Effect of Hydrophobic Polypeptide Length on Performances of Thermo-Sensitive Hydrogels.

Project description:Thermosensitive gels are commonly used as drug carriers in medical fields, mainly due to their convenient processing and easy functionalization. However, their overall performance has been severely affected by their unsatisfying biocompatibility and biodegradability. To this end, we synthesized poly(l-alanine) (PLAla)-based thermosensitive hydrogels with different degrees of polymerization by ring-opening polymerization. The obtained mPEG45−PLAla copolymers showed distinct transition temperatures and degradation abilities. It was found that slight changes in the length of hydrophobic side groups had a decisive effect on the gelation behavior of the polypeptide hydrogel. Longer hydrophobic ends led to a lower gelation temperature of gel at the same concentration, which implied better gelation capability. The hydrogels showed rapid gelling, enhanced biocompatibility, and better degradability. Therefore, this thermosensitive hydrogel is a promising material for biomedical application.

DOX/IL-2/IFN-γ co-loaded thermo-sensitive polypeptide hydrogel for efficient melanoma treatment.

Project description:Melanoma has been a serious threat to the human health; however, effective therapeutic methods of this cancer are still limited. Combined local therapy is a crucial approach for achieving a superior anti-tumor efficacy. In this paper, a chemo-immunotherapy system of DOX, IL-2 and IFN-γ based on poly(γ-ethyl-L-glutamate)-poly(ethylene glycol)-poly(γ-ethyl-L-glutamate) (PELG-PEG-PELG) hydrogel was developed for local treatment of melanoma xenograft. The drug release process of this system exhibited a short term of burst release (the first 3 days), followed by a long-term sustained release (the following 26 days). The hydrogel degraded completely within 3 weeks without obvious inflammatory responses in the subcutaneous layer of rats, showing a good biodegradability and biocompatibility. The DOX/IL-2/IFN-γ co-loaded hydrogel also showed enhanced anti-tumor effect against B16F10 cells in vitro, through increasing the ratio of cell apoptosis and G2/S phage cycle arrest. Moreover, the combined strategy presented improved therapy efficacy against B16F10 melanoma xenograft without obvious systemic side effects in a nude mice model, which was likely related to both the enhanced tumor cell apoptosis and the increased proliferation of the CD3+/CD4+ T-lymphocytes and CD3+/CD8+ T-lymphocytes. Overall, the strategy of localized co-delivery of DOX/IL-2/IFN-γ using the polypeptide hydrogel provided a promising approach for efficient melanoma therapy.

Tunable Thermo-Responsive Properties of Hydroxybutyl Chitosan Oligosaccharide.

Project description:In this study, a simple method was used to synthesize novel thermosensitive hydroxybutyl chitosan oligosaccharide (HBCOS) by introducing hydroxybutyl groups to C6-OH of chitosan oligosaccharide (COS) chain. The variation in light scattering demonstrated that HBCOS had good thermosensitive properties and the particle size of HBCOS changed from 2.21-3.58 to 281.23-4,162.40 nm as the temperature increased to a critical temperature (LCST). The LCST of HBCOS (10 mg/ml) decreased from 56.25°C to 40.2°C as the degrees of substitution (DSs) increased from 2.96 to 4.66. The LCST of HBCOS with a DS of 4.66 decreased to 33.5°C and 30°C as the HBCOS and NaCl concentrations increased to 50 mg/ml and 4% (w/v), respectively. Variable-temperature FTIR spectroscopy confirmed that dehydration of hydrophobic chains and the transition of hydrogen bonds were the driving forces for the phase transition of HBCOS. Moreover, HBCOS was not cytotoxic at different concentrations. This work generated a novel thermosensitive HBCOS with tunable thermoresponsive properties and excellent biocompatibility, which may be a potential nanocarrier for the biomedical application.

The thermo-sensitive gene expression signatures of spermatogenesis.

Project description:BACKGROUND:Spermatogenesis in most mammals (including human and rat) occurs at ~?3 °C lower than body temperature in a scrotum and fails rapidly at 37 °C inside the abdomen. The present study investigates the heat-sensitive transcriptome and miRNAs in the most vulnerable germ cells (spermatocytes and round spermatids) that are primarily targeted at elevated temperature in a bid to identify novel targets for contraception and/or infertility treatment. METHODS:Testes of adult male rats subjected to surgical cryptorchidism were obtained at 0, 24, 72 and 120 h post-surgery, followed by isolation of primary spermatocytes and round spermatids and purification to >?90% purity using a combination of trypsin digestion, centrifugal elutriation and density gradient centrifugation techniques. RNA isolated from these cells was sequenced by massive parallel sequencing technique to identify the most-heat sensitive mRNAs and miRNAs. RESULTS:Heat stress altered the expression of a large number of genes by ?2.0 fold, out of which 594 genes (286?; 308?) showed alterations in spermatocytes and 154 genes (105?; 49?) showed alterations in spermatids throughout the duration of experiment. 62 heat-sensitive genes were common to both cell types. Similarly, 66 and 60 heat-sensitive miRNAs in spermatocytes and spermatids, respectively, were affected by ?1.5 fold, out of which 6 were common to both the cell types. CONCLUSION:The study has identified Acly, selV, SLC16A7(MCT-2), Txnrd1 and Prkar2B as potential heat sensitive targets in germ cells, which may be tightly regulated by heat sensitive miRNAs rno-miR-22-3P, rno-miR-22-5P, rno-miR-129-5P, rno-miR-3560, rno-miR-3560 and rno-miR-466c-5P.

