The complete mitochondrial genome of Entylia carinata (Hemiptera: Membracidae).

ABSTRACT: The complete mitochondrial genome of the treehopper species, Entylia carinata, was sequenced using the next-generation sequencing method. The completely assembled mitochondrial genome is 15,483?bp long. The genome follows the typical invertebrate mitochondrial gene arrangement, and includes 13 protein-coding genes, 22 tRNA genes, two rRNA genes and one A?+?T-rich region. Two tandem repeats were also identified within the A?+?T-rich region.

Project description:The complete Eclipophleps carinata mitogenome was sequenced through Illumina HiSeq 2500 platform and the resulting data were analyzed in this paper. The mitochondrial genome of E. carinata is a typical circular DNA molecule of 15,781 bp with 37 genes and 74.5% A + T content, which encoded 13 protein-coding genes (PCGs), 22 tRNA genes, two rRNA genes, and the control region. The E. carinata mitochondrial genome and 27 mitochondrial genomes (downloaded from NCBI) were employed to construct phylogenetic tree, in which Ruspolia dubia and Teleogryllus occipitails were the outgroups. Phylogenetic reconstruction validated the taxonomic status of E. carinata, which was placed in the monophyletic Gomphocerinae in Acrididae.

The complete mitogenome of Lophops carinata (Hemiptera: Fulgoroidea: Lophopidae).

Project description:In the present study, the complete mitochondrial genome of Lophops carinata (Hemiptera: Lophopidae) was sequenced for the first time through next-generation sequencing. The complete mitogenome of L. carinata is 15,553 bp in length, with the typical gene content and arrangement usually observed in Hexapods. The mitogenome consisted of 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes, and 1 D-loop. The overall nucleotide composition of the mitogenome was 44.6% A, 14.0% C, 8.3% G, and 33.2% T, with an A + T bias of 77.8%. Phylogenetic analyses from 12 Fulgoroidea species by maximum likelihood were consistent and well supported the basal position of Delphacidae, a close affinity among the families Ricaniidae, Issidae, and Flatidae, and a close relationship between Achilidae and Fulgoridae. And L. carinata belong to a separate lineage, located in the middle of the phylogenetic tree.

Project description:In this paper, we determined the complete mitochondrial genome of Chinese Daphnia carinata for the first time by the long and accurate polymerase chain reaction and primer-walking methods. It was 15,245 bp in length, with an A + T content of 70.35%, containing 37 typical animal mitochondrial genes and an A + T-rich region. The COI gene started with ACTA. All the 22 typical tRNA genes had a classical cloverleaf structure except for trnS1, in which the D-stem pairings in the DHU arm were absent.

The Complete Mitochondrial Genome of Ugyops sp. (Hemiptera: Delphacidae).

Project description:The complete mitochondrial genome (mitogenome) of Ugyops sp. (Hemiptera: Delphacidae) was sequenced, making it the first determined mitogenome from the subfamily Asiracinae, the basal clade of the family Delphacidae. The mitogenome was 15,259 bp in length with A + T content of 77.65% and contained 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), two ribosomal RNA genes (rRNAs), and a control region. The gene order was identical with that of the ancestral insect. The nucleotide composition analysis indicated that the whole mitogenome was strongly A-skewed (0.288) and highly C-skewed (-0.270). For PCGs on the J-strand, the AT skew was positive, and the GC skew was negative. All PCGs started with canonical ATN codons, except for cox1 and nad5, which used CTG and GTG as start codon, respectively. All tRNAs could fold into typical cloverleaf secondary structures, with the exception of trnS1 (AGN), in which the dihydrouridine arm was reduced to a simple loop. The control region included a poly-T stretch downstream of the small rRNA gene (rrnS), a subregion of higher A + T content and tandemly repeated sequence near trnI. The mitogenome of Ugyops sp. could be very helpful in exploring the diversity and evolution of mitogenomes in Delphacidae.

The phylogenetic analyses based on concatenated thirteen protein-coding genes of mitogenomes recover the monophyly of Auchenorrhyncha (Fulgoromorpha + Cicadomorpha) and Sternorrhyncha as a sister group to Auchenorrhyncha. Complete mitochondrial genome of Liorhyssus hyalinus (Hemiptera: Rhopalidae).

