Project description:PurposeThe goal of the present study is to develop polymeric matrix films loaded with a combination of free diclofenac sodium (DFSfree) and DFS:Ion exchange resin complexes (DFS:IR) for immediate and sustained release profiles, respectively.MethodsEffect of ratio of DFS and IR on the DFS:IR complexation efficiency was studied using batch processing. DFS:IR complex, DFSfree, or a combination of DFSfree + DFS:IR loaded matrix films were prepared by melt-cast technology. DFS content was 20% w/w in these matrix films. In vitro transcorneal permeability from the film formulations were compared against DFS solution, using a side-by-side diffusion apparatus, over a 6 h period. Ocular disposition of DFS from the solution, films and corresponding suspensions were evaluated in conscious New Zealand albino rabbits, 4 h and 8 h post-topical administration. All in vivo studies were carried out as per the University of Mississippi IACUC approved protocol.ResultsComplexation efficiency of DFS:IR was found to be 99% with a 1:1 ratio of DFS:IR. DFS release from DFS:IR suspension and the film were best-fit to a Higuchi model. In vitro transcorneal flux with the DFSfree + DFS:IR(1:1)(1 + 1) was twice that of only DFS:IR(1:1) film. In vivo, DFS solution and DFS:IR(1:1) suspension formulations were not able to maintain therapeutic DFS levels in the aqueous humor (AH). Both DFSfree and DFSfree + DFS:IR(1:1)(3 + 1) loaded matrix films were able to achieve and maintain high DFS concentrations in the AH, but elimination of DFS from the ocular tissues was much faster with the DFSfree formulation.ConclusionDFSfree + DFS:IR combination loaded matrix films were able to deliver and maintain therapeutic DFS concentrations in the anterior ocular chamber for up to 8 h. Thus, free drug/IR complex loaded matrix films could be a potential topical ocular delivery platform for achieving immediate and sustained release characteristics.

Project description:Protein mobility at solid-liquid interfaces can affect the performance of applications such as bioseparations and biosensors by facilitating reorganization of adsorbed protein, accelerating molecular recognition, and informing the fundamentals of adsorption. In the case of ion-exchange chromatographic beads with small, tortuous pores, where the existence of surface diffusion is often not recognized, slow mass transfer can result in lower resin capacity utilization. We demonstrate that accounting for and exploiting protein surface diffusion can alleviate the mass-transfer limitations on multiple significant length scales. Although the surface diffusivity has previously been shown to correlate with ionic strength (IS) and binding affinity, we show that the dependence is solely on the binding affinity, irrespective of pH, IS, and resin ligand density. Different surface diffusivities give rise to different protein distributions within the resin, as characterized using confocal microscopy and small-angle neutron scattering (length scales of micrometer and nanometer, respectively). The binding dependence of surface diffusion inspired a protein-loading approach in which the binding affinity, and hence the surface diffusivity, is modulated by varying IS. Such gradient loading increased the protein uptake efficiency by up to 43%, corroborating the importance of protein surface diffusion in protein transport in ion-exchange chromatography.

Project description:We introduce fluoride-selective anion exchange resin sorbents as sensitisers into membranes for water-gated field effect transistors (WGTFTs). Sorbents were prepared via metal (La or Al)-loading of a commercial macroporous aminophosphonic acid resin, PurometTM MTS9501, and were filled into a plasticised poly(vinyl chloride) (PVC) phase transfer membrane. We found a potentiometric response (membrane potential leading to WGTFT threshold shift) to fluoride following a Langmuir-Freundlich (LF) adsorption isotherm with saturated membrane potential up to ~480 mV, extremely low characteristic concentration c1/2 = 1/K, and picomolar limit of detection (LoD), even though ion exchange did not build up charge on the resin. La-loading gave a superior response compared to Al-loading. Membrane potential characteristics were distinctly different from charge accumulating sensitisers (e.g., organic macrocycles) but similar to the Cs+ (cation) selective ion-exchanging zeolite mineral 'mordenite'. We propose a mechanism for the observed threshold shift and investigate interference from co-solutes. Strong interference from carbonate was brought under control by 'diluting' metal loading in the resin. This work sets a template for future studies using an entirely new 'family' of sensitisers in applications where very low limit of detection is essential such as for ions of arsenic, mercury, copper, palladium, and gold.

Project description:Arid areas often contain brackish groundwater that has a salinity exceeding 500 mg/L. This poses several challenges to the users of the water such as a salty taste and damage to household appliances. Desalination can be one of the key solutions to significantly lower the salinity and solute content of the water. However, the technology requires high energy inputs as well as managing waste products. This paper presents the fabrication of ultrafiltration heterogeneous ion exchange membranes for brackish groundwater treatment. Scanning electron microscopy (SEM) images showed a relatively uniform resin particle distribution within the polymer matrix. The mean roughness of the cation exchange membrane (CEM) and anion exchange membrane (AEM) surfaces increased from 42.12 to 317.25 and 68.56 to 295.95 nm, respectively, when resin loading was increased from 1 to 3.5 wt %. Contact angle measures suggested a more hydrophilic surface (86.13 to 76.26° and 88.10 to 74.47° for CEM and AEM, respectively) was achieved with greater resin loading rates. The ion exchange capacity (IEC) of the prepared membranes was assessed using synthetic groundwater in a dead-end filtration system and removal efficiency of K+, Mg2+, and Ca2+ were 56.0, 93.5, and 85.4%, respectively, for CEM with the highest resin loading. Additionally, the anion, NO3- and SO42- removal efficiency was 84.2% and 52.4%, respectively, for the AEM with the highest resin loading. This work demonstrates that the prepared ultrafiltration heterogeneous ion exchange membranes have potential for selective removal for of ions by ion exchange, under filtration conditions at low pressure of 0.05 MPa.

