Cytotoxic Labdane Diterpenes from Hedychium ellipticum Buch.-Ham. ex Sm.

ABSTRACT: In order to reveal the constituents and their biological activities, we carried out a phytochemical study on Hedychium ellipticum Buch.-Ham. ex Sm. (Zingiberaceae). Ten labdane diterpenoids (1-10) were isolated from the rhizomes of H. ellipticum for the first time. Their structures were identified on the basis of spectroscopic analyses including two-dimensional NMR and comparison with literature data. All of these compounds were evaluated for their antimycobacterial activity against Mycobacterium tuberculosis and cytotoxicity against KB, MCF7, NCI-H187 and Vero cells. The result showed that compounds 1 and 7 exhibited moderate activity against Mycobacterium tuberculosis and compounds 4, 6 and 7 displayed remarkable cytotoxic activity. This is the first report on the presence of all compounds in H. ellipticum and the first time that their structure activity relationship has been discussed.

New Cytotoxic Seco-Type Triterpene and Labdane-Type Diterpenes from Nuxia oppositifolia.

Project description:Chromatographic purification of the n-hexane and dichloromethane extracts of Nuxia oppositifolia aerial parts, growing in Saudi Arabia, resulted in the isolation and characterization of three new labdane-type diterpene acids, 2?-acetoxy-labda-7-en-15-oic acid (1), 2?-acetoxy-7-oxolabda-8-en-15-oic acid (2), 2?-acetoxy-6-oxolabda-7-en-15-oic acid (3), and one new seco-triterpene, 3,4-seco olean-12-en-3,30 dioic acid (4), together with 10 known lupane, oleanane and ursane-type triterpenes, as well as the common phytosterols, ?-sitosterol and stigmasterol (5-16). Their structures have been assigned on the basis of different spectroscopic techniques including 1D and 2D NMR. Moreover, 13 of the isolated compounds were tested on the human cancer cell lines HeLa (cervical), A549 (lung) and MDA (breast), and most of the compounds showed potent cytotoxic activities in vitro.

Dimeric labdane diterpenes: synthesis and antiproliferative effects.

Project description:Several diterpenes with the labdane skeleton show biological activity, including antiproliferative effects. Most of the research work on bioactive labdanes has been carried out on naturally occurring diterpenes and semisynthetic derivatives, but much less is known on the effects of diterpene dimers. The aim of the present work was to synthesize dimeric diterpenes from the labdane imbricatolic acid using esters, ethers and the triazole ring as linkers. Some 18 new derivatives were prepared and the compounds were evaluated for antiproliferative activity on human normal fibroblasts (MRC-5) and the following human tumor cell lines: AGS, SK-MES-1, J82 and HL-60. The diethers 8-10, differing in the number of CH? units in the linker, presented better antiproliferative activity with a maximum effect for the derivative 9. The best antiproliferative effect against HL-60 cells was found for compounds 3 and 17, with IC?? values of 22.3 and 23.2 ?M, lower than that found for the reference compound etoposide (2.23 ?M). The compounds 9, 17 and 11 were the most active derivatives towards AGS cells with IC?? values of 17.8, 23.4 and 26.1 ?M. A free carboxylic acid function seems relevant for the effect as several of the compounds showed less antiproliferative effect after methylation.

Sequencing and comparative genomics analysis in Senecio scandens Buch.-Ham. Ex D. Don, based on full-length cDNA library.

