Predictive cues reduce but do not eliminate intrinsic response bias.

ABSTRACT: Predictive cues induce large changes in people's choices by biasing responses towards the expected stimulus category. At the same time, even in the absence of predictive cues, humans often exhibit substantial intrinsic response biases. Despite the ubiquity of both of these biasing effects, it remains unclear how predictive cues interact with intrinsic bias. To understand the nature of this interaction, we examined data across three previous experiments that featured a combination of neutral cues (revealing intrinsic biases) and predictive cues. We found that predictive cues decreased the intrinsic bias to about half of its original size. This result held both when bias was quantified as the criterion location estimated using signal detection theory and as the probability of choosing a particular stimulus category. Our findings demonstrate that predictive cues reduce but do not eliminate intrinsic response bias, testifying to both the malleability and rigidity of intrinsic biases.

Cognitive bias modification for energy drink cues.

Project description:Energy drink consumption is increasing worldwide, especially among young adults, and has been associated with physical and mental health problems. In two experiments, we tested the prediction that energy drink consumption is in part driven by biased cognitive processing (attentional and approach biases), with a view to modifying these to reduce consumption. Young adults (18-25 years) who regularly consume energy drinks completed the dot probe (Exp.1; N = 116) or approach-avoidance task (Exp.2; N = 110) to measure attentional and approach bias for energy drink cues, respectively. They then underwent a cognitive bias modification protocol where they were trained to direct their attention away from pictures of energy drink cans (Exp.1), or to push a joystick away from themselves in response to these pictures (Exp.2). Following a post-training assessment of attentional (Exp.1) or approach bias (Exp.2), energy drink consumption was measured by an ostensible taste test. Regular energy drink consumers showed both an attentional and an approach bias for energy drink cues. Cognitive bias modification successfully reduced both biases. However, neither attentional nor approach bias modification significantly reduced energy drink intake. The results lend some support to incentive sensitisation theory which emphasises the role of biased decision-making processes related to addictive behaviours.

Attentional bias toward cigarette cues in active smokers.

Project description:While it is well documented that substance users exhibit attentional bias toward addiction-related stimuli, the exact mechanism remains unclear.To differentiate between distinct aspects of attentional allocation in the smoking-cue attentional bias observed in smokers.Active smokers (AS) and non-smoking controls completed spatial cueing tasks with pairs of smoking and neutral pictorial cues to measure attentional capture, and an attentional blink task with either a smoking or neutral image appearing behind the first target (T1) to measure aspects of attention separate from capture. In addition, we tested groups of sports enthusiasts, and non-enthusiasts in corresponding tasks replacing smoking images with sports-related images to address the possibility that effects found in the smoking study were due simply to greater stimulus familiarity.Smoking cues reflexively capture smokers' attention, as AS showed a greater bias toward smoking cues in short stimulus-onset asynchrony (SOA; the time between the onset of two stimuli) trials, but not in trials with a longer SOA. These effects represent a facilitation of responding to smoking- versus neutral-cued targets, and were absent in the sports control task. The attentional blink effects were similar in the smoking- and sports-cue experiments: the special T1 resulted in better detection of the second target for the smokers and sports enthusiasts.Stimulus familiarity may contribute to some aspects of attentional bias in regular nicotine users, but selective quick capture of attention by smoking cues may be nicotine-habit specific.

Pleasant olfactory cues can reduce cigarette craving.

Project description:Cigarette craving is a cardinal feature of smoking, which is the leading preventable cause of death. Despite its clinical relevance, there remains a pressing need to develop new approaches for controlling craving. Although olfactory cues (OCs) are especially well suited to reduce affectively charged cravings, there has been surprisingly little research on the topic. We investigated the strategic use of OCs to reduce cigarette craving. Abstinent smokers (N = 232) initially sampled and rated a series of OCs. Participants then were exposed to in vivo smoking cues, which produced robust cigarette cravings. During peak craving, they were randomly assigned to sniff one of three types of OCs (all of which they had previously sampled) while their craving, and a set of responses thought to be associated with craving, were assessed. OCs that a participant had rated as pleasant reduced craving more than did exposure to odor blank (i.e., neutral) or tobacco-related OCs. This effect persisted over the course of 5 min. In addition, smokers with the most specific autobiographical memory systems were most responsive to the craving-reducing effects of pleasant OCs. About 90% of participants reported they could imagine using a pleasant OC to curb their craving in the natural environment. The present data suggest that OCs show promise for controlling cravings and highlight the need to conduct further research to test whether OCs may prove useful alone or in combination with existing approaches as a smoking cessation intervention. Aversive stimuli bias corticothalamic responses to motivationally significant cues.