Synthetic Thermo-Responsive Terpolymers as Tunable Scaffolds for Cell Culture Applications.

Project description:The use of tailored synthetic hydrogels for in vitro tissue culture and biomanufacturing provides the advantage of mimicking the cell microenvironment without issues of batch-to-batch variability. To that end, this work focused on the design, characterization, and preliminary evaluation of thermo-responsive, transparent synthetic terpolymers based on N-isopropylacrylamide, vinylphenylboronic acid, and polyethylene glycol for cell manufacturing and in vitro culture applications. Polymer physical properties were characterized by FT-IR, 1H-NMR, DLS, rheology, and thermal-gravimetric analysis. Tested combinations provided polymers with a lower critical solution temperature (LCST) between 30 and 45 °C. Terpolymer elastic/shear modulus varied between 0.3 and 19.1 kPa at 37 °C. Cellular characterization indicated low cell cytotoxicity on NIH-3T3. Experiments with the ovarian cancer model SKOV-3 and Jurkat T cells showed the terpolymers' capacity for cell encapsulation without interfering with staining or imaging protocols. In addition, cell growth and high levels of pluripotency demonstrated the capability of terpolymer to culture iPSCs. Characterization results confirmed a promising use of terpolymers as a tunable scaffold for cell culture applications.

Tunable thermo-reversible bicontinuous nanoparticle gel driven by the binary solvent segregation.

Project description:Bicontinuous porous structures through colloidal assembly realized by non-equilibrium process is crucial to various applications, including water treatment, catalysis and energy storage. However, as non-equilibrium structures are process-dependent, it is very challenging to simultaneously achieve reversibility, reproducibility, scalability, and tunability over material structures and properties. Here, a novel solvent segregation driven gel (SeedGel) is proposed and demonstrated to arrest bicontinuous structures with excellent thermal structural reversibility and reproducibility, tunable domain size, adjustable gel transition temperature, and amazing optical properties. It is achieved by trapping nanoparticles into one of the solvent domains upon the phase separation of the binary solvent. Due to the universality of the solvent driven particle phase separation, SeedGel is thus potentially a generic method for a wide range of colloidal systems.

The Hsp70 Chaperone System Stabilizes a Thermo-sensitive Subproteome

Project description:Stress-inducible molecular chaperones have essential roles in maintaining protein homeostasis, but the extent to which they affect global proteome stability remains unclear. Here, we analyzed the effects of the DnaK (Hsp70) system on protein stability in Escherichia coli using pulse proteolysis combined with quantitative proteomics. We quantified ~1500 soluble proteins and found ~500 of them to be protease-sensitive under normal growth conditions, indicating a high prevalence of conformationally dynamic states. Acute heat stress resulted in unfolding of an additional ~200 proteins, reflected in exposure of otherwise buried hydrophobic regions. Overexpression of the DnaK chaperone system markedly stabilized most thermo-sensitive proteins, including numerous ribosomal proteins as well as large, hetero-oligomeric proteins that frequently contain the evolutionary ancient c.37 fold (P-loop nucleoside triphosphate hydrolases).

Stimulus-Responsiveness of Thermo-Sensitive Polymer Hybridized with N-Doped Carbon Quantum Dots and Its Applications in Solvent Recognition and Fe<sup>3+</sup> Ion Detection.

Project description:To fabricate N-CQDs hybrid thermo-sensitive polymer (poly-N-CQDs), N-doped carbon quantum dots (N-CQDs) with strong blue fluorescence and poly(N-isopropylacrylamide-co-acrylic acid) (poly(NIPAAm-co-AAc)) copolymer with thermo-sensitivity were synthesized, respectively. Subsequently, the coupling reaction between. the -COOH groups of poly(NIPAAm-co-AAc) and the -NH2 groups on the surface of the N-CQDs was carried out. The fluorescence spectra show that the coil-globule transition of the poly-N-CQDs coincided with intensity changes in the scattering peak at excitation wavelength with the temperature variations. The phase transition temperature and the fluorescent intensity of poly-N-CQDs can be regulated by modulating the composition and concentration of poly-N-CQDs as well as the temperature and pH of the local medium. The thermo-sensitivity and fluorescent properties of the poly-N-CQDs displayed good stability and reversibility. The fluorescence intensity and emission wavelengths of the poly-N-CQDs significantly changed in different solvents for solvent recognition. The poly-N-CQDs was employed as a fluorescent probe for Fe3+ detection ranging from 0.025 to 1 mM with a limit of detection (LOD) of 9.49 μM. The hybrid polymer materials have the potential to develop an N-CQDs-based thermo-sensitive device or sensor.

Thermo-tunable hybrid photonic crystal fiber based on solution-processed chalcogenide glass nanolayers.

Project description:The possibility to combine silica photonic crystal fiber (PCF) as low-loss platform with advanced functional materials, offers an enormous range of choices for the development of fiber-based tunable devices. Here, we report a tunable hybrid silica PCF with integrated As2S3 glass nanolayers inside the air-capillaries of the fiber based on a solution-processed glass approach. The deposited high-index layers revealed antiresonant transmission windows from ~500 nm up to ~1300 nm. We experimentally demonstrate for the first time the possibility to thermally-tune the revealed antiresonances by taking advantage the high thermo-optic coefficient of the solution-processed nanolayers. Two different hybrid fiber structures, with core diameter 10 and 5 μm, were developed and characterized using a supercontinuum source. The maximum sensitivity was measured to be as high as 3.6 nm/°C at 1300 nm. The proposed fiber device could potentially constitute an efficient route towards realization of monolithic tunable fiber filters or sensing elements.
