Project description:The first C-SCF3 /SeCF3 cross-coupling reactions using gold redox catalysis [(MeDalphos)AuCl], AgSCF3 or Me4 NSeCF3 , and organohalides as substrates are reported. The new methodology enables a one-stop shop synthesis of aryl/alkenyl/alkynyl trifluoromethylthio- and selenoethers with a broad substrate scope (>60 examples with up to 97 % isolated yield). The method is scalable, and its robustness is evidenced by the late-stage functionalization of various bioactive molecules, which makes this reaction an attractive alternative in the synthesis of trifluoromethylthio- and selenoethers for pharmaceutical and agrochemical research and development.

Base-Promoted Cascade Reactions for the Synthesis of 3,3-Dialkylated Isoindolin-1-ones and 3-Methyleneisoindolin-1-ones.

Project description:Cascade reactions of ortho-carbonyl-substituted benzonitriles with ((chloromethyl)sulfonyl)benzenes as pronucleophiles led to new isoindolin-1-ones with a tetrasubstituted C-3 position or to (Z)-3-(sulfonyl-methylene)isoindolin-1-ones. The reactions start from readily available materials, are carried out under mild conditions, and do not require metal catalysis. Promoted only by the cheap and environmentally benign K2CO3 as the base, up to six elemental steps can be combined in a single pot. Hence, a sequential one-pot cascade/β-elimination/alkylation furnished useful intermediates for the synthesis of aristolactam natural products. The observed selectivity and the mechanism were investigated by DFT studies.

Bismuth(III) reagents in steroid and terpene chemistry.

Project description:Steroid and terpene chemistry still have a great impact on medicinal chemistry. Therefore, the development of new reactions or "greener" processes in this field is a contemporaneous issue. In this review, the use of bismuth(III) salts, as "ecofriendly" reagents/catalysts, on new chemical processes involving steroids and terpenes as substrates will be focused. Special attention will be given to some mechanistic considerations concerning selected reactions.

Difluorocarbene-Derived Trifluoromethylthiolation and [(18)F]Trifluoromethylthiolation of Aliphatic Electrophiles.

Project description:The first trifluoromethylthiolation and [(18)F]trifluoromethylthiolation of alkyl electrophiles with in situ generated difluorocarbene in the presence of elemental sulfur and external (radioactive) fluoride ion is described. This transition-metal-free approach is high yielding, compatible with a variety of functional groups, and operated under mild reaction conditions. The conceptual advantage of this exogenous-fluoride-mediated transformation enables unprecedented syntheses of [(18)F]CF3S-labeled molecules from most commonly used [(18)F]fluoride ions. The rapid radiochemical reaction time (≤1 min) and high functional-group tolerance allow access to a variety of aliphatic [(18)F]CF3S compounds in high yields.

Base-Promoted Chemodivergent Formation of 1,4-Benzoxazepin-5(4H)-ones and 1,3-Benzoxazin-4(4H)-ones Switched by Solvents.

Project description:The KOH-promoted chemodivergent benzannulation of ortho-fluorobenzamides with 2-propyn-1-ol can afford either 1,4-benzoxazepin-5(4H)-ones or 1,3-benzoxazin-4(4H)-ones in good yields with high selectivity, depending greatly upon the use of solvents. In the case of using DMSO, the intermolecular benzannulation produced seven-membered benzo-fused heterocycles of 1,4-benzoxazepin-5(4H)-ones, whereas in MeCN, the six-membered benzo-fused heterocycles of 1,3-benzoxazin-4(4H)-ones were formed. The KOH-promoted benzannulation proceeded most probably through the C-F nucleophilic substitution of ortho-fluorobenzamides with 2-propyn-1-ol to give the intermediate of ortho-[(2-propynyl)oxy]benzamide, which underwent the intramolecular hydroamidation in a different manner to afford either seven- or six-membered benzo-fused heterocycles.

Decarboxylative trifluoromethylthiolation of pyridylacetates.

Project description:Decarboxylative trifluoromethylthiolation of lithium pyridylacetates was achieved using N-(trifluoromethylthio)benzenesulfonimide as the electrophilic trifluoromethylthiolation reagent. The reaction afforded the corresponding trifluoromethyl thioethers in good yield. Furthermore, the preparation of lithium pyridylacetates by saponification of the corresponding methyl esters and subsequent decarboxylative trifluoromethylthiolation were performed in a one-pot fashion.

Deoxygenative trifluoromethylthiolation of carboxylic acids.

Project description:Here we describe a deoxygenative trifluoromethylthiolation method that yields trifluoromethyl thioesters from readily available carboxylic acids. The method is built upon an "umpolung" strategy where triphenylphosphine is used to first activate an electrophilic trifluoromethylthiolating reagent and then serves as an oxygen acceptor for the deoxygenation. The method is mild, efficient, broad-scope, and tolerant. It can be applied for the late-stage functionalization of numerous natural products and drug molecules containing a carboxylic acid group. The trifluoromethyl thioesters can be converted into trifluoromethyl thioethers by Pd-catalyzed decarbonylation.

Electron delocalization of robust high-nuclear bismuth-oxo clusters for promoted CO2 electroreduction.

Project description:The integration of high activity, selectivity and stability in one electrocatalyst is highly desirable for electrochemical CO2 reduction (ECR), yet it is still a knotty issue. The unique electronic properties of high-nuclear clusters may bring about extraordinary catalytic performance; however, construction of a high-nuclear structure for ECR remains a challenging task. In this work, a family of calix[8]arene-protected bismuth-oxo clusters (BiOCs), including Bi4 (BiOC-1/2), Bi8Al (BiOC-3), Bi20 (BiOC-4), Bi24 (BiOC-5) and Bi40Mo2 (BiOC-6), were prepared and used as robust and efficient ECR catalysts. The Bi40Mo2 cluster in BiOC-6 is the largest metal-oxo cluster encapsulated by calix[8]arenes. As an electrocatalyst, BiOC-5 exhibited outstanding electrochemical stability and 97% Faraday efficiency for formate production at a low potential of -0.95 V vs. RHE, together with a high turnover frequency of up to 405.7 h-1. Theoretical calculations reveal that large-scale electron delocalization of BiOCs is achieved, which promotes structural stability and effectively decreases the energy barrier of rate-determining *OCHO generation. This work provides a new perspective for the design of stable high-nuclear clusters for efficient electrocatalytic CO2 conversion.

The procedure exploits the peculiar properties of arylazo sulfones (having electron-rich or electron-poor substituents on the (hetero)aromatic ring) as photochemical precursors of aryl radicals and S-trifluoromethyl arylsulfonothioates as easy-to-handle trifluoromethylthiolating agents.
