Scope and Limitations of ?-Valerolactone (GVL) as a Green Solvent to be Used with Base for Fmoc Removal in Solid Phase Peptide Synthesis.

ABSTRACT: GVL is a green solvent used in Fmoc-based solid-phase peptide synthesis. It is susceptible to ring opening in the presence of bases such as piperidines, which are used to remove the Fmoc protecting group. Here we studied the formation of the corresponding acyl piperidides by time-dependent monitoring using NMR. The results, corroborated by theoretical calculations, indicate that a solution of piperidines in GVL should be prepared daily for a better Fmoc removal.

Scope and Limitations of Fmoc Chemistry SPPS-Based Approaches to the Total Synthesis of Insulin Lispro via Ester Insulin.

Project description:We have systematically explored three approaches based on 9-fluorenylmethoxycarbonyl (Fmoc) chemistry solid phase peptide synthesis (SPPS) for the total chemical synthesis of the key depsipeptide intermediate for the efficient total chemical synthesis of insulin. The approaches used were: stepwise Fmoc chemistry SPPS; the "hybrid method", in which maximally protected peptide segments made by Fmoc chemistry SPPS are condensed in solution; and, native chemical ligation using peptide-thioester segments generated by Fmoc chemistry SPPS. A key building block in all three approaches was a Glu[O-?-(Thr)] ester-linked dipeptide equipped with a set of orthogonal protecting groups compatible with Fmoc chemistry SPPS. The most effective method for the preparation of the 51 residue ester-linked polypeptide chain of ester insulin was the use of unprotected peptide-thioester segments, prepared from peptide-hydrazides synthesized by Fmoc chemistry SPPS, and condensed by native chemical ligation. High-resolution X-ray crystallography confirmed the disulfide pairings and three-dimensional structure of synthetic insulin lispro prepared from ester insulin lispro by this route. Further optimization of these pilot studies could yield an efficient total chemical synthesis of insulin lispro (Humalog) based on peptide synthesis by Fmoc chemistry SPPS.

Amine-directed hydroboration: scope and limitations.

Project description:Iodine activation induces intramolecular hydroboration of homoallylic and bis-homoallylic amine boranes with good to excellent control of regiochemistry compared to control experiments using excess THF*BH 3. Deuterium labeling and other evidence confirm that the iodine-induced hydroboration reaction of homoallylic amine boranes occurs via an intramolecular mechanism equivalent to the classical 4-center process and without competing retro-hydroboration. Longer carbon chain tethers result in lower regioselectivity, whereas the shorter tether in allylic amines results in a switch to dominant intermolecular hydroboration. Regioselectivity in THF*BH 3 control experiments is higher for the allylic amine boranes compared to the iodine activation experiments, whereas the reverse is true for homoallylic amine borane activation.

γ-Valerolactone as Bio-Based Solvent for Nanofiltration Membrane Preparation.

Project description:γ-Valerolactone (GVL) was selected as a renewable green solvent to prepare membranes via the process of phase inversion. Water and ethanol were screened as sustainable non-solvents to prepare membranes for nanofiltration (NF). Scanning electron microscopy was applied to check the membrane morphology, while aqueous rose Bengal (RB) and magnesium sulphate (MgSO4) feed solutions were used to screen performance. Cellulose acetate (CA), polyimide (PI), cellulose triacetate (CTA), polyethersulfone (PES) and polysulfone (PSU) membranes were fine-tuned as materials for preparation of NF-membranes, either by selecting a suitable non-solvent for phase inversion or by increasing the polymer concentration in the casting solution. The best membranes were prepared with CTA in GVL using water as non-solvent: with increasing CTA concentration (10 wt% to 17.5 wt%) in the casting solution, permeance decreased from 15.9 to 5.5 L/m2·h·bar while RB rejection remained higher than 94%. The polymer solubilities in GVL were rationalized using Hansen solubility parameters, while membrane performances and morphologies were linked to viscosity measurements and cloudpoint determination of the casting solutions to better understand the kinetic and thermodynamic aspects of the phase inversion process.

Palladium Catalyst Recycling for Heck-Cassar-Sonogashira Cross-Coupling Reactions in Green Solvent/Base Blend.