Project description:Senecio scandens Buch.-Ham. ex D. Don, an important antibacterial source of Chinese traditional medicine, has a widespread distribution in a few ecological habitats of China. We generated a full-length complementary DNA (cDNA) library from a sample of elite individuals with superior antibacterial properties, with satisfactory parameters such as library storage (4.30 × 106 CFU), efficiency of titre (1.30 × 106 CFU/mL), transformation efficiency (96.35%), full-length ratio (64.00%) and redundancy ratio (3.28%). The BLASTN search revealed the facile formation of counterparts between the experimental sample and Arabidopsis thaliana in view of high-homology cDNA sequence (90.79%) with e-values <1e - 50. Sequence similarities to known proteins indicate that the entire sequences of the full-length cDNA clones consist of the major of functional genes identified by a large set of microarray data from the present experimental material. For other Compositae species, a large set of full-length cDNA clones reported in the present article will serve as a useful resource to facilitate further research on the transferability of expressed sequence tag-derived simple sequence repeats (EST-SSR) development, comparative genomics and novel transcript profiles.

Labdane Diterpenes from the Fruits of Sinopodophyllum emodi.

Project description:Two new labdane diterpenes, sinoditerpene A (1) and B (2), were isolated from the fruits of Sinopodophyllum emodi, along with two known analogues 3 and 4. Their structures were established on the basis of extensive spectroscopic analysis. The isolation of compounds 1-4 represents the first report of diterpenes from the genus Sinopodophyllum. The cytotoxic activities of all isolated compounds were evaluated in comparison with 5-fluorouracil against the MCF-7 and HepG2 cell lines, towards which 3 showed more potent cytotoxicity.

Metabolomics reveals immunomodulation as a possible mechanism for the antibiotic effect of Persicaria capitata (Buch.-Ham. ex D. Don) H.Gross.

Project description:Introduction:In spite of advances in antibiotics, urinary tract infection (UTI) is still among the most common reasons for antibiotic medication worldwide. Persicaria capitata (Buch.-Ham. ex D. Don) H.Gross (P. capitata) is a herbal medicine used by the Miao people in China to treat UTI. However studies of its mechanism are challenging, owing to the complexity of P. capitata with multiple constituents acting on multiple metabolic pathways. Objective:The objective of this study was to explore the working mechanism of P. capitata on urinary tract infection. Methods:Relinqing® granule, which is solely made from aqueous extracts of the whole P. capitata plant, was used in this study. Urine metabolomics based on gas chromatography-mass spectroscopy was employed to assess the metabolic changes caused by administration of Relinqing® granule in a UTI mouse model. Female specific-pathogen-free Kunming mice were divided into control group (mock infection, saline treatment), model group (E.coli infection, saline treatment), Relinqing® group (E.coli infection, Relinqing® granule treatment), ciprofloxacin group (E.coli infection, ciprofloxacin treatment), and sham-Relinqing® group (no surgery, Relinqing® granule treatment). Results:The results showed that after the treatments, urine levels of itaconic acid in Relinqing® group increased by 4.9 fold and 11.3 fold compared with model and ciprofloxacin groups respectively. Itaconic acid is an endogenous antibacterial metabolite produced by macrophages, which also functions as a checkpoint for metabolic reprogramming of macrophage. Conclusion:Our findings suggest that this herbal medicine can cure urinary tract infection through modulation of immune system.

The complete chloroplast genome characteristics of <i>Polygala crotalarioides</i> Buch.-Ham. ex DC. (<i>Polygalaceae</i>) from Yunnan, China.

Project description:Polygala crotalarioides Buch.-Ham. ex DC. (Polygalaceae) is a perennial herbaceous plant widely distributed in southwest China. Its chloroplast genome sequence was obtained using Illumina NovaSeq sequencing technology, and then was assembled, annotated, and characterized. The total length of chloroplast genome is 164,269 bp and it contained a large (LCS, 82,697 bp) and a small (SSC, 8083 bp) single copy region, separated by a pair of inverted repeats (IRA/IRB, 36,744 bp). The overall GC content of genome was 36.8% and corresponding values of the LSC, SSC, and IR regions are 34.9%, 29.5%, and 39.7%, respectively. There are 135 genes that were predicted including 89 protein-coding, 38 tRNA, and 8 rRNA genes. The maximum-likelihood (ML) phylogenetic tree reconstructed by IQ-TREE indicated that P. crotalarioides had a strong genetic relationship with P. tenuifolia, P. sibirica, and P. japonica. In summary, the complete chloroplast genome of P. crotalarioides is reported for the first time, which is of great significance for future research on the evolutionary phylogeny and protection of Polygala.

