Light-driven micro-tool equipped with a syringe function.

ABSTRACT: Leveraging developments in microfabrication open new possibilities for optical manipulation. With the structural design freedom from three-dimensional printing capabilities of two-photon polymerization, we are starting to see the emergence of cleverly shaped 'light robots' or optically actuated micro-tools that closely resemble their macroscopic counterparts in function and sometimes even in form. In this work, we have fabricated a new type of light robot that is capable of loading and unloading cargo using photothermally induced convection currents within the body of the tool. We have demonstrated this using silica and polystyrene beads as cargo. The flow speeds of the cargo during loading and unloading are significantly larger than when using optical forces alone. This new type of light robot presents a mode of material transport that may have a significant impact on targeted drug delivery and nanofluidics injection.

Standing Air Bubble-Based Micro-Hydraulic Capacitors for Flow Stabilization in Syringe Pump-Driven Systems.

Project description:Unstable liquid flow in syringe pump-driven systems due to the low-speed vibration of the step motor is commonly observed as an unfavorable phenomenon, especially when the flow rate is relatively small. Upon the design of a convenient and cost-efficient microfluidic standing air bubble system, this paper studies the physical principles behind the flow stabilization phenomenon of the bubble-based hydraulic capacitors. A bubble-based hydraulic capacitor consists of three parts: tunable microfluidic standing air bubbles in specially designed crevices on the fluidic channel wall, a proximal pneumatic channel, and porous barriers between them. Micro-bubbles formed in the crevices during liquid flow and the volume of the bubble can be actively controlled by the pneumatic pressure changing in the proximal channel. When there is a flowrate fluctuation from the upstream, the flexible air-liquid interface would deform under the pressure variation, which is analogous to the capacitive charging/discharging process. The theoretical model based on Euler law and the microfluidic equivalent circuit was developed to understand the multiphysical phenomenon. Experimental data characterize the liquid flow stabilization performance of the flow stabilizer with multiple key parameters, such as the number and the size of microbubbles. The developed bubble-based hydraulic capacitor could minimize the flow pulses from syringe pumping by 75.3%. Furthermore, a portable system is demonstrated and compared with a commercial pressure-driven flow system. This study can enhance the understanding of the bubble-based hydraulic capacitors that would be beneficial in microfluidic systems where the precise and stable liquid flow is required.

Light-Driven Reversible Shaping of Individual Azopolymeric Micro-Pillars.

Project description:Azopolymers are known to exhibit a strong light responsivity known as athermal photofluidization. Although the underlying physics is still under debate, athermal photofluidization has been demonstrated to trigger mass-migration according to the polarization of a proper illumination light. Here, a polymer blend is proposed wherein a commercial azo-polyelectrolyte is mixed with a passive polymer. The blend is patterned as an array of micro-pillars that are individually exposed to visible laser illumination. Thanks to the interplay between the two blend components, a reversible and controlled deformation of the micro-pillars by periodically tuning the laser polarization in time is demonstrated. A reduced mobility of the azo-compound allows to repeatibly elongate and rotate micro-pillars along specific directions, with no significant material flow outisde the initial volume and no significant degradation of the structure morphology over several cycles. The proposed work suggests new degrees of freedom in controlling the mechanical features of micro-patterned light-responsive materials that can be usefully exploited in many application fields.

Enhanced Light-Harvesting Efficiency and Adaptation: A Review on Visible-Light-Driven Micro/Nanomotors

Project description:As visible light accounts for a larger proportion of solar energy and is harmless to living organisms, it has the potential to be the energy source of micro/nanomotors, which transform visible-light energy into mechanical motion, for different applications, especially in environmental remediation. However, how to precisely control the motion of visible-light-driven micro/nanomotors (VLD-MNMs) and efficiently utilize the weak visible-light photon energy to acquire rapid motion are significant challenges. This review summarizes the most critical aspects, involving photoactive materials, propulsion mechanisms, control methods, and applications of VLD-MNMs, and discusses strategies to systematically enhance the energy-harvesting efficiency and adaptation. At first, the photoactive materials have been divided into inorganic and organic photoactive materials and comprehensively discussed. Then, different propulsion mechanisms of the current VLD-MNMs are presented to explain the improvement in the actuation force, speed, and environmental adaptability. In addition, considering the characteristics of easy control of VLD-MNMs, we summarized the direction, speed, and cluster control methods of VLD-MNMs for different application requirements. Subsequently, the potential applications of VLD-MNMs, e.g., in environmental remediation, micropumps, cargo delivery, and sensing in microscale, are presented. Finally, discussions and suggestions for future directions to enhance the energy-harvesting efficiency and adaptation of VLD-MNMs are provided.

Indirect optical trapping using light driven micro-rotors for reconfigurable hydrodynamic manipulation.

Project description:Optical tweezers are a highly versatile tool for exploration of the mesoscopic world, permitting non-contact manipulation of nanoscale objects. However, direct illumination with intense lasers restricts their use with live biological specimens, and limits the types of materials that can be trapped. Here we demonstrate an indirect optical trapping platform which circumvents these limitations by using hydrodynamic forces to exert nanoscale-precision control over aqueous particles, without directly illuminating them. Our concept is based on optically actuated micro-robotics: closed-loop control enables highly localised flow-fields to be sculpted by precisely piloting the motion of optically-trapped micro-rotors. We demonstrate 2D trapping of absorbing particles which cannot be directly optically trapped, stabilise the position and orientation of yeast cells, and demonstrate independent control over multiple objects simultaneously. Our work expands the capabilities of optical tweezers platforms, and represents a new paradigm for manipulation of aqueous mesoscopic systems.

