Whole-Genome Sequences of Two Beak and Feather Disease Viruses in the Endangered Swift Parrot (Lathamus discolor).

ABSTRACT: Two complete genomes of beak and feather disease virus (BFDV) were characterized from Lathamus discolor, the Australian swift parrot. This is the first report of BFDV complete genome sequences in this host. The completed BFDV genomes consist of 1,984 nucleotides encoding two open reading frames with 99.7% pairwise nucleotide identity.

Molecular detection and analysis of beak and feather disease viruses in Iran.

Project description:The beak and feather disease virus (BFDV) is one of the few pathogens capable of causing extinction of psittacines. To determine the prevalence and the nature of BFDV mutation, this study investigated the presence of the BFDV among 1,095 individual birds of the 17 psittacine species in Iran followed by analyzing the DNA sequences of seven replication-associated protein (rep) and 10 capsid (cap) genomes of the virus. The BFDV was found to be the foremost pathogen among more than 12 psittacine species, and phylogenetic analysis showed that the BFDV GenBank-published sequences from Poland, Saudi Arabia, South Africa, Taiwan, and Thailand were most similar to those of this study. Evolutionary analysis concluded that arginine, leucine, and glycine were the amino acids frequently involved in the least-conserved substitution patterns of BFDV, and conversely, methionine, glutamine, and tryptophan were the amino acids that exhibited ultra-high conservation through the substitution patterns. The high substitution rate of arginine to lysine and glycine to serine also made greater contribution to the BFDV gene mutation. The relative synonymous codon usage between two genes revealed that the cap genome encoded proteins frequently used fewer codons, while the rep genome encoded proteins used more codons only at moderate frequency, explaining the broader divergence of the cap compared to the rep sequence. The data analysis also introduced a new variant of BFDV that exists in the rep and cap sequences of budgerigars. While the existence of more new variants was suspected, more solid evidence is required to substantiate this suspicion.

Virome of Australia's most endangered parrot in captivity evidenced of harboring hitherto unknown viruses.

Project description:ImportanceThe impact of circulating viruses on the critically endangered, orange-bellied parrot (OBP) population can be devastating. The OBP already faces numerous threats to its survival in the wild, including habitat loss, predation, and small population impacts. Conservation of the wild OBP population is heavily reliant on supplementation using OBPs from a managed captive breeding program. These birds may act as a source for introduction of a novel disease agent to the wild population that may affect survival and reproduction. It is, therefore, essential to monitor and assess the health of OBPs and take appropriate measures to prevent and control the spread of viral infections. This requires knowledge of the existing virome to identify novel and emerging viruses and support development of appropriate measures to manage associated risk. By monitoring and protecting these animals from emerging viral diseases, we can help ensure their ongoing survival and preserve the biodiversity of our planet.

Feather Pecking and Cannibalism in Non-Beak-Trimmed Laying Hen Flocks-Farmers' Perspectives.

Project description:Pecking-related problems are common in intensive egg production, compromising hen welfare, causing farmers economic losses and negatively affecting sustainability. These problems are often controlled by beak trimming, which in Finland is prohibited. An online questionnaire aimed to collect information from farmers about pecking-related problems in Finnish laying hen flocks, important risk factors and the best experiences to prevent the problems. Additionally, the farmers' attitudes towards beak trimming were examined. We received 35 responses, which represents about 13% of all Finnish laying hen farms with ≥300 laying hens. The majority of respondents stated that a maximum of 5⁻7% incidence of feather pecking or 1⁻2% incidence of cannibalism would be tolerable. The majority of respondents (74%) expressed that they would definitely not use beak-trimmed hens. Only two respondents indicated that they would probably use beak-trimmed hens were the practice permitted. Among risk factors, light intensity earned the highest mean (6.3), on a scale from 1 (not important) to 7 (extremely important). Other important problems included those that occurred during rearing, feeding, flock management and problems with drinking water equipment (mean 5.9, each). The most important intervention measures included optimal lighting and feeding, flock management, and removing the pecker and victim. Concluding, Finnish farmers had strong negative attitudes towards beak trimming. The study underlines the importance of flock management, especially lighting and feeding, in preventing pecking problems and indicates that it is possible to incorporate a non-beak-trimming policy into sustainable egg production.