Project description:A new approach to process Saccharina latissima algal biomass was developed using sodium citrate and a polyvalent cation-specific resin to sequentially extract the alginate into several usable fractions. The fractionation was performed in a cyclic manner, utilizing a stepwise removal of the native polyvalent ions present in the algae to isolate fractions of alginate with different solubility in the presence of these ions. Sodium citrate was used in different concentrations in the extraction solution to remove polyvalent cations to adjust the alginate liberation while AMBERLITE IRC718 resin was added to further remove these ions and regenerate the extraction solution. Alginate was recovered by acid precipitation and analyzed for its uronic acid composition and molecular weight, and the carbohydrate compositions of the insoluble and soluble parts of the algal biomass residue were determined. Finally, the fractionation method was assessed with a life cycle analysis to determine the energy and water efficiency as well as the greenhouse gas emissions and the results were compared to conventional alkaline extraction. The results indicate that the energy and water use as well as the emissions are considerably lower for the cyclic extraction in comparison with the conventional methods.

Project description:Esterification kinetics on acetic acid with isopropyl alcohol was studied in an intensified fixed bed reactor at 333-353 K with Amberlyst 36 Wet. The effects of volume flow rate, molar ratio of reactants, catalyst loading, and operating temperature were investigated and optimized. The method of UNIFAC was applied to calculate the activity coefficient of each component for correcting the nonideality of the solution. Reaction enthalpy, entropy, and Gibbs free energy were calculated in different cases. The pseudohomogeneous model, Eley-Rideal model, and Langmuir-Hinshelwood-Hougen-Watson model were used to establish kinetic equations of the reaction conducted in the IFBR. It was proved that the LHHW model can accurately describe the esterification kinetics in the intensified fixed bed reactor.

Project description:Conventional pollution monitoring strategies for heavy metals are often costly and unpractical. Innovative sampling and analytical approaches are therefore needed to efficiently monitor large areas. This study presents a novel, simple, fast, and inexpensive method to monitor heavy metal pollution that uses cation-exchange resin sachets and the micro-XRF core-scanning technique (XRF-CS). The resin passive samplers act as concentrators of cationic species and can be readily deployed spatially and temporally to record pollution signals. The large number of analytical tasks are then overcome by the fast and non-destructive XRF-CS to precisely assess elemental concentrations. Quantifying element loading involves direct comparison with a set of identically prepared and scanned resin reference standards containing Ca, Ti, Cr, Mn, Ni, Cu, Zn, Pb. The results show that within the test range (from 0-1000?s?mg kg-1), the calibration lines have excellent regressions (R2???0.97), even at the shortest exposure time (1?s). A pilot field survey of a suspected polluted area in central Taiwan, where 30 resin sachets had been deployed, identified a pollution hot spot in a rapid and economical manner. Therefore, this approach has the potential to become a valuable tool in environmental monitoring and forensics.

Project description:Ion-exchange resins are commonly used to treat complications such as hyperkalemia, hyperphosphatemia, and hypercholesterolemia. Gastrointestinal complications may occur as side effects of such treatments. Sodium and calcium polystyrene sulfonate (PS-Ca) are cation-exchange resins comprising an insoluble structure that binds to potassium ions in the digestive tract and exchanges them with sodium and calcium ions, respectively, to promote their elimination. PS crystals are rhomboid, refractive, and basophilic in hematoxylin and eosin staining. To differentiate PS crystals from other ion-exchange resin crystals such as sevelamer and cholestyramine, periodic acid-Schiff, Ziehl-Neelsen, and Congo red staining are usually performed. Here, correlative light and electron microscopy (CLEM)-energy-dispersive X-ray spectroscopy and the NanoSuit method (CENM) was applied to perform a definitive identification of ion-exchange resins. CENM could detect sulfur in PS crystals without destroying the glass slides. Notably, PS retained its ion-exchange ability to bind potassium in paraffin sections. Differential diagnosis of anion-exchange resins, such as sevelamer and cholestyramine, was possible using these characteristics. The phosphorus:carbon ratio was higher in sevelamer than in cholestyramine after soaking paraffin sections in a phosphate solution. Therefore, CENM may be used for the differential pathological diagnosis of ion-exchange resins in paraffin sections.

Project description:Artificial photosynthesis relies on the availability of semiconductors that are chemically stable and can efficiently capture solar energy. Although metal oxide semiconductors have been investigated for their promise to resist oxidative attack, materials in this class can suffer from chemical and photochemical instability. Here we present a methodology for evaluating corrosion mechanisms and apply it to bismuth vanadate, a state-of-the-art photoanode. Analysis of changing morphology and composition under solar water splitting conditions reveals chemical instabilities that are not predicted from thermodynamic considerations of stable solid oxide phases, as represented by the Pourbaix diagram for the system. Computational modelling indicates that photoexcited charge carriers accumulated at the surface destabilize the lattice, and that self-passivation by formation of a chemically stable surface phase is kinetically hindered. Although chemical stability of metal oxides cannot be assumed, insight into corrosion mechanisms aids development of protection strategies and discovery of semiconductors with improved stability.
