Sequential Pathology of a Genotype XIII Newcastle Disease Virus from Bangladesh in Chickens on Experimental Infection.

Project description:The sequential pathology of a genotype XIII Bangladeshi strain of Newcastle disease virus (NDV) was studied in 5-weeks old chickens. Layer chickens of ISA Brown breed were inoculated through the intranasal and intraocular routes with the BD-C161/2010 strain of NDV and examined at different times post-infection (pi). NDV-infected chickens showed depression at 3 days pi (dpi) followed by dropped wings, paralysis and death starting at 4 dpi. Lungs of infected chickens showed hemorrhagic lesions starting at 24 hours pi (hpi) that was followed by pallor and slight contraction by 2 to 3 dpi and subsequently developed into severe hemorrhagic pneumonia with mononuclear cell infiltration. Hemorrhagic and necrotizing lesions were found in different visceral organs including proventriculus, intestine, gut-associated lymphoid tissues, liver and kidneys starting at 3 dpi that progressed rapidly. Severe lymphoid depletion was observed in the thymus, spleen and bursa of Fabricius starting at 1-3 dpi followed by hemorrhages, necrosis, inflammation and atrophy at 4-5 dpi. In the brain, mild neuronal lesions such as focal to diffuse encephalitis with encephalomalacia was observed at 2-3 dpi and moderate and diffuse meningoencephalitis with encephalomalacia at advanced stages. In conclusion, the BD-C161/2010 strain of NDV produced lesions typical of velogenic viscerotropic pathotype of NDV.

Virulent isolates pigeon/Pakistan/Lahore/21A/2015 and pigeon/Pakistan/Lahore/25A/2015 were obtained from racing pigeons sampled in the Pakistani province of Punjab during 2015. Phylogenetic analysis of the fusion protein genes and complete genomes classified the isolates as members of NDV class II, genotype VI.

Isolation and characterization of genotype XIII Newcastle disease virus from Emu in India.

Project description:Newcastle disease virus (NDV) infects at least 241 species of pet and free-living birds in addition to domesticated avian species. Wild, feral and domesticated birds are recognized reservoirs of NDV, and contribute to the epidemiology of NDV in the domesticated poultry. The biological and molecular characterization of velogenic NDV (vNDV) from emus is limited. In this study, 54 tissues were collected from eight Emu flocks between May 2010 and January 2012 from highly poultry-dense areas of India including Namakkal, Hyderabad and Bareilly regions. The presence of vNDV was confirmed by virus isolation, fusion (F)-gene based RT-PCR, sequencing of the cleavage site and the virulence scored. One out of eight flocks received from Hyderabad region was found positive for NDV and the in vivo pathotyping revealed the isolate to be vNDV type. The sequence analysis revealed that the isolate had cleavage site of 112-R-R-R-K-R-F-117, which is typical for vNDV. Sequence and phylogenetic analysis of the partial 'F' gene coding regions suggested that the NDV strain belongs to genotype XIII. The Emu isolate had 98-100 % nucleotide identity with the vNDVs previously reported in poultry flocks of India. The findings of present study based on the biological and molecular characterization of Emu-origin vNDV, highlights the circulation of genotype XIII in Emus for the first time in the country. There is need to understand the possible spill over of these genetically diverse NDV strains into the commercial poultry and their possible implications in disease control strategies.

Project description:The first complete genome sequence of a strain of Newcastle disease virus (NDV) of genotype XVII is described here. A velogenic strain (duck/Nigeria/903/KUDU-113/1992) was isolated from an apparently healthy free-roaming domestic duck sampled in Kuru, Nigeria, in 1992. Phylogenetic analysis of the fusion protein gene and complete genome classified the isolate as a member of NDV class II, genotype XVII.

Hemagglutinin-neuraminidase Sequence and Phylogenetic Analysis of Two Newcastle Disease Virus Isolated from Chickens in Iran.

Project description:Newcastle disease is a highly contagious viral infection affecting many species of birds that can spread fast between poultry houses and cause a heavy economic burden on the poultry industry all around the world. Fusion and hemagglutinin-neuraminidase (HN) protein are important in the pathogenesis of the Newcastle disease virus (NDV). The HN protein is a critical viral protein with multiple functions and plays a key role in the formation of the virulence of NDV. Head of HN protein is responsible for receptor binding, neuraminidase activity. This study aimed to investigate the sequence homology of hemagglutinin-neuraminidase of two NDV isolates sampled from infected farms in Iran. The samples were collected from flocks that had been vaccinated by both types of live and killed vaccines for NDV. After isolation of NDV, the viruses were subjected to the polymerase chain reaction (PCR) amplification using two pairs of specific primers designed for the HN gene to amplify the complete HN gene (1730bp). Afterward, the PCR products were sequenced and analyzed by phylogenetic tree construction software. Based on the analysis, substantial sequence homology among Iranian isolates is within the range of 97.1-100%. Moreover, the sequence homology searching revealed a level of similarity between HN sequences of Iranian isolates and the HN sequences from other countries, particularly Asian ones. For instance, a high homology ratio (95.34%) was found between Iranian isolates and the sequences registered on online molecular databases from China. Based on phylogenetic analysis, the NDV isolates belong to the VIId genotype. Finally, it can be concluded that monitoring the circulation of NDVs among poultry and other birds can help to obtain an insight into the evolution of NDVs and control of panzootic viruses in the future.

Complete genome sequence of a subgenotype VIId Newcastle disease virus circulating predominantly in chickens in China.

Project description:At least 11 genotypes of class II viruses have been identified since the discovery of Newcastle disease virus (NDV) in 1926. Here, we reported the complete genome sequence of a prevalent NDV variant from China, belonging to subgenotype VIId in class II. The similar viruses have been the predominant strains circulating in China for the past decade, which occupied over 80% of Chinese prevalent strains and were phylogenetically different from currently available vaccines.

Complete genome sequence analysis of a Newcastle disease virus isolated from a wild egret.

Project description:We report here the complete genomic sequence of a novel Newcastle disease virus (NDV) strain, egret/China/Guangxi/2011, isolated from an egret in Guangxi Province, southern China. A phylogenetic analysis based on a fusion gene comparison with different NDV strains revealed that egret/China/Guangxi/2011 was phylogenetically close to genotype VIIa NDV, and the deduced amino acid sequence was (112)R-R-R-K-R-F(117) at the fusion protein cleavage site. The whole nucleotide sequence had the highest homology (93.3%) with the sequence of strain chicken/Sukorejo/019/10 (GenBank accession number HQ697255). This study will help us to understand the epidemiology and molecular characteristics of Newcastle disease virus in a migratory egret.
