The complete chloroplast genome of Euphorbia ebracteolata Hayata (Euphorbiaceae).

ABSTRACT: Euphorbia ebracteolata is a perennial medicinal plant and widely used in China for thousands of years. The complete chloroplast genome reported here is 163,090 bp in length, including two inverted repeats (IRs) of 26,699 bp, which are separated by a large single-copy (LSC) and a small single-copy (SSC) of 91,943 and 17,749 bp, respectively. The whole chloroplast genome of E. ebracteolata contains 112 genes, including 78 protein-coding genes, 30 transfer RNA, and 4 ribosome RNA. Phylogenetic analysis result strongly indicated that E. ebracteolata is closely related to E. helioscopia.

Complete chloroplast genome of Euphorbia micractina Boiss (Euphorbiaceae: Euphorbia).

Project description:Euphorbia micractina Boiss is a plant with high medicinal value. Yet, its molecular biology is not fully understood. In this study, we sequenced the whole chloroplast genome (CP) sequence of E. micractina to study its phylogenetic relationship in Euphorbiaceae. The total length of the chloroplast genome of E. micractina is 162,056 bp, including a large single-copy (LSC) region of 89,936bp bp, a small single-copy (SSC) region of 18,376 bp, and a pair of identical inverted repeat regions (IRs) of 11,367 bp. The genome has 128 genes, including 84 protein-coding genes, 36 transfer RNA (tRNA) genes, and 8 ribosomal RNA (rRNA) genes. The overall GC content of the plastome is 35.7%. The phylogenetic analysis of E. micractina with 30 related species suggested a closest taxonomical relationship with Euphorbia pekinensis in the Euphorbiaceae family.

The complete chloroplast genome of Euphorbia prostrata (Euphorbiaceae).

Project description:Euphorbia prostrata Aiton 1789, an annual herb, is native to tropical and subtropical Americas and was introduced into many parts of the Old-World including Korea. We determined the genomic characteristics of the complete chloroplast genome of E. prostrata with a de novo assembly strategy. The complete chloroplast genome was 162,858 bp long, and harbored 86 protein coding, 37 tRNA and 8 rRNA. The genome showed the typical quadripartite structure consisting of a large single copy (LSC) (90,580 bp), a small single copy (SSC) (18,570 bp) and a pair of inverted repeats (IRs) (26,854 bp). The phylogenetic tree inferred from chloroplast genomes of 25 taxa, which belong to Euphorbiaceae indicated that the Euphorbia is a monophyletic group and E. prostrata is closely related with E. humifusa, E. thymifolia and E. hirta, forming a well-supported clade. Our findings can be valuable for understanding the phylogenetic relationships and the evolution in Euphorbiaceae and will provide basic information for future studies on E. prostrata from genomic perspective.

The complete chloroplast genome sequence of Euphorbia lathyris L. (Euphorbiaceae).

Project description:Euphorbia lathyris L. is a well-known bioenergy plant cultivated in many parts of the world. In this study, we sequenced the complete chloroplast (cp) genome sequence of E. lathyris to investigate its phylogenetic relationship in the family Euphorbiaceae. The cp genome was 163,738 bp in length, consisting of a pair of inverted repeats (IRa and IRb: 26,837 bp) separated by a large single-copy region (LSC: 91,783 bp) and a small single-copy region (SSC: 18,281 bp). The GC content of whole cp genome is 35.6%. Annotation showed the presence of 113 unique genes with 79 protein-coding genes, four tRNA genes, and 30 rRNA genes. Phylogenetic analysis indicated that E. lathyris was in the basal position of subgen. Esula, closely related to sect. Esula and sect. Helioscopiae.

Characterization of the complete chloroplast genome of Euphorbia pekinensis Rupr. (Euphorbiaceae).

Project description:Euphorbia pekinensis Rupr. 1859 is a medicinal herb endemic to China and distributed throughout the country, particularly across the northern part of the mainland. However, the systematic classification of Euphorbiaceae remains controversial. Therefore, studying the chloroplast genome of E. pekinensis is crucial for the resolution of this taxonomic dispute, clarification of the systematic status of Euphorbia, and establishment of an accurate classification system for Euphorbiaceae. In this study, we sequenced the complete chloroplast genome of E. pekinensis using Illumina sequencing technology and annotated it using GeSeq. The complete chloroplast genome was 162,002-bp-long with a guanine-cytosine (GC) content of 35.7%. It included one large single-copy (LSC), one small single-copy (SSC), and two inverted repeat sequence regions (IRa and IRb), which were 90,225 bp, 18,067 bp, and 26,855 bp in length, respectively, and are indicative of a typical tetrad structure. The genome encoded 129 functional genes, comprising 85 protein-coding genes, 36 tRNA genes, and eight rRNA genes. According to the maximum-likelihood phylogenetic tree that was constructed using 16 complete chloroplast genomes, E. pekinensis was found to be closely related to E. ebracteolata. Therefore, the complete chloroplast genome of E. pekinensis provides a better understanding of Euphorbia genetics.

