This mitochondrial genome has 34,787?bp in length and consists of 65 genes including 37 protein-coding, three rRNA, and 25 tRNA genes. The overall GC content of the genome is 36.5%.

Project description:Sargassum kjellmanianum plays an important role in marine ecosystem and biological resource utilization. Here, we analyzed the characteristics and evolution of mitochondrial genome of S. kjellmanianum by paired-end sequencing. The complete mitogenome comprises a circular DNA molecule measuring 34,723 bp in length with 35 coding genes. The circular genome consisted of seven protein-coding genes, 25 tRNA genes, and three rRNA genes, with a total length of 26,553 bp. The overall A + T content was 63.43%, higher than the G + C content (36.57%). Phylogenetic analysis confirmed S. kjellmanianum was closely clustered with S. muticum.

Project description:Sargassum hemiphyllum (Turner) C. Agardh is an important brown macroalga. In this study, we presented the complete chloroplast genome of its variety S. hemiphyllum var. chinense using genome skimming approach. Circular mapping revealed its sequence length was 124,319 bp, with a large single-copy region (LSC, 73,505 bp) and a small single copy region (SSC, 39,922 bp) separated by a pair of inverted repeats (IRs, 5446 bp). Its chloroplast genome contained 173 genes, including 139 protein-coding, 6 rRNA, and 28 tRNA genes. The phylogenetic analysis indicated that S. hemiphyllum var. chinense was closely related with S. confusum.

In this study, we characterized the complete chloroplast genome sequence of S. fusiforme using PacBio long-read sequencing technology. It had a circular mapping molecular with the length of 124,286?bp, with a large single-copy region (LSC, 73,437?bp) and a small single copy region (SSC, 40,131?bp) separated by a pair of inverted repeats (IRs, 5,359?bp). The cp genome contained 173 genes including 139 protein-coding, 6 rRNA, and 28 tRNA genes. The phylogenomic analysis indicated that S. fusiforme is closely related to S. thunbergii.

Historical biogeography of the widespread macroalga Sargassum (Fucales, Phaeophyceae).

Project description:Sargassum is a cosmopolitan brown algal genus spanning the three ocean basins of the Atlantic, Pacific and Indian Oceans, inhabiting temperate, subtropical and tropical habitats. Sargassum has been postulated to have originated in the Oligocene epoch approximately 30 mya according to a broad phylogenetic analysis of brown macroalgae, but its diversification to become one of the most widespread and speciose macroalgal genera remains unclear. Here, we present a Bayesian molecular clock study, which analyzed data from the order Fucales of the brown algal crown radiation (BACR) group to reconstruct a time-calibrated phylogeny of the Sargassum clade. Our phylogeny included a total of 120 taxa with 99 Sargassum species sampled for three molecular markers - ITS-2, cox3 and rbcLS - calibrated with an unambiguous Sargassaceae fossil from between the lower and middle Miocene. The analysis revealed a much later origin of Sargassum than expected at about 6.7 mya, with the genus diversifying since approximately 4.3 mya. Current geographic distributions of Sargassum species were then analyzed in conjunction with the time-calibrated phylogeny using the dispersal-extinction-cladogenesis (DEC) model to estimate ancestral ranges of clades in the genus. Results strongly support origination of Sargassum in the Central Indo-Pacific (CIP) region with subsequent independent dispersal events into other marine realms. The longer history of diversification in the ancestral CIP range could explain the much greater diversity there relative to other marine areas today. Analyses of these dynamic processes, when fine-tuned to a higher spatial resolution, enable the identification of evolutionary hotspots and provide insights into long-term dispersal patterns.

Complete mitochondrial genome and phylogenetic analysis of brown alga Scytosiphon lomentaria (Ectocarpales, Phaeophyceae).

Project description:The present study provides a mitochondrial genome sequence of Scytosiphon lomentaria, which is better than the previously established data in China. This mitogenome is 39,971 bp in length and consists of 35 protein-coding genes, 3 ribosomal-RNA genes, 24 transfer-RNA genes, and 5 open reading frames. Overall, the base composition was as follows: A, 29.7%; C, 12.8%; G, 19.2%; and T, 38.4%. The phylogenetic analysis based on the protein-coding genes indicated that the S. lomentaria collected in the samples was at an earlier differentiation position. These results contribute to the further understanding of Phaeophyceae phylogenetic relationships and species identification.

Complete Plastid Genome of the Brown Alga Costaria costata (Laminariales, Phaeophyceae).

Project description:Costaria costata is a commercially and industrially important brown alga. In this study, we used next-generation sequencing to determine the complete plastid genome of C. costata. The genome consists of a 129,947 bp circular DNA molecule with an A+T content of 69.13% encoding a standard set of six ribosomal RNA genes, 27 transfer RNA genes, and 137 protein-coding genes with two conserved open reading frames (ORFs). The overall genome structure of C. costata is nearly the same as those of Saccharina japonica and Undaria pinnatifida. The plastid genomes of these three algal species retain a strong conservation of the GTG start codon while infrequently using TGA as a stop codon. In this regard, they differ substantially from the plastid genomes of Ectocarpus siliculosus and Fucus vesiculosus. Analysis of the nucleic acid substitution rates of the Laminariales plastid genes revealed that the petF gene has the highest substitution rate and the petN gene contains no substitution over its complete length. The variation in plastid genes between C. costata and S. japonica is lower than that between C. costata and U. pinnatifida as well as that between U. pinnatifida and S. japonica. Phylogenetic analyses demonstrated that C. costata and U. pinnatifida have a closer genetic relationship. We also identified two gene length mutations caused by the insertion or deletion of repeated sequences, which suggest a mechanism of gene length mutation that may be one of the key explanations for the genetic variation in plastid genomes.
