Data on rhizosphere pH, phosphorus uptake and wheat growth responses upon TiO2 nanoparticles application.

ABSTRACT: In this study, the data sets and analyses provided the information on the characterization of titanium dioxide nanoparticles (TiO2 NPs), and their impacts on rhizosphere pH, and soil-bound phosphorus (P) availability to plants together with relevant parameters. For this purpose, wheat (Triticum aestivum L.) was cultivated in the TiO2 NPs amended soil over a period of 60 days. After harvesting, the soil and plants were analyzed to examine the rhizosphere pH, P availability in rhizosphere soil, uptake in roots and shoots, biomass produced, chlorophyll content and translocation to different plant parts monitored by SEM and EDX techniques in response to different dosages of TiO2 NPs. The strong relationship can be found among TiO2 NPs application, P availability, and plant growth.

Impact of Two Phosphorus Fertilizer Formulations on Wheat Physiology, Rhizosphere, and Rhizoplane Microbiota.

Project description:Phosphorus (P) is the second most important macronutrient for crop growth and a limiting factor in food production. Choosing the right P fertilizer formulation is important for crop production systems because P is not mobile in soils, and placing phosphate fertilizers is a major management decision. In addition, root microorganisms play an important role in helping phosphorus fertilization management by regulating soil properties and fertility through different pathways. Our study evaluated the impact of two phosphorous formulations (polyphosphates and orthophosphates) on physiological traits of wheat related to yield (photosynthetic parameters, biomass, and root morphology) and its associated microbiota. A greenhouse experiment was conducted using agricultural soil deficient in P (1.49%). Phenotyping technologies were used at the tillering, stem elongation, heading, flowering, and grain-filling stages. The evaluation of wheat physiological traits revealed highly significant differences between treated and untreated plants but not between phosphorous fertilizers. High-throughput sequencing technologies were applied to analyse the wheat rhizosphere and rhizoplane microbiota at the tillering and the grain-filling growth stages. The alpha- and beta-diversity analyses of bacterial and fungal microbiota revealed differences between fertilized and non-fertilized wheat, rhizosphere, and rhizoplane, and the tillering and grain-filling growth stages. Our study provides new information on the composition of the wheat microbiota in the rhizosphere and rhizoplane during growth stages (Z39 and Z69) under polyphosphate and orthophosphate fertilization. Hence, a deeper understanding of this interaction could provide better insights into managing microbial communities to promote beneficial plant-microbiome interactions for P uptake.

Differential responses of the rhizosphere microbiome structure and soil metabolites in tea (Camellia sinensis) upon application of cow manure.

Project description:BackgroundThe rhizosphere is the narrow zone of soil immediately surrounding the root, and it is a critical hotspot of microbial activity, strongly influencing the physiology and development of plants. For analyzing the relationship between the microbiome and metabolome in the rhizosphere of tea (Camellia sinensis) plants, the bacterial composition and its correlation to soil metabolites were investigated under three different fertilization treatments (unfertilized, urea, cow manure) in different growing seasons (spring, early and late summer).ResultsThe bacterial phyla Proteobacteria, Bacteroidetes, Acidobacteria and Actinobacteria dominated the rhizosphere of tea plants regardless of the sampling time. These indicated that the compositional shift was associated with different fertilizer/manure treatments as well as the sampling time. However, the relative abundance of these enriched bacteria varied under the three different fertilizer regimes. Most of the enriched metabolic pathways stimulated by different fertilizer application were all related to sugars, amino acids fatty acids and alkaloids metabolism. Organic acids and fatty acids were potential metabolites mediating the plant-bacteria interaction in the rhizosphere. Bacteria in the genera Proteiniphilum, Fermentimonas and Pseudomonas in spring, Saccharimonadales and Gaiellales in early summer, Acidobacteriales and Gaiellales in late summer regulated relative contents of organic and fatty acids.ConclusionThis study documents the profound changes to the rhizosphere microbiome and bacterially derived metabolites under different fertilizer regimes and provides a conceptual framework towards improving the performance of tea plantations.

Drivers of Phosphorus Uptake by Barley Following Secondary Resource Application.

Project description:Minable rock phosphate is a finite resource. Replacing mineral phosphorus (P) fertilizer with P-rich secondary resources is one way to manage P more efficiently, but the importance of physicochemical and microbial soil processes induced by secondary resources for plant P uptake is still poorly understood. Using radioactive-labeling techniques, the fertilization effects of dairy manure, fish sludge, meat bone meal, and wood ash were studied as P uptake by barley after 44 days and compared with those of water-soluble mineral P (MinP) and an unfertilized control (NoP) in a pot experiment with an agricultural soil containing little available P at two soil pH levels, approximately pH 5.3 (unlimed soil) and pH 6.2 (limed soil). In a parallel incubation experiment, the effects of the secondary resources on physicochemical and microbial soil processes were studied. The results showed that the relative agronomic efficiency compared with MinP decreased in the order: manure ≥fish sludge ≥wood ash ≥meat bone meal. The solubility of inorganic P in secondary resources was the main driver for P uptake by barley (Hordeum vulgare). The effects of secondary resources on physicochemical and microbial soil processes were of little overall importance. Application of organic carbon with manure resulted in microbial P immobilization and decreased uptake by barley of P derived from the soil. On both soils, P uptake by barley was best explained by a positive linear relationship with the H2O + NaHCO3-soluble inorganic P fraction in fertilizers or by a linear negative relationship with the HCl-soluble inorganic P fraction in fertilizers.

