Complete Genome Sequence of a Genotype XVII Newcastle Disease Virus, Isolated from an Apparently Healthy Domestic Duck in Nigeria.

ABSTRACT: The first complete genome sequence of a strain of Newcastle disease virus (NDV) of genotype XVII is described here. A velogenic strain (duck/Nigeria/903/KUDU-113/1992) was isolated from an apparently healthy free-roaming domestic duck sampled in Kuru, Nigeria, in 1992. Phylogenetic analysis of the fusion protein gene and complete genome classified the isolate as a member of NDV class II, genotype XVII.

Phylogenetic relationship and genotype variation of six Newcastle disease viruses isolated from duck in Indonesia.

Project description:Background and aimNewcastle disease viruses (NDVs) are frequently acquired from all ages and types of bird species. In general, ducks are considered as potential reservoirs for different genotypes of NDV and are resistant even to velogenic NDV strains. This research was conducted to genotypically and phylogenetically characterize NDV isolates collected from unvaccinated ducks from Indonesia.Materials and methodsA total of 200 samples were collected through cloacal swabs and were inoculated in the allantoic sacs of 8-day-old specific pathogen-free eggs. Hemagglutination (HA) activity was analyzed through a HA test, and isolated viruses were characterized by reverse transcription-polymerase chain reaction targeting the complete fusion (F)-gene of NDV using three primer sets. One primer set was specific for the F protein cleavage site sequences of velogenic, mesogenic, and lentogenic NDV strains.ResultsThe results demonstrated that three isolates (NDV/Duck/B104/19, NDV/Duck/B125/19, and NDV/Duck/BK43/19) belonged to genotype VII and one (NDV/Duck/TD19/19) to genotype VI. Other isolates (NDV/Duck/A74/19 and NDV/Duck/M147/19) belonged to genotype II Class II. Based on the F protein cleavage site and the pathogenicity tests, two isolates (NDV/Duck/B104/19 and NDV/Duck/B125/19) were categorized as velogenic viruses and four (NDV/Duck/BK43/19, NDV/Duck/TD19/19, NDV/Duck/A74/19, and NDV/Duck/M147/19) as lentogenic viruses.ConclusionThe results indicate that NDVs from unvaccinated ducks from Indonesia carry various genotypes and pathotypes of NDVs; therefore, these viruses are still circulating in the environment and might pose a risk of Newcastle disease outbreak.

This strain is classified as a member of subgenotype XIVb of class II.

Identification of a Putative Novel Genotype of Avian Hepatitis E Virus from Apparently Healthy Chickens in Southwestern Nigeria.

Project description:Avian hepatitis E virus (aHEV) is the major etiological agent of hepatitis-splenomegaly syndrome (HSS), big liver and spleen disease (BLSD), and hepatic rupture hemorrhage syndrome (HRHS) in chickens. Infections with aHEV cause a significant decrease in egg production and increased mortality in chickens worldwide. However, studies on the prevalence of aHEV in Nigeria are scarce. In this study, serum (n = 88) and fecal samples (n = 110) obtained from apparently healthy layer chickens from three states in southwestern Nigeria were analyzed by nested reverse transcription-polymerase chain reaction (nRT-PCR) targeting the helicase and capsid gene for the presence of aHEV. Avian HEV was detected in 12.5% (n = 11/88) of serum samples and 9.1% (n = 10/110) of fecal samples tested. Phylogenetic analysis showed that five of the twelve identified aHEV sequences belonged to genotype 2. The remaining seven sequences were only distantly related to other known aHEV isolates. After amplification of the near-complete ORF2 fragment (1618 bp) and part of the ORF1 (582 bp) of isolate YF40_aHEV_NG phylogenetic analysis revealed a nucleotide sequence identity between 79.0 and 82.6% and 80.1 and 83.5%, respectively, to other known aHEV strains, indicating that the Nigerian isolate YF40_aHEV_NG belongs to a novel aHEV genotype. This is the first report of co-circulation of aHEV genotypes in chickens in Nigeria.

Phylogenetic analysis of the fusion protein genes and complete genomes classified the isolates as members of NDV class II, genotype VI.

Electrocardiographic pattern of apparently healthy African adolescent athletes in Nigeria.

Project description:BackgroundStrategies to prevent sudden cardiac death (SCD) among young athletes have become topical worldwide and unrecognized cardiac pathology has been identified as a leading cause. Black ethnicity has been reported as an independent predictor of abnormal electrocardiography (ECG) findings among athletes and the frequency and significance of training-related ECG findings versus findings suggestive of an underlying pathology in the young African athletes is crucial.MethodsThis cross sectional study aimed to determine the prevalence and distribution of ECG patterns in young athletes and controls. A total of 360 participants (180 athletes and 180 controls) were recruited from six secondary schools in Lagos, Nigeria between November 2014 and July 2015. Evaluation included interviewer-administered questionnaires for relevant history, physical examination and resting 12 - lead ECG for each participant.ResultsAbnormal ECG patterns were found in 48.3% of athletes and 35.6% of controls. Training-related ECG findings occurred in 33.3% of athletes and 18.3% of controls. Athletes and controls had 7.7% prevalence of training un-related ECG patterns respectively. Left ventricular hypertrophy was the most common ECG finding among the athletes and male athletes had a higher prevalence of ECG abnormalities compared to females.ConclusionAdolescent athletes in Nigeria have a high prevalence of training-related ECG patterns and athletes and non-athletes alike have similar proportions of ECG findings suggestive of underlying structural heart disease. Cardiovascular evaluation including ECG should be performed for young athletes prior to competition at any level and should also be considered as part of pre-school entry assessment for all children.

Identification of a Genotype IX Newcastle Disease Virus in a Guangxi White Duck.

Project description:We report the complete genomic sequence of a novel Newcastle disease virus (NDV) strain, duck/China/Guangxi19/2011, isolated from a white duck in Guangxi Province, southern China. Phylogenetic analysis based on a fusion gene comparison with different NDV strains revealed that duck/China/Guangxi19/2011 is phylogenetically close to genotype IX NDV, and the deduced amino acid sequence of the fusion protein cleavage site was 112R-R-Q-R-R-F117. The whole nucleotide sequence had the highest homology (99.7%) to the sequence of strain F48E8 (GenBank accession number FJ436302). This study will help us understand the epidemiology and molecular characteristics of genotype IX Newcastle disease virus in ducks.
