A Pilot-scale Benthic Microbial Electrochemical System (BMES) for Enhanced Organic Removal in Sediment Restoration.

ABSTRACT: A benthic microbial electrochemical systems (BMES) of 195?L (120?cm long, 25?cm wide and 65?cm height) was constructed for sediment organic removal. Sediment from a natural river (Ashi River) was used as test sediments in the present research. Three-dimensional anode (Tri-DSA) with honeycomb structure composed of carbon cloth and supporting skeleton was employed in this research for the first time. The results demonstrated that BMES performed good in organic-matter degradation and energy generation from sediment and could be considered for river sediments in situ restoration as novel method. Community analysis from the soil and anode using 16S rDNA gene sequencing showed that more electrogenic functional bacteria was accumulated in anode area when circuit connected than control system.

Project description:Western Antarctica, one of the fastest warming locations on Earth, is a unique environment that is underexplored with regards to biodiversity. Although pelagic microbial communities in the Southern Ocean and coastal Antarctic waters have been well-studied, there are fewer investigations of benthic communities and most have a focused geographic range. We sampled surface sediment from 24 sites across a 5500 km region of Western Antarctica (covering the Ross Sea to the Weddell Sea) to examine relationships between microbial communities and sediment geochemistry. Sequencing of the 16S and 18S rRNA genes showed microbial communities in sediments from the Antarctic Peninsula (AP) and Western Antarctica (WA), including the Ross, Amundsen, and Bellingshausen Seas, could be distinguished by correlations with organic matter concentrations and stable isotope fractionation (total organic carbon; TOC, total nitrogen; TN, and δ(13)C). Overall, samples from the AP were higher in nutrient content (TOC, TN, and NH4 (+)) and communities in these samples had higher relative abundances of operational taxonomic units (OTUs) classified as the diatom, Chaetoceros, a marine cercozoan, and four OTUs classified as Flammeovirgaceae or Flavobacteria. As these OTUs were strongly correlated with TOC, the data suggests the diatoms could be a source of organic matter and the Bacteroidetes and cercozoan are grazers that consume the organic matter. Additionally, samples from WA have lower nutrients and were dominated by Thaumarchaeota, which could be related to their known ability to thrive as lithotrophs. This study documents the largest analysis of benthic microbial communities to date in the Southern Ocean, representing almost half the continental shoreline of Antarctica, and documents trophic interactions and coupling of pelagic and benthic communities. Our results indicate potential modifications in carbon sequestration processes related to change in community composition, identifying a prospective mechanism that links climate change to carbon availability.

Project description:Dam removal is an increasingly important method of stream restoration, but most removal efforts are under-studied in their effects. In order to better understand the effects of such removals on the stream ecosystem, we examined changes in stream macroinvertebrate communities from 2011-2016. Comparisons were focused above, below, and before and after the October 2012 removal of the Brown Bridge Dam on the Boardman River in Michigan (USA), as well as to new channel sites created in its former reservoir (2013-2015). Using linear mixed-effect models on the percent abundance of ecologically sensitive taxa (% Ephemeroptera, Plecoptera, Trichoptera (EPT)), total density of all macroinvertebrates, overall taxa richness, and Functional Feeding Groups, along with multivariate analyses on the community matrix, we examined differences in community composition among sites and years. EPT declined downstream of the dam immediately after dam removal, but recovered in the second year, becoming dominant within 2-4 years. Downstream sites before removal had different community composition than upstream sites and downstream sites after removal (p<0.001), while upstream and downstream sites after removal converged towards similarity. New channel (restored) %EPT, density, and taxa richness were not different from upstream sites in any year following removal, but new channel sites were the most distinct in community composition, possessing multiple indicator taxa characteristic of unique new conditions. The invasive New Zealand mud snail (Potamopyrgus antipodarum) was absent from all sites prior to dam removal, but appeared at low densities in upstream sites in 2013, had spread to all sites by 2015, and showed large increases at all sites by 2016. Managers employing dam removal for stream restoration should include post-removal monitoring for multiple years following removal and conduct risk analysis regarding potential effects on colonization of invasive invertebrate species.