Horizontal gene transfer from a flowering plant to the insular pine Pinus canariensis (Chr. Sm. Ex DC in Buch).

Project description:Horizontal gene transfer (HGT) is viewed as very common in the plant mitochondrial (mt) genome, but, to date, only one case of HGT has been found in gymnosperms. Here we report a new case of HGT, in which a mt nad5-1 fragment was transferred from an angiosperm to Pinus canariensis. Quantitative assay and sequence analyses showed that the foreign nad5-1 is located in the mt genome of P. canariensis and is nonfunctional. An extensive survey in the genus Pinus revealed that the angiosperm-derived nad5-1 is restricted to P. canariensis and present across the species' range. Molecular dating based on chloroplast DNA suggested that the HGT event occurred in the late Miocene after P. canariensis split from its closest relatives, and that the foreign copy became fixed in P. canariensis owing to drift during its colonization of the Canary Islands. The mechanism of this HGT is unclear but it was probably achieved through either direct cell-cell contact or external vectors. Our discovery provides evidence for an important role of HGT in plant mt genome evolution.

Signatures of volcanism and aridity in the evolution of an insular pine (Pinus canariensis Chr. Sm. Ex DC in Buch).

Project description:Oceanic islands of volcanic origin provide useful templates for the study of evolution because they are subjected to recurrent perturbations that generate steep environmental gradients that may promote adaptation. Here we combine population genetic data from nuclear genes with the analysis of environmental variation and phenotypic data from common gardens to disentangle the confounding effects of demography and selection to identify the factors of importance for the evolution of the insular pine P. canariensis. Eight nuclear genes were partially sequenced in a survey covering the entire species range, and phenotypic traits were measured in four common gardens from contrasting environments. The explanatory power of population substrate age and environmental indices were assessed against molecular and phenotypic diversity estimates. In addition, neutral genetic variability (FST) and the genetic differentiation of phenotypic variation (QST) were compared in order to identify the evolutionary forces acting on these traits. Two key factors in the evolution of the species were identified: (1) recurrent volcanic activity has left an imprint in the genetic diversity of the nuclear genes; (2) aridity in southern slopes promotes local adaptation in the driest localities of P. canariensis, despite high levels of gene flow among populations.

Climatic Signals in Tree Rings of Heritiera fomes Buch.-Ham. in the Sundarbans, Bangladesh.

Project description:Mangroves occur along the coastlines throughout the tropics and sub-tropics, supporting a wide variety of resources and services. In order to understand the responses of future climate change on this ecosystem, we need to know how mangrove species have responded to climate changes in the recent past. This study aims at exploring the climatic influences on the radial growth of Heritiera fomes from a local to global scale. A total of 40 stem discs were collected at breast height position from two different zones with contrasting salinity in the Sundarbans, Bangladesh. All specimens showed distinct tree rings and most of the trees (70%) could be visually and statistically crossdated. Successful crossdating enabled the development of two zone-specific chronologies. The mean radial increment was significantly higher at low salinity (eastern) zone compared to higher salinity (western) zone. The two zone-specific chronologies synchronized significantly, allowing for the construction of a regional chronology. The annual and monsoon precipitation mainly influence the tree growth of H. fomes. The growth response to local precipitation is similar in both zones except June and November in the western zone, while the significant influence is lacking. The large-scale climatic drivers such as sea surface temperature (SST) of equatorial Pacific and Indian Ocean as well as the El Niño-Southern Oscillation (ENSO) revealed no teleconnection with tree growth. The tree rings of this species are thus an indicator for monsoon precipitation variations in Bangladesh. The wider distribution of this species from the South to South East Asian coast presents an outstanding opportunity for developing a large-scale tree-ring network of mangroves.