Project description:The identification of a green, versatile, user-friendly, and efficient methodology is necessary to facilitate the use of Heck-Cassar-Sonogashira (HCS) cross-coupling reaction in drug discovery and industrial production in the pharmaceutical segment. The Heck-Cassar and Sonogashira protocols, using N-hydroxyethylpyrrolidone (HEP)/water/N,N,N',N'-tetramethyl guanidine (TMG) as green solvent/base mixture and sulfonated phosphine ligands, allowed to recycle the catalyst, always guaranteeing high yields and fast conversion under mild conditions, with aryl iodides, bromides, and triflates. No catalyst leakage or metal contamination of the final product were observed during the HCS recycling. To our knowledge, a turnover number (TON) up to 2375, a turnover frequency (TOF) of 158 h-1 , and a process mass intensity (PMI) around 7 that decreased around 3 after solvent, base, and palladium recovery, represent one of the best results to date using a sustainable protocol. The Heck-Cassar protocol using sSPhos was successfully applied to the telescoped synthesis of Erlotinib (TON: 1380; TOF: 46 h-1 ).

Removal of Copper Corrosion Products by Using Green Deep Eutectic Solvent and Bio-Derivative Cellulose Membrane.

Project description:A safe and environmentally friendly material for corrosion removal from metals is proposed in this article. Electrochemically corroded copper was selected as a target material, and a deep eutectic solvent (DES) composed of choline chloride and ascorbic acid, in a molar ratio of 2:1, was developed to this end. Aqueous solutions of the DES with a concentration above 70 wt% were found to be effective in the dissolution of patina and less aggressive towards other materials such as CaCO3, which is the main component of limestone. These concentrated DES solutions were integrated with either cotton swabs or cellulose-based membranes and used for the cleaning of electrochemically corroded copper. The membrane containing 80 wt% DES aqueous solution exhibited the most desirable cleaning ability in terms of speed and area selectivity. X-ray diffraction analysis of the corroded copper before and after the application of the membrane was performed to demonstrate the successful corrosion removal.

A sustainable metal and base-free direct amidation of esters using water as a green solvent.

Project description:Herein, we report a simple and efficient synthetic approach for direct amidation of esters via C(acyl)-O bond cleavage without any additional reagents or catalysts, using only water as a green solvent. Subsequently, the reaction byproduct is recovered and utilized for the next phase of ester synthesis. This method emphasized metal-free, additive-free, and base-free characteristics making it a new, sustainable, and eco-friendly way to realize direct amide bond formation. In addition, the synthesis of the drug molecule diethyltoluamide and the Gram-scale synthesis of a representative amide are demonstrated.

The Amadori Rearrangement for Carbohydrate Conjugation: Scope and Limitations.

Project description:The Amadori rearrangement was investigated for the synthesis of C-glycosyl-type neoglycoconjugates. Various amines including diamines, amino-functionalized glycosides, lysine derivatives, and peptides were conjugated with two different heptoses to generate non-natural C-glycosyl-type glycoconjugates of the d-gluco and d-manno series. With these studies, the scope and limitations of the Amadori rearrangement as a conjugation method have been exemplified with respect to the carbohydrate substrate, as well as the amino components.

Sugar-assisted glycopeptide ligation with complex oligosaccharides: scope and limitations.

Project description:We have previously shown sugar-assisted ligation (SAL) to be a useful method for the convergent construction of glycopeptides. However to date SAL has only been carried out on systems where the thiol auxiliary is attached to a monosaccharide. For SAL to be truly applicable to the construction of fully elaborated glycopeptides and glycoproteins, it must be possible to carry out the reaction when the thiol auxiliary is attached to more elaborate sugars, as these are frequently what are observed in nature. Here we examine the effects of glycosylation at C-3, C-4, and C-6 of the C-2 auxiliary-containing glycan. Model glycopeptides where synthesized chemoenzymatically and reacted with peptide thioesters used in our previous work. These studies reveal that SAL is sensitive to extended glycosylation on the auxiliary-containing sugar. While it is possible to carry out SAL with extended glycosylation at C-4 and C-6, the presence of glycosylation at C-3 prevents the ligation from occurring. Additionally, with glycosylation at C-4 the ligation efficiency is affected by the identity of the N-terminal AA, while the nature of the C-terminal residue of the peptide thioester does not appear to affect ligation efficiency. These studies provide useful guidelines in deciding when it is appropriate to use SAL in the synthesis of complex glycopeptides and glycoproteins and how to choose ligation junctions for optimal yield.