Project description:Making predictions about future rewards or punishments is fundamental to adaptive behavior. These processes are influenced by prior experience. For example, prior exposure to aversive stimuli or stressors changes behavioral responses to negative- and positive-value predictive cues. Here, we demonstrate a role for medial prefrontal cortex (mPFC) neurons projecting to the paraventricular nucleus of the thalamus (PVT; mPFC→PVT) in this process. We found that a history of aversive stimuli negatively biased behavioral responses to motivationally relevant cues in mice and that this negative bias was associated with hyperactivity in mPFC→PVT neurons during exposure to those cues. Furthermore, artificially mimicking this hyperactive response with selective optogenetic excitation of the same pathway recapitulated the negative behavioral bias induced by aversive stimuli, whereas optogenetic inactivation of mPFC→PVT neurons prevented the development of the negative bias. Together, our results highlight how information flow within the mPFC→PVT circuit is critical for making predictions about motivationally-relevant outcomes as a function of prior experience.

Dual randomization of oligonucleotides to reduce the bias in ribosome-profiling libraries

Project description:Protein translation is at the heart of cellular metabolism and its in-depth characterization is key for many lines of research. Recently, ribosome profiling became the state-of-the-art method to quantitatively characterize translation dynamics at a transcriptome-wide level. However, the strategy of library generation affects its outcomes. Here, we present a modified ribosomeprofiling protocol starting from yeast, human cells and vertebrate brain tissue. We use a DNA linker carrying four randomized positions at its 5â and a reverse-transcription (RT) primer with three randomized positions to reduce artifacts during library preparation. The use of seven randomized nucleotides allows to efficiently detect library-generation artifacts. We find that the effect of polymerase chain reaction (PCR) artifacts is relatively small for global analyses when sufficient input material is used. However, when input material is limiting, our strategy improves the sensitivity of gene-specific analyses. Furthermore, randomized nucleotides alleviate the skewed frequency of specific sequences at the 3â end of ribosome-protected fragments (RPFs) likely resulting from ligase specificity. Finally, strategies that rely on dual ligation show a high degree of gene-coverage variation. Taken together, our approach helps to remedy two of the main problems associated with ribosome-profiling data. This will facilitate the analysis of translational dynamics and increase our understanding of the influence of RNA modifications on translation. Prior probability cues bias sensory encoding with increasing task exposure.

Project description:When observers have prior knowledge about the likely outcome of their perceptual decisions, they exhibit robust behavioural biases in reaction time and choice accuracy. Computational modelling typically attributes these effects to strategic adjustments in the criterion amount of evidence required to commit to a choice alternative - usually implemented by a starting point shift - but recent work suggests that expectations may also fundamentally bias the encoding of the sensory evidence itself. Here, we recorded neural activity with EEG while participants performed a contrast discrimination task with valid, invalid, or neutral probabilistic cues across multiple testing sessions. We measured sensory evidence encoding via contrast-dependent steady-state visual-evoked potentials (SSVEP), while a read-out of criterion adjustments was provided by effector-selective mu-beta band activity over motor cortex. In keeping with prior modelling and neural recording studies, cues evoked substantial biases in motor preparation consistent with criterion adjustments, but we additionally found that the cues produced a significant modulation of the SSVEP during evidence presentation. While motor preparation adjustments were observed in the earliest trials, the sensory-level effects only emerged with extended task exposure. Our results suggest that, in addition to strategic adjustments to the decision process, probabilistic information can also induce subtle biases in the encoding of the evidence itself.

