Sigma Receptors as Endoplasmic Reticulum Stress "Gatekeepers" and their Modulators as Emerging New Weapons in the Fight Against Cancer.

ABSTRACT: Despite the interest aroused by sigma receptors (SRs) in the area of oncology, their role in tumor biology remains enigmatic. The predominant subcellular localization and main site of activity of SRs are the endoplasmic reticulum (ER). Current literature data, including recent findings on the sigma 2 receptor subtype (S2R) identity, suggest that SRs may play a role as ER stress gatekeepers. Although SR endogenous ligands are still unknown, a wide series of structurally unrelated compounds able to bind SRs have been identified. Currently, the identification of novel antiproliferative molecules acting via SR interaction is a challenging task for both academia and industry, as shown by the fact that novel anticancer drugs targeting SRs are in the preclinical-stage pipeline of pharmaceutical companies (i.e., Anavex Corp. and Accuronix). So far, no clinically available anticancer drugs targeting SRs are still available. The present review focuses literature advancements and provides a state-of-the-art overview of SRs, with emphasis on their involvement in cancer biology and on the role of SR modulators as anticancer agents. Findings from preclinical studies on novel anticancer drugs targeting SRs are presented in brief.

Project description:The root of Panax notoginseng (P. notoginseng) is one of the most highly valuable medicinal herbs in China owing to its pronounced hemostatic and restorative properties. Despite this important fact, growing P. notoginseng is seriously limited by root-rot diseases. In studies aimed at developing a solution to this problem, environment-friendly essential oils (EOs) of five medicinal plants of the family Zingiberaceae were tested for their inhibitory effects on the growth of three main soil pathogens associated with the root-rot diseases of P. notoginseng. The results showed that the EOs of Alpinia katsumadai Hayata and Zingiber officinale Roscoe promote significant reductions in the mycelium growth of the pathogen in vitro at a concentration of 50 mg mL-1, which is much higher than that needed (5 mg mL-1) to reduce growth by the positive control, flutriafol. Furthermore, the chemical components of the two EOs were determined by using GC-MS analysis. Eucalyptol was found to account for more than 30% of the oils of the two plants, with the second major components being geranyl acetate and ?-terpineol. These substances display different degrees of fungistasis in vitro. To further determine the effects of the EO of Zingiber officinale (Z. officinale) in vivo, soilless cultivation of P. notoginseng with pathogen inoculation was conducted in a greenhouse. Addition of the petroleum ether extract (approximately equal to EO) of Z. officinale to the culture matrix causes a large decrease in both the occurrence and severity of the P. notoginseng root-rot disease. The decreasing trend of net photosynthetic rate (Pn), stomatal conductance (gs), intercellular CO2 concentration (Ci), and transpiration rate (Tr) were all alleviated. In addition, the activities of catalase (CAT), peroxidase (POD), and the malondialdehyde (MDA) content were also largely reduced after pathogen infection, with the root activity being higher than that of the control. Taken together, the findings reveal that the EOs from plants might serve as promising sources of eco-friendly natural pesticides with less chemical resistance.

Project description:The prevalence of diabetes-associated cognitive dysfunction (DACD) has increased to 13.5%. Dementia, as the most severe DACD, is the second leading cause of death in patients with diabetes mellitus. Hence, the potential mechanisms of DACD for slowing or halting its progression need to be urgently explored. Given that the sigma-1 receptor (Sig-1R), a chaperone protein located in the endoplasmic reticulum (ER)-mitochondrion contact membranes to regulate ER stress (ERS), is associated with cognitive outcomes in neurodegenerative diseases, this study aimed to investigate the role of astrocytic Sig-1R in DACD and its underlying mechanism. Here, we examined the levels of ERS and complement component 3/3a (C3/C3a) from primary astrocytes with different concentrations of glucose and treatment. Subsequently, HT22 neurons were cultured in different astrocyte-conditioned medium, and the expression of synaptic proteins was detected. We constructed type 1 diabetes mellitus (T1DM) model to evaluate the astrocytic Sig-1R mechanism on synapse and cognitive function changes. In vitro, high glucose concentration downregulated Sig-1R and aggravated ERS in astrocytes, resulting in synapse deficits. PRE-084, a high-affinity and selective Sig-1R agonist, inhibited astrocytic ERS and complement cascades and restored synaptic damage, while the Sig-1R antagonist displayed the opposite results. Moreover, C3a receptor antagonist (C3aRA) could mimic the effect of PRE-084 and exerted neuroprotective effects. In vivo, PRE-084 substantially reduced ER-mitochondrion contact, activation of ERS, and C3/C3a secretion in mice with T1DM. Additionally, the synaptic loss and neurobehavioral dysfunction of mice with T1DM were less pronounced in both the PRE-084 and C3aRA treatment groups. These findings demonstrated that Sig-1R activation reduced the astrocytic ER-mitochondrion contact, ERS activation, and complement-mediated synaptic damage in T1DM. This study suggested the mechanisms and potential therapeutic approaches for treating DACD.
