolate reductase (MTHFR) C677T is associated with elevated homocysteine levels, cardiovascular disease and stroke, which supports a causal relationship between hyperhomocysteinemia and vascular disease. However, MTHFR variants have also been reported to be associated with smoking behavior, which could be an important confounder. METHODOLOGY/PRINCIPAL FINDINGS: We analyzed the MTHFR variants C677T and A1298C in two independent samples of 525 and 535 individuals, respectively. 21% of the non-smokers, but only 12% of the smokers were homozygous carriers of both MTHFR wildtype alleles, i.e. 677CC and 1298AA (Chi²?=?15.8; p<0.001; binary regression). Plasma homocysteine levels were higher in smokers (13.9±4.1 µmol/L) than in non-smokers (12.6±4.0 µmol/L; F?=?11.4; p?=?0.001; ANOVA). Smoking MTHFR 677TT individuals had the highest plasma homocysteine levels (16.2±5.2 µmol/L), non-smoking 677CC individuals had the lowest (12.2±13.6 µmol/L). CONCLUSIONS/SIGNIFICANCE: In our study samples, MTHFR variants and smoking behaviour were associated with homocysteine plasma levels. In addition, the MTHFR variants were associated with smoking behaviour. Such an association may be a relevant confounder between MTHFR variants, homocysteine plasma levels and vascular diseases.