Columnar-Intrinsic Cues Shape Premotor Input Specificity in Locomotor Circuits.

Project description:Control of movement relies on the ability of circuits within the spinal cord to establish connections with specific subtypes of motor neuron (MN). Although the pattern of output from locomotor networks can be influenced by MN position and identity, whether MNs exert an instructive role in shaping synaptic specificity within the spinal cord is unclear. We show that Hox transcription-factor-dependent programs in MNs are essential in establishing the central pattern of connectivity within the ventral spinal cord. Transformation of axially projecting MNs to a limb-level lateral motor column (LMC) fate, through mutation of the Hoxc9 gene, causes the central afferents of limb proprioceptive sensory neurons to target MNs connected to functionally inappropriate muscles. MN columnar identity also determines the pattern and distribution of inputs from multiple classes of premotor interneurons, indicating that MNs broadly influence circuit connectivity. These findings indicate that MN-intrinsic programs contribute to the initial architecture of locomotor circuits.

Excluding spatial sampling bias does not eliminate oversplitting in DNA-based species delimitation analyses.

Project description:DNA barcoding and DNA-based species delimitation are major tools in DNA taxonomy. Sampling has been a central debate in this context, because the geographical composition of samples affects the accuracy and performance of DNA barcoding. Performance of complex DNA-based species delimitation is to be tested under simpler conditions in absence of geographic sampling bias. Here, we present an empirical dataset sampled from a single locality in a Southeast-Asian biodiversity hotspot (Laos: Phou Pan mountain). We investigate the performance of various species delimitation approaches on a megadiverse assemblage of herbivorous chafer beetles (Coleoptera: Scarabaeidae) to infer whether species delimitation suffers in the same way from exaggerate infraspecific variation despite the lack of geographic genetic variation that led to inconsistencies between entities from DNA-based and morphology-based species inference in previous studies. For this purpose, a 658 bp fragment of the mitochondrial cytochrome c oxidase subunit 1 (cox1) was analyzed for a total of 186 individuals of 56 morphospecies. Tree-based and distance-based species delimitation methods were used. All approaches showed a rather limited match ratio (max. 77%) with morphospecies. Poisson tree process (PTP) and statistical parsimony network analysis (TCS) prevailingly over-splitted morphospecies, while 3% clustering and Automatic Barcode Gap Discovery (ABGD) also lumped several species into one entity. ABGD revealed the highest congruence between molecular operational taxonomic units (MOTUs) and morphospecies. Disagreements between morphospecies and MOTUs have to be explained by historically acquired geographic genetic differentiation, incomplete lineage sorting, and hybridization. The study once again highlights how important morphology still is in order to correctly interpret the results of molecular species delimitation.

Project description:This study investigated attentional bias toward game-related cues in Internet gaming disorder (IGD) using electrophysiological markers of late positive potential (LPP) and identifying the sources of LPP. In addition, the association between LPP and decision-making ability was investigated. The IGD (n = 40) and healthy control (HC; n = 39) participants viewed a series of game-related and neutral pictures, while their event-related potentials (ERPs) were recorded. LPPs were calculated as the mean amplitudes between 400 and 700 ms at the centro-parietal (CP3, CP1, Cpz, CP2, and CP4) and parietal (P3, P1, Pz, P2, and P4) electrode sites. The source activations of LPP were estimated using standardized low-resolution brain electromagnetic tomography (sLORETA). In addition, decision-making ability was evaluated by the Cambridge Gambling Task. Higher LPP amplitudes were found for game-related cues in the IGD group than in the HC group. sLORETA showed that the IGD group was more active in the superior and middle temporal gyri, which are involved in social perception, than in the HC group, whereas it was less active in the frontal area. Individuals with IGD have deficits in decision-making ability. In addition, in the HC group, the lower the LPP when looking at the game-related stimuli, the better the quality of decision-making, but not in the IGD group. Enhanced LPP amplitudes are associated with emotional arousal to gaming cues and decision-making deficits in IGD. In addition, source activities suggest that patients with IGD perceive game-related cues as social stimuli. LPP can be used as a neurophysiological marker of IGD.