Phosphate solubilizing bacteria stimulate wheat rhizosphere and endosphere biological nitrogen fixation by improving phosphorus content.

Project description:Phosphate (P) availability often limits biological nitrogen fixation (BNF) by diazotrophic bacteria. In soil, only 0.1% of the total P is available for plant uptake. P solubilizing bacteria can convert insoluble P to plant-available soluble P (ionic P and low molecular-weight organic P). However, limited information is available about the effects of synergistic application of diazotrophic bacteria and P solubilizing bacteria on the nitrogenase activity of rhizosphere and nifH expression of endosphere. In this study, we investigated the effects of co-inoculation with a diazotrophic bacterium (Paenibacillus beijingensis BJ-18) and a P-solubilizing bacterium (Paenibacillus sp. B1) on wheat growth, plant and soil total N, plant total P, soil available P, soil nitrogenase activity and the relative expression of nifH in plant tissues. Co-inoculation significantly increased plant biomass (length, fresh and dry weight) and plant N content (root: 27%, shoot: 30%) and P content (root: 63%, shoot: 30%). Co-inoculation also significantly increased soil total N (12%), available P (9%) and nitrogenase activity (69%) compared to P. beijingensis BJ-18 inoculation alone. Quantitative real-time PCR analysis showed co-inoculation doubled expression of nifH genes in shoots and roots. Soil nitrogenase activity and nifH expression within plant tissues correlated with P content of soil and plant tissues, which suggests solubilization of P by Paenibacillus sp. B1 increased N fixation in soils and the endosphere. In conclusion, P solubilizing bacteria generally improved soil available P and plant P uptake, and considerably stimulated BNF in the rhizosphere and endosphere of wheat seedlings.

Controlling cellular uptake of nanoparticles with pH-sensitive polymers.

Project description:The major challenge in cancer therapy is to efficiently translocate drug molecules into cancer tumors without doing any damage to healthy tissues. Since there exist pH gradients between tumor and normal tissues, pH-sensitive materials may have great potential to overcome such challenge. Here, we report one new type of pH-responsive drug delivery system where pH-sensitive polymers are introduced to control the cellular uptake of nanoparticles under different pH environments through dissipative particle dynamics simulations. Interestingly, the behavior of cellular uptake of nanoparticles here exhibits "smart" pH-responsive properties: for lower and higher pH, the nanoparticles can be taken up by cell membranes, while for pH in middle range, the endocytosis is blocked. Further, it is found that receptor-ligand interactions as well as surface charge property of nanoparticles and membranes can also have important impacts on the endocytosis. The present study may give some significant insights into future stimulus-responsive medical materials design.

Localized metal solubilization in the rhizosphere of Salix smithiana upon sulfur application.

Project description:A metal-accumulating willow was grown under greenhouse conditions on a Zn/Cd-polluted soil to investigate the effects of sulfur (S(0)) application on metal solubility and plant uptake. Soil porewater samples were analyzed 8 times during 61 days of growth, while DGT-measured metal flux and O2 were chemically mapped at selected times. Sulfur oxidation resulted in soil acidification and related mobilization of Mn, Zn, and Cd, more pronounced in the rooted compared to bulk soil. Chemical imaging revealed increased DGT-measured Zn and Cd flux at the root-soil interface. Our findings indicated sustained microbial S(0) oxidation and associated metal mobilization close to root surfaces. The localized depletion of O2 along single roots upon S(0) addition indicated the contribution of reductive Mn (oxy)hydoxide dissolution with Mn eventually becoming a terminal electron acceptor after depletion of O2 and NO3(-). The S(0) treatments increased the foliar metal concentrations (mg kg(-1) dwt) up to 10-fold for Mn, (5810 ± 593), 3.3-fold for Zn (3850 ± 87.0), and 1.7-fold for Cd (36.9 ± 3.35), but had no significant influence on biomass production. Lower metal solubilization in the bulk soils should translate into reduced leaching, offering opportunities for using S(0) as environmentally favorable amendment for phytoextraction of metal-polluted soils.

Selenium uptake, translocation, subcellular distribution and speciation in winter wheat in response to phosphorus application combined with three types of selenium fertilizer.