Project description:The mitogenome of Liorhyssus hyalinus (Fabricius, 1794), decoded using next-generation sequencing, is the first report of Liorhyssus. The mitogenomic size was 16,355 bp with 41.99% A, 33.44% T, 14.53% C, and 10.05% G (OM328158). The phylogenetic tree, constructed with the amino acid sequences of 13 protein-coding genes, showed that L. hyalinus clustered together with other species in Rhopalidae, which supported the monophyly of each family in Pentatomomorpha.

Structural Characteristics of Mitochondrial Genomes of Eight Treehoppers (Hemiptera: Membracidae: Centrotinae) and Their Phylogenetic Implications.

Project description:Complete mitochondrial genomes were newly sequenced for eight species of the treehopper subfamily Centrotinae (Hemiptera: Membracidae), four of which represent genera for which mitogenomes were not previously available. The new mitogenomes are generally similar in overall structure, gene order, base composition, and nucleotide content to those of previously sequenced species of the subfamily. Phylogenetic analyses were conducted using both maximum likelihood and Bayesian inference methods based on three separate nucleotide sequence datasets in which RNA gene sequences and/or third codon positions were either included or excluded from the concatenated protein-coding gene alignments. The results are consistent with previous phylogenies based on morphology and partial nuclear genome data, except for the lack of support for the monophyly of Leptocentrini. These results show that mitogenome sequences are informative of both ancient and recent divergence patterns within Centrotinae.

Comparative Genomics of the Dual-Obligate Symbionts from the Treehopper, Entylia carinata (Hemiptera: Membracidae), Provide Insight into the Origins and Evolution of an Ancient Symbiosis.

Project description:Insect species in the Auchenorrhyncha suborder (Hemiptera) maintain ancient obligate symbioses with bacteria that provide essential amino acids (EAAs) deficient in their plant-sap diets. Molecular studies have revealed that two complementary symbiont lineages, "Candidatus Sulcia muelleri" and a betaproteobacterium ("Ca. Zinderia insecticola" in spittlebugs [Cercopoidea] and "Ca. Nasuia deltocephalinicola" in leafhoppers [Cicadellidae]) may have persisted in the suborder since its origin ?300?Ma. However, investigation of how this pair has co-evolved on a genomic level is limited to only a few host lineages. We sequenced the complete genomes of Sulcia and a betaproteobacterium from the treehopper, Entylia carinata (Membracidae: ENCA), as the first representative from this species-rich group. It also offers the opportunity to compare symbiont evolution across a major insect group, the Membracoidea (leafhoppers?+?treehoppers). Genomic analyses show that the betaproteobacteria in ENCA is a member of the Nasuia lineage. Both symbionts have larger genomes (Sulcia?=?218?kb and Nasuia?=?144?kb) than related lineages in Deltocephalinae leafhoppers, retaining genes involved in basic cellular functions and information processing. Nasuia-ENCA further exhibits few unique gene losses, suggesting that its parent lineage in the common ancestor to the Membracoidea was already highly reduced. Sulcia-ENCA has lost the abilities to synthesize menaquinone cofactor and to complete the synthesis of the branched-chain EAAs. Both capabilities are conserved in other Sulcia lineages sequenced from across the Auchenorrhyncha. Finally, metagenomic sequencing recovered the partial genome of an Arsenophonus symbiont, although it infects only 20% of individuals indicating a facultative role.

The complete mitochondrial genome of Nysius fuscovittatus (Hemiptera: Lygaeidae).

Project description:Here, we sequenced and annotated the complete mitochondrial genome (mitogenome) of Nysius fuscovittatus (Hemiptera: Lygaeidae). This mitogenome was 14575 bp long, including 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), 2 ribosomal RNA unit genes (rrnL and rrnS), and a putative control region. All genes were arranged in the same order as that of most true bugs. Eleven PCGs started with a typical ATN, and the remaining two PCGs started with TTA (nad4L) and TTG (cox1). The N. fuscovittatus mitogenome with an A + T content of 76.42% showed a positive AT-skew (0.15) and a negative GC-skew (-0.15). With the exception of trnS1 that lacked the dihydrouridine arm, all tRNAs had a typical cloverleaf secondary structure. Phylogenetic analysis based on the concatenated nucleotide sequences of the 13 PCGs showed that N. fuscovittatus clustered with other three Lygaeidae species.