A Compact, Syringe-Assisted, Vacuum-Driven Micropumping Device.

Project description:In this paper, a simple syringe‑assisted pumping method is introduced. The proposed fluidic micropumping system can be used instead of a conventional pumping system which tends to be large, bulky, and expensive. The micropump was designed separately from the microfluidic channels and directly bonded to the outlet of the microfluidic device. The pump components were composed of a dead‑end channel which was surrounded by a microchamber. A syringe was then connected to the pump structure by a short tube, and the syringe plunger was manually pulled out to generate low pressure inside the microchamber. Once the sample was loaded in the inlet, air inside the channel diffused into the microchamber through the PDMS (polydimethylsiloxane) wall, acting as a dragging force and pulling the sample toward the outlet. A constant flow with a rate that ranged from 0.8 nl · s - 1 to 7.5 nl · s - 1 was achieved as a function of the geometry of the pump, i.e., the PDMS wall thickness and the diffusion area. As a proof-of-concept, microfluidic mixing was demonstrated without backflow. This method enables pumping for point-of-care testing (POCT) with greater flexibility in hand-held PDMS microfluidic devices.

Light-Driven Liquid Crystal Circular Dammann Grating Fabricated by a Micro-Patterned Liquid Crystal Polymer Phase Mask.

Project description:As one of the diffractive optical elements, circular Dammann grating has shown its excellent versatility in practical applications. The electrically switchable Dammann grating has been extensively investigated; however, the research on the optically tunable circular Dammann grating has received less attention and reports on this subject have been insufficient in the past decade. In this paper, three-order and eight-order binary-phase liquid crystal circular Dammann gratings with two mutually orthogonal photo-induced alignments in every two adjacent alignment domains, fabricated by a micro-patterned liquid crystal polymer phase mask, are proposed to generate annular uniform-intensity patterns in the far field. A simple maskless optical tuning of an eight-order liquid crystal circular Dammann grating is demonstrated by controlling the polarization of an ultraviolet light as well as the energy dose. The proposed liquid crystal circular Dammann gratings with high efficiencies and desirable uniformities exhibit outstanding optical as well as electrical tunabilities, enabling the widespread prospective applications in adaptive photonic chips stimulated flexibly by only light or by the combination of light and electric field.

Ultra-low-current driven InGaN blue micro light-emitting diodes for electrically efficient and self-heating relaxed microdisplay.

Project description:InGaN-based micro-light-emitting diodes have a strong potential as a crucial building block for next-generation displays. However, small-size pixels suffer from efficiency degradations, which increase the power consumption of the display. We demonstrate strategies for epitaxial structure engineering carefully considering the quantum barrier layer and electron blocking layer to alleviate efficiency degradations in low current injection regime by reducing the lateral diffusion of injected carriers via reducing the tunneling rate of electrons through the barrier layer and balanced carrier injection. As a result, the fabricated micro-light-emitting diodes show a high external quantum efficiency of 3.00% at 0.1 A/cm2 for the pixel size of 10 × 10 μm2 and a negligible Jmax EQE shift during size reduction, which is challenging due to the non-radiative recombination at the sidewall. Furthermore, we verify that our epitaxy strategies can result in the relaxation of self-heating of the micro-light-emitting diodes, where the average pixel temperature was effectively reduced.

Open-source spring-driven syringe pump with 3D-printed components for microfluidic applications.

Project description:The operation of microfluidic devices requires precise and constant fluid flow. Microfluidic systems in low-resource settings require a portable, inexpensive, and electricity-free pumping approach due to the rising demand for microfluidics in point-of-care testing (POCT). Open-source alternatives, employing 3D printing and motors, offer affordability. However, using motors require electrical power, which often relies on external sources, hindering the on-site use of open-source pumps. This study introduces a spring-driven, 3D-printed syringe pump, eliminating the need for an external power source. The syringe pump is operated by the flat spiral spring's torque. By manually winding up the mainspring, the syringe pump can be operated without electricity. Various flow rates can be achieved by utilizing different syringe sizes and choosing the right gear combinations. All the parts of the syringe pump can be fabricated by 3D printing, requiring no additional components that require electricity. It operates by winding a mainspring and is user-friendly, allowing flow rate adjustments by assembling gears that modulate syringe plunger pushing velocity. The fabrication cost is $25-30 and can be assembled easily by following the instructions. We expect that the proposed syringe pump will enable the utilization of microfluidic technologies in resource-limited settings, promoting the adoption of microfluidics. Light-driven biocatalytic oxidation.

Project description:Enzymes are the catalyst of choice for highly selective reactions, offering nature-inspired approaches for sustainable chemical synthesis. Oxidative enzymes (e.g., monooxygenases, peroxygenases, oxidases, or dehydrogenases) catalyze a variety of enantioselective oxyfunctionalization and dehydrogenation reactions under mild conditions. To sustain the catalytic cycles of these enzymes, constant supply with or withdrawal of reducing equivalents (electrons) is required. Being redox by nature, photocatalysis appears a 'natural choice' to accomplish the electron-relay role, and many photoenzymatic oxidation reactions have been developed in the past years. In this contribution, we critically summarize the current developments in photoredoxbiocatalysis, highlight some promising concepts but also discuss the current limitations.
