To showcase the Networks' success during phase two (2012-2016), and to set out the strategy for phase three (2017-2019), the Directed Assembly Network held a meeting at the Royal Society in London, United Kingdom on 14 and 15 December 2016. Project description:Breast cancer is the most common cancer worldwide. The occurrence of breast cancer is associated with many risk factors, including genetic and hereditary predisposition. Breast cancers are highly heterogeneous. Treatment strategies for breast cancer vary by molecular features, including activation of human epidermal growth factor receptor 2 (HER2), hormonal receptors (estrogen receptor [ER] and progesterone receptor [PR]), gene mutations (e.g., mutations of breast cancer 1/2 [BRCA1/2] and phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit alpha [PIK3CA]) and markers of the immune microenvironment (e.g., tumor-infiltrating lymphocyte [TIL] and programmed death-ligand 1 [PD-L1]). Early-stage breast cancer is considered curable, for which local-regional therapies (surgery and radiotherapy) are the cornerstone, with systemic therapy given before or after surgery when necessary. Preoperative or neoadjuvant therapy, including targeted drugs or immune checkpoint inhibitors, has become the standard of care for most early-stage HER2-positive and triple-negative breast cancer, followed by risk-adapted post-surgical strategies. For ER-positive early breast cancer, endocrine therapy for 5-10 years is essential. Advanced breast cancer with distant metastases is currently considered incurable. Systemic therapies in this setting include endocrine therapy with targeted agents, such as CDK4/6 inhibitors and phosphoinositide 3-kinase (PI3K) inhibitors for hormone receptor-positive disease, anti-HER2 targeted therapy for HER2-positive disease, poly(ADP-ribose) polymerase inhibitors for BRCA1/2 mutation carriers and immunotherapy currently for part of triple-negative disease. Innovation technologies of precision medicine may guide individualized treatment escalation or de-escalation in the future. In this review, we summarized the latest scientific information and discussed the future perspectives on breast cancer.

Project description:Even the rather traditional consulting industry is not spared from digitization. Digital platforms are known to foster convergence and generativity. For the consulting industry, digital platforms offer the potential to win new customer groups who have not previously purchased consulting services before. If digital platforms for the mediation of consulting services are established by incumbent consulting companies, the new platform-based business models for the consulting market will appear. Since network effects fuel platforms, the launch phase of a platform is a serious challenge. In this paper, we examine the launch of a digital platform for the algorithmic consulting (AC) approach, due to its promising market potential. In order to research the adaptability of digital platforms for AC consulting, we perform a qualitative study of electronic documents on the actions of three successful crowdsourcing platforms. The preliminary results comprise 14 success factors for the platform launch that will be validated in a follow-up practical application. The open nature of platform design in the areas of customer access, cooperation with other platforms, interfaces, and communication appeared to be particularly important for a successful platform launch. All identified success factors can be applied to AC services and the consulting market in the future.

Project description:Nudges influence behavior by changing the environment in which decisions are made, without restricting the menu of options and without altering financial incentives. This paper assesses past empirical research on nudging and provides recommendations for future work in this area by discussing examples of successful and unsuccessful nudges and by analyzing 174 articles that estimate nudge treatment effects. Researchers in disciplines spanning the behavioral sciences, using varied data sources, have documented that many different types of nudges succeed in changing behavior in a wide range of domains. Nudges that automate some aspect of the decision-making process have an average effect size, measured by Cohen's d, that is 0.193 larger than that of other nudges. Our analyses point to the need for future research to pay greater attention to (1) determining which types of nudges tend to be most impactful; (2) using field and laboratory research approaches as complementary methods; (3) measuring long-run effects of nudges; (4) considering effects of nudges on non-targeted outcomes; and (5) examining interaction effects among nudges and other interventions.