Project description:BackgroundSelenium (Se) deficiency causes a series of health disorders in humans, and Se concentrations in the edible parts of crops can be improved by altering exogenous Se species. However, the uptake, transport, subcellular distribution and metabolism of selenite, selenate and SeMet (selenomethionine) under the influence of phosphorus (P) has not been well characterized.ResultsThe results showed that increasing the P application rate enhanced photosynthesis and then increased the dry matter weight of shoots with selenite and SeMet treatment, and an appropriate amount of P combined with selenite treatment increased the dry matter weight of roots by enhancing root growth. With selenite treatment, increasing the P application rate significantly decreased the concentration and accumulation of Se in roots and shoots. P1 decreased the Se migration coefficient, which could be attributed to the inhibited distribution of Se in the root cell wall, but increased distribution of Se in the root soluble fraction, as well as the promoted proportion of SeMet and MeSeCys (Se-methyl-selenocysteine) in roots. With selenate treatment, P0.1 and P1 significantly increased the Se concentration and distribution in shoots and the Se migration coefficient, which could be attributed to the enhanced proportion of Se (IV) in roots but decreased proportion of SeMet in roots. With SeMet treatment, increasing the P application rate significantly decreased the Se concentration in shoots and roots but increased the proportion of SeCys2 (selenocystine) in roots.ConclusionCompared with selenate or SeMet treatment, treatment with an appropriate amount of P combined with selenite could promote plant growth, reduce Se uptake, alter Se subcellular distribution and speciation, and affect Se bioavailability in wheat.

TiO2 nanoparticles functionalized by Pd nanoparticles for gas-sensing application with enhanced butane response performances.

Project description:Pd functionalized TiO2 nanoparticles were synthesized by a facile hydrothermal method. The structure, morphology, surface chemical states and surface area were characterized by X-ray diffraction (XRD), transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), and N2 adsorption-desorption isotherms, respectively. The as-synthesized pure and Pd functionalized TiO2 nanoparticles were used to fabricate indirect-heating gas sensor, and the gas-sensing characteristics towards butane were investigated. At the optimum temperature, the sensors possess good response, selectivity, response/recovery, repeatability as well as long-term stability. Especially for the high response, the response of 7.5 mol% Pd functionalized TiO2 nanoparticles based sensor reaches 33.93 towards 3000 ppm butane, which is about 9 times higher than that of pure TiO2 nanoparticles. The response and recovery time are 13 and 8 s, respectively. Those values demonstrate the potential of using as-synthesized Pd functionalized TiO2 nanoparticles as butane gas detection, particularly in the dynamic monitoring. Apart from these, a possible mechanism related to the enhanced sensing performance is also investigated.

The responses of root morphology and phosphorus-mobilizing exudations in wheat to increasing shoot phosphorus concentration.

Project description:The adaptations of root growth and rhizosphere processes for soil phosphorus (P) acquisition have been investigated intensively in wheat (Triticum aestivum). However, only a few studies paid attention to these responses to shoot P status. This study aimed at investigating the responses of root morphology and P-mobilizing exudation to increasing shoot P concentration. A broad range of wheat shoot P concentrations (1.0-7.1 mg per g dry weight) was set up with 11 rates of P supply: 0-1200 mg P per kg soil. Root morphology and exudation parameters were measured after 37 days of plant growth. Shoot dry biomass reached a maximum when shoot P concentration was 4.63 mg per g dry weight. The maximum shoot P concentration for total root length, specific root length and the proportion of fine root (diameter ≤ 0.2 mm) length to total root length was 3 mg per g dry weight. Rhizosphere acidification was positively correlated with shoot P concentration when this was <5 mg per g dry weight. Shoot P concentration did not change acid phosphatase activity in the rhizosphere. Citrate concentration in the rhizosphere was suppressed by increasing shoot P concentration. In contrast, malate concentration in the rhizosphere showed a positive correlation with shoot P concentration. In conclusion, wheat root morphological and P-mobilizing exudation traits showed different behaviours with increasing P deficiency stress. Maintaining root biomass and length is the major strategy rather than root exudation for wheat to cope with extreme P deficiency.

Uptake of pH-Sensitive Gold Nanoparticles in Strong Polyelectrolyte Brushes.

Project description:The impact of electrostatic attraction on the uptake of gold nanoparticles (AuNPs) into positively charged strong poly-[2-(Methacryloyloxy) ethyl] trimethylammonium chloride (PMETAC) polyelectrolyte brushes was investigated. In this work, PMETAC brushes were synthesized via surface-initiated atom transfer radical polymerization (Si-ATRP). PMETAC/AuNP composite materials were prepared by incubation of the polymer brush coated samples into 3-mercaptopropionic acid-capped AuNP (5 nm in diameter) suspension. The electrostatic interactions were tuned by changing the surface charge of the AuNPs through variations in pH value, while the charge of the PMETAC brush was not affected. Atomic-force microscopy (AFM), ellipsometry, UV/Vis spectroscopy, gravimetric analysis and transmission electron microscopy (TEM) were employed to study the loading and penetration into the polymer brush. The results show that the number density of attached AuNPs depends on the pH value and increases with increasing pH value. There is also strong evidence that the particle assembly is dependent on the pH value of the AuNP suspension. Incubation of PMETAC brushes in AuNP suspension at pH 4 led to the formation of a surface layer on top of the brush (2D assembly) due to sterical hindrance of the clustered AuNPs, while incubation in AuNP suspension at pH 8 led to deeper particle penetration into the brush (3D assembly). The straightforward control of particle uptake and assembly by tuning the charge density of the nanoparticle surface is a valuable tool for the development of materials for colorimetric sensor applications.