Characterization of the complete chloroplast genome of Euphorbia helioscopia Linn. (Euphorbiaceae), a traditional Chinese medicine.

Project description:Euphorbia helioscopia Linn. known as a traditional Chinese medicine of Euphorbiaceae, which contains terpenes, steroids, flavonoids, acetophenones, tannins, phenylpropanoids, cerebrosides and so on. Euphorbia helioscopia L. was used to treat malignant tumors and chronic obstructive pulmonary diseases such as cough, phlegm-turbidity, asthma, and chronic bronchitis. The complete chloroplast genome was assembled by Illumina paired-end reads data. The length of circular cp genome distribution in 160,041 bp, containing a large single-copy region (LSC) of 88,832 bp, a small single-copy region (SSC) of 17,145 bp and a pair of inverted repeat (IR) regions of 27,032 bp. In addition, 11 genes possess a single intron, while the other two genes (ycf3, clpP) have a couple of introns. The GC content of entire Euphorbia helioscopia L. cp genome, LSC, SSC and IR regions are 35.9, 33.1, 30.3, and 42.3%, respectively. From the NJ phylogenetic tree analysis showed that Euphorbia helioscopia L. and Euphorbia esula are closely related to each other within the family Euphorbiaceae.

Project description:Euphorbia kansui T.N. Liou ex S.B. Ho (Euphorbiaceae) is a perennial herb plant endemic to China. This species has important economic and medicinal values. In this study, we first characterized the complete nucleotide sequence of chloroplast (cp) genome of E. kansui using the Illumina Hiseq platform. The cp genome was 161,061 bp in length, comprising of a large single copy (LSC) region of 91,288 bp, a small single copy (SSC) region of 17,086 bp, and two inverted repeat regions of 26,343 bp each. The cp genome contains 130 genes, including 86 protein-coding genes, 8 ribosomal RNAs (rRNAs), and 36 transfer RNAs (tRNAs). The phylogenetic analysis indicated that E. kansui was placed as a sister to the congeneric Euphorbia esula.

Inhibitory Effects of Euphorbia ebracteolata Hayata Extract ECB on Melanoma-Induced Hyperplasia of Blood Vessels in Zebrafish Embryos.

Project description:Melanoma is a serious malignant form of skin cancer. Euphorbiaceae compound B (ECB, 2,4-dihydroxy-6-methoxy-3-methylacetophenone) is an acetophenone compound that is isolated from Euphorbia ebracteolata Hayata (EEH), an herbaceous perennial, and has antitumor activity. Here, we transplanted human melanoma cells into zebrafish embryos to establish a zebrafish/melanoma model. We showed that this model can be used to evaluate the therapeutic effect of EEH and ECB and discussed its potential mechanism of action. The results showed that ECB was an active ingredient of EEH in inhibiting melanoma-induced hyperplasia of blood vessels in zebrafish embryos, similar to the angiogenic inhibitor vatalanib. ECB inhibited the number and length of subintestinal veins (p < 0.05), as well as the distribution of melanoma in zebrafish embryos (p < 0.05). More importantly, unlike vatalanib, ECB only inhibited melanoma-induced abnormal and excessive growth of blood vessels in xenografts. In addition, ECB inhibited the mRNA expression of vegfr2 and vegfr3 in zebrafish. Both vegfr2 and vegfr3 are essential genes that regulate blood vessel formation and upregulate the expression of p53 and casp3a genes in zebrafish. Together, the above-mentioned results indicate that ECB has a potential antimelanoma effect in vivo, which may be mediated by inhibiting vascular endothelial growth factor receptors.

Two novel diterpenoid heterodimers, Bisebracteolasins A and B, from Euphorbia ebracteolata Hayata, and the cancer chemotherapeutic potential of Bisebracteolasin A.

Project description:Rare ent-abietane-rosane diterpenoid heterodimers, Bisebracteolasins A and B (1 and 2, respectively), were isolated from the roots of Euphorbia ebracteolata Hayata. Their structures and absolute configurations were elucidated from spectroscopic data and X-ray diffraction analysis. Compounds 1 and 2 exhibited moderate cytotoxic effects against five cancer cell lines. Compound 1 showed more effective antiproliferative activities against human tumour cells, HL-60 and SMMC-7721, with IC50 values of 2.61 and 4.08 μM, respectively, than 2. Both compounds 1 and 2 inhibit the colorectal cancer stem cell line P6C with IC50 values of 16.48 and 34.76 μM, respectively. Moreover, preliminary biological tests showed compound 1 exhibited inhibitory activity towards tumoursphere formation and migration of the P6C cell line. Overall, we identified two novel diterpenoid heterodimers, and Bisebracteolasin A exhibits therapeutic potential in impeding tumour growth and metastatic ability of cancer stem cells.