Project description:Integrating cover crops (CC) in rotations provides multiple ecological services, but it must be ensured that management does not increase pre-emptive competition with the subsequent crop. This experiment was conducted to study the effect of kill date on: (i) CC growth and N content; (ii) the chemical composition of residues; (iii) soil inorganic N and potentially mineralizable N; and (iv) soil water content. Treatments were fallow and a CC mixture of barley (Hordeum vulgare L.) and vetch (Vicia sativa L.) sown in October and killed on two different dates in spring. Above-ground biomass and chemical composition of CC were determined at harvest, and ground cover was monitored based on digital image analysis. Soil mineral N was determined before sowing and after killing the CC, and potentially mineralizable N was measured by aerobic incubation at the end of the experiment. Soil water content was monitored daily to a depth of 1.1 m using capacitance sensors. Under the present conditions of high N availability, delaying kill date increased barley above-ground biomass and N uptake from deep soil layers; little differences were observed in vetch. Postponing kill date increased the C/N ratio and the fiber content of plant residues. Ground cover reached >80% by the first kill date (∼1250°C days). Kill date was a means to control soil inorganic N by balancing the N retained in the residue and soil, and showed promise for mitigating N losses. The early kill date decreased the risk of water and N pre-emptive competition by reducing soil depletion, preserving rain harvested between kill dates and allowing more time for N release in spring. The soil potentially mineralizable N was enhanced by the CC and kill date delay. Therefore kill date is a crucial management variable for maximizing the CC benefits in agricultural systems.

Project description:Transparent conducting oxides (TCOs) are electrical conductive materials with comparably low absorption of electromagnetic waves within the visible region of the spectrum. They are usually prepared with thin film technologies and used in opto-electrical apparatus such as solar cells, displays, opto-electrical interfaces and circuitries. Here, based on a modern database-system, aspects of up-to-date material selections and applications for transparent conducting oxides are sketched, and references for detailed information are given. As n-type TCOs are of special importance for thin film solar cell production, indium-tin oxide (ITO) and the reasonably priced aluminum-doped zinc oxide (ZnO:Al), are discussed with view on preparation, characterization and special occurrences. For completion, the recently frequently mentioned typical p-type delafossite TCOs are described as well, providing a variety of references, as a detailed discussion is not reasonable within an overview publication.

Project description:The number and type of synthetic chemicals that are being produced worldwide continues to increase significantly. While these industrial chemicals provide numerous benefits, there is no doubt that some have potential to damage the environment and health. Toxicity must be evaluated and use must be carefully controlled and monitored in order to minimize potential damage. DNA microarray technology has become an important new technique in toxicology. We are using the yeast Saccharomyces cerevisiae as a model organism for toxicological study because it is a simple, fast-growing eukaryote that has been thoroughly characterized. In order to evaluate toxicity by newly synthesized or mixture chemicals, toxicity-induced gene expression alteration profiles by known chemicals should be collected. In our study, cells need to be exposed with same experimental cellular condition, semi lethal (IC50), respectively. In the case of round up (CAS; 40465-66-5), the exposure dose was decided as 1500 times dilution by growth curve with continuously diluted exposure. Roundup is the brand name of a systemic, broad-spectrum herbicide contains the active ingredient glyphosate. Glyphosate is classed as a moderately toxic herbicide and in EPA toxicity class III. // Genomic profile of roundup treatment of yeast using DNA microarray analysis: The herbicide Roundup, which contains glyphosate as the active ingredient, was first introduced in 1974 and has enjoyed widespread use in Japan and elsewhere in the world. Roundup-induced reactions occurring in the yeast Saccharomyces cerevisiae may have a predictive value for understanding responses in higher eukaryotes, and we applied yeast DNA microarray analysis for this purpose. Functional characterization of up-regulated open reading frames (ORFs) following Roundup treatment suggests that Roundup affects membrane structures and cellular organelles. Expression profiles induced by treatments with detergents, oils and hydrostatic pressure were similar to those following Roundup treatment based on cluster analysis. Glyphosate alone was not found to inhibit yeast growth at the concentration contained in the Roundup treatment used for microarray analysis. The toxicity of Roundup appeared to be due to detergent in the product. Keywords: stress response

Project description:Protein phosphorylation is one of the most common post-translational modifications (PTMs), which can regulate protein activity and localization, as well as protein–protein interactions in numerous cellular processes. Phosphopeptide enrichment techniques enabled plant researchers to acquire insight in phosphorylation-controlled signaling networks in various plant species. However, most phosphoproteome analyses of plant samples still involve stable isotope labeling, peptide fractionation, and demand lots of mass spectrometry (MS) time. Here, we present a simple workflow to probe, map and catalogue plant phosphoproteomes, requiring low starting materials, no labeling, no fractionation, and limited analysis time. Following optimization of the different experimental steps on Arabidopsis material, we transferred our workflow to maize, a major monocot crop, to study stress signaling upon drought.

