Bismuth(III) reagents in steroid and terpene chemistry.

ABSTRACT: Steroid and terpene chemistry still have a great impact on medicinal chemistry. Therefore, the development of new reactions or "greener" processes in this field is a contemporaneous issue. In this review, the use of bismuth(III) salts, as "ecofriendly" reagents/catalysts, on new chemical processes involving steroids and terpenes as substrates will be focused. Special attention will be given to some mechanistic considerations concerning selected reactions.

Hydrothermal Crystallization of Bismuth Oxychlorides (BiOCl) Using Different Shape Control Reagents.

Project description:Bismuth oxychloride photocatalysts were obtained using solvothermal synthesis and different additives (CTAB-cetyltrimethylammonium bromide, CTAC-cetyltrimethylammonium chloride, PVP-polyvinylpyrrolidone, SDS-sodium dodecylsulphate, U-urea and TU-thiourea). The effect of the previously mentioned compounds was analyzed applying structural (primary crystallite size, crystal phase composition, etc.), morphological (particle geometry), optical (band gap energy) parameters, surface related properties (surface atoms' oxidation states), and the resulted photocatalytic activity. A strong dependency was found between the surface tension of the synthesis solutions and the overall morpho-structural parameters. The main finding was that the characteristics of the semiconductors can be tuned by modifying the surface tension of the synthesis mixture. It was observed after the photocatalytic degradation, that the white semiconductor turned to grey. Furthermore, we attempted to explain the gray color of BiOCl catalysts after the photocatalytic decompositions by Raman and XPS studies.

Phosphite Esters: Reagents for Exploring S-Nitrosothiol Chemistry.

Project description:The reactions between S-nitrosothiols and phosphite esters, including P(OPh)3, P(OBn)3, and P(OEt)3, were studied. Two different conjugated adducts, thiophosphoramidates and phosphorothioates, were formed, depending on the structures of the S-nitrosothiol substrate (e.g., primary vs tertiary). These reactions proceeded under mild conditions, and the reaction mechanisms were studied using experiments and calculations.

Organometallic Gold(III) Reagents for Cysteine Arylation.

Project description:An efficient method for chemoselective cysteine arylation of unprotected peptides and proteins using Au(III) organometallic complexes is reported. The bioconjugation reactions proceed rapidly (<5 min) at ambient temperature in various buffers and within a wide pH range (0.5-14). This approach provides access to a diverse array of S-aryl bioconjugates including fluorescent dye, complex drug molecule, affinity label, poly(ethylene glycol) tags, and a stapled peptide. A library of Au(III) arylation reagents can be prepared as air-stable, crystalline solids in one step from commercial reagents. The selective and efficient arylation procedures presented in this work broaden the synthetic scope of cysteine bioconjugation and serve as promising routes for the modification of complex biomolecules.

The UbiX flavin prenyltransferase reaction mechanism resembles class I terpene cyclase chemistry.

Project description:The UbiX-UbiD enzymes are widespread in microbes, acting in concert to decarboxylate alpha-beta unsaturated carboxylic acids using a highly modified flavin cofactor, prenylated FMN (prFMN). UbiX serves as the flavin prenyltransferase, extending the isoalloxazine ring system with a fourth non-aromatic ring, derived from sequential linkage between a dimethylallyl moiety and the FMN N5 and C6. Using structure determination and solution studies of both dimethylallyl monophosphate (DMAP) and dimethyallyl pyrophosphate (DMAPP) dependent UbiX enzymes, we reveal the first step, N5-C1' bond formation, is contingent on the presence of a dimethylallyl substrate moiety. Hence, an SN1 mechanism similar to other prenyltransferases is proposed. Selected variants of the (pyro)phosphate binding site are unable to catalyse subsequent Friedel-Crafts alkylation of the flavin C6, but can be rescued by addition of (pyro)phosphate. Thus, retention of the (pyro)phosphate leaving group is required for C6-C3' bond formation, resembling pyrophosphate initiated class I terpene cyclase reaction chemistry.

Amphiphilic Iodine(III) Reagents for the Lipophilization of Peptides in Water.

Project description:We report the functionalization of cysteine residues with lipophilic alkynes bearing a silyl group or an alkyl chain using amphiphilic ethynylbenziodoxolone reagents (EBXs). The reactions were carried out in buffer (pH 6 to 9), without organic co-solvent or removal of oxygen, either at 37 °C or room temperature. The transformation led to a significant increase of peptide lipophilicity and worked for aromatic thiols, homocysteine, cysteine, and peptides containing 4 to 18 amino acids. His6 -Cys-Ubiquitin was also alkynylated under physiological conditions. Under acidic conditions, the thioalkynes were converted into thioesters, which could be cleaved in the presence of hydroxylamine.

Catalytic, Tunable, One-Step Bismuth(III) Triflate Reaction with Alcohols: Dehydration Versus Dimerization.

Project description:Bi(OTf)3·xH2O is a powerful catalyst for the dehydration of tertiary alcohols into alkenes in apolar solvents. The reaction proceeds smoothly and selectively, with amounts as low as 0.01 mol % catalyst, in yields up to 93%. Moreover, in polar solvents, Bi(OTf)3·xH2O (0.1-1 mol %) selectively catalyzes the dimerization of the alcohols instead, forming new C-C bonds, in yields up to 96%. This mild, efficient, economic, and eco-friendly method is applicable across different chemical classes and amenable to several functional groups.

Tweaking the Charge Transfer: Bonding Analysis of Bismuth(III) Complexes with a Flexidentate Phosphane Ligand.

Project description:To account for the charge transfer and covalent character in bonding between P and Bi centers, the electronic structures of [P(C6H4-o-CH2SCH3)3BiCln](3-n)+ (n = 0-3) model species have been investigated computationally. On the basis of this survey a synthetic target compound with a dative P→Bi bond has been selected. Consecutively, the highly reactive bismuth cage [P(C6H4-o-CH2SCH3)3Bi]3+ has been accessed experimentally and characterized. Importantly, our experiments (single-crystal X-ray diffraction and solid-state NMR spectroscopy) and computations (NBO and AIM analysis) reveal that the P···Bi bonding in this trication can be described as a dative bond. Here we have shown that our accordion-like molecular framework allows for tuning of the interaction between P and Bi centers.

Crystal structure, antibacterial and cytotoxic activities of a new complex of bismuth(III) with sulfapyridine.

Project description:A new complex of Bi(III) and sulfapyridine was synthesized and characterized by elemental analysis, atomic absorption spectrometry, conductivity analysis, electrospray ionization mass spectrometry (ESI-MS), infrared spectroscopy, and single crystal X-ray diffraction methods. The antimicrobial and the cytotoxic activities of the compound were investigated. Elemental and conductivity analyses are in accordance to the formulation [BiCl₃(C₁₁H₁₁N₃O₂S)₃]. The structure of the complex reveals a distorted octahedral geometry around the bismuth atom, which is bound to three sulfonamidic nitrogens from sulfapyridine, acting as a monodentate ligand, and to three chloride ions. The presence of the compound in solution was confirmed by ESI-MS studies. The complex is 3 times more potent than the ligand against Salmonella typhimurium, 4 times against Staphylococcus aureus, Shigella dysenteriae, and Shigella sonnei and 8 times more potent against Pseudomonas aeruginosa and Escherichia coli. The compound inhibits the growth of chronic myelogenous leukemia cells with an IC₅₀ value of 44 μM whereas the free ligand has no effect up to 100 μM.