Project description:Proteins represent the most sophisticated building blocks available to an organism and to the laboratory chemist. Yet, in contrast to nearly all other types of molecular building blocks, the designed self-assembly of proteins has largely been inaccessible because of the chemical and structural heterogeneity of protein surfaces. To circumvent the challenge of programming extensive non-covalent interactions to control protein self-assembly, we have previously exploited the directionality and strength of metal coordination interactions to guide the formation of closed, homoligomeric protein assemblies. Here, we extend this strategy to the generation of periodic protein arrays. We show that a monomeric protein with properly oriented coordination motifs on its surface can arrange, on metal binding, into one-dimensional nanotubes and two- or three-dimensional crystalline arrays with dimensions that collectively span nearly the entire nano- and micrometre scale. The assembly of these arrays is tuned predictably by external stimuli, such as metal concentration and pH.

Project description:Utilization of a self-assembled two-dimensional DNA nanostructure to arrange single-walled carbon nanotubes (SWNTs) into predetermined structures at controllable angles is presented. A specially designed DNA three-way junction (3WJ) composed of three double-stranded DNA arms containing single-stranded overhang sequences was prepared by annealing of partially complementary ssDNA sequences and ultrasonicated with SWNTs, resulting in DNA-3WJ/SWNT hybrid nanostructures. Utilization of DNA-3WJ not only allowed the precise dispersion of SWNTs but also acted as a rigid template for the self-assembly of SWNTs into three-armed junctions at an angle of approximately 120° to each other as visualized by scanning electron microscopy and atomic force microscopy. Prepared DNA-3WJ/SWNT nanostructures were also demonstrated to have the appropriate binding sites for fluorophores, providing a simple method for the fluorescent labeling of SWNTs. When ssDNA sequences forming the DNA-3WJ are ultrasonicated with SWNTs, followed by annealing of resulting ssDNA wrapped SWNTs, instead of hybrid junctions composed of three SWNT molecules, a web-like structure composed of interconnected SWNT junctions was obtained. The design approaches demonstrated here provide simple methods for the arrangement of SWNTs into desired nanostructures utilizing pre-assembled DNA nanostructures as linkers in aqueous solution through noncovalent interactions which can greatly contribute to efforts along the controlled assembly of SWNTs.

Project description:Gold standard datasets on protein complexes are key to inferring and validating protein-protein interactions. Despite much progress in characterizing protein complexes in the yeast Saccharomyces cerevisiae, numerous researchers still use as reference the manually curated complexes catalogued by the Munich Information Center of Protein Sequences database. Although this catalogue has served the community extremely well, it no longer reflects the current state of knowledge. Here, we report two catalogues of yeast protein complexes as results of systematic curation efforts. The first one, denoted as CYC2008, is a comprehensive catalogue of 408 manually curated heteromeric protein complexes reliably backed by small-scale experiments reported in the current literature. This catalogue represents an up-to-date reference set for biologists interested in discovering protein interactions and protein complexes. The second catalogue, denoted as YHTP2008, comprises 400 high-throughput complexes annotated with current literature evidence. Among them, 262 correspond, at least partially, to CYC2008 complexes. Evidence for interacting subunits is collected for 68 complexes that have only partial or no overlap with CYC2008 complexes, whereas no literature evidence was found for 100 complexes. Some of these partially supported and as yet unsupported complexes may be interesting candidates for experimental follow up. Both catalogues are freely available at: http://wodaklab.org/cyc2008/.
