Complete mitochondrial genome of the acrobat ant Crematogaster teranishii Santschi, 1930 (Formicidae; Hymenoptera).

ABSTRACT: The genus Crematogaster is a diverse group of ants found around the world. We have completed the mitochondrial genome of Crematogaster teranishii, which is the first mitochondrial genome of the genus. The mitochondrial genome is 17,442 bp long and 20.3% in GC ratio, which is similar to those of other ants. It contains 13 protein-coding genes, two ribosomal RNAs, 22 transfer RNAs, and a control region with same gene order to other myrmicine species. The intergenic region between nad3 and trnA was unusually long compared to other ant species. Phylogenetic analysis showed that C. teranishii was closely related to other members of tribe Crematogastrini.

Project description:Crematogaster matsumurai (Forel 1901) is an important arboreal ant species commonly found on Phyllostachys heterocycla (Carr.) in Lishui, Zhejiang, China. This study analyzed the mitochondrial genome sequence of C. matsumurai and discussed its phylogenetic relationship in Hymenoptera. The circular mitochondrial genome was 16,028 bp long, including a standard set of 22 transfer RNAs (tRNAs), two ribosomal RNAs (rRNAs), and 13 protein-coding genes (PCGs), which showed the typical insect mitochondrial genome arrangement. The AT and GC contents of the mitochondrial genome sequence were 76.92% and 23.08%, respectively. The maximum-likelihood (ML) phylogenetic analysis based on whole mitochondrial genome sequences showed that C. matsumurai is closest to Crematogaster teranishii.

The complete mitochondrial genome of a parthenogenetic ant Monomorium triviale (Hymenoptera: Formicidae).

Project description:Monomorium is one of the most species-rich yet taxonomically problematic ant genus. An East Asian species, M. triviale Wheeler, W.M., 1906, is reproduced by obligate thelytokous parthenogenesis and performs strict reproductive division of labor. We sequenced the M. triviale mitogenome using next-generation sequencing methods. The circular mitogenome of M. triviale was 16,290 bp in length, consisting of 13 protein-coding genes, two ribosomal RNA genes, 22 transfer RNAs, and a single non-coding region of 568 bp. The base composition was AT-biased (82%). Gene order rearrangements were detected and likely to be unique to the genus Monomorium. We announce the M. triviale mitogenome as additional genomic resources for elucidating phylogenetic and taxonomic problems of Monomorium and comparative genomics of parthenogenetic ant species.

The complete mitochondrial genome of yellow crazy ant, Anoplolepis gracilipes (Hymenoptera: Formicidae).

Project description:The yellow crazy ant Anoplolepis gracilipes is an invasive species that threatens biodiversity in introduced ecosystems. We sequenced the A. gracilipes mitogenome using next-generation sequencing methods. The circular mitogenome of A. gracilipes was 16,943 bp included 13 protein-coding genes, two ribosomal RNA genes, 22 transfer RNAs, and a single large non-coding region of 893 bp. The base composition was AT-biased (72%). Three genomic rearrangements compared to ancestral insects were found. Phylogenetic analysis based on the concatenated nucleotide sequences of the 13 protein-coding genes supports A. gracilipes belonging to the Formicinae subfamily. We announce the A. gracilipes mitogenome as a DNA reference for further population genetic, phylogenetic, and evolutionary analyses.

Trans-generational immunization in the acrobat ant Crematogaster scutellaris.

Project description:Trans-generational immunization is defined as the transmission of an enhanced resistance to a pathogen from parents to offspring. By using the host-parasite system of the ant Crematogaster scutellaris and the entomopathogenic fungus Metarhizium anisopliae, we describe this phenomenon for the first time in ants. We exposed four groups of hibernating queens to different treatments (i) a non-lethal dose of live conidiospores in Triton, (ii) a dose of heat-killed conidiospores in Triton, (iii) a control Triton solution, and (iv) a naive control. We exposed their first workers to a high dose of conidiospores and measured mortality rates. Workers produced by queens exposed to live conidiospores survived longer than those belonging to the other groups, while exposure to Triton and dead spores had no effect. Starved workers showed a significantly higher mortality. The treatments did not influence queen mortality, nor the number of offspring they produced at the emergence of the first worker, showing no evidence of immunization costs-at least for these parameters in the first year of colony development. We propose that trans-generational immunization represents an important component of social immunity that could affect colony success, particularly during the critical phase of claustral foundation.

Project description:The genus Lasius is a conspicuous and popular genus of ants found in the Holarctic regions. We have completed the mitochondrial genome of Lasius spathepus as the first mitochondrial genome of Lasius. The mitochondrial genome is 18,951 bp long, which is the sixth longest ant mitochondrial genome known to science. It contains 13 protein-coding genes, 2 ribosomal RNAs, 22 transfer RNAs, and a control region in a gene order shared with other species of subfamily Formicinae. The control region is 2,147 bp long, longest of all ants. Phylogenetic analysis shows L. spathepus groups with Nylanderia flavipes of the same tribe Lasiini.

Complete mitochondrial genome of the H3 haplotype Argentine ant Linepithema humile (Mayr, 1868) (Formicidae; Hymenoptera).

Project description:The argentine ant, Linepithema humile (Mayr, 1867), is an invasive ant species that has spread across the world. We have determined the mitochondrial genome of L. humile collected in South Korea, which is 15,934 bp containing 10 SNPs and 5 INDELs compared to the previous mitogenome. Most SNPs were found in cox3, followed by cytb. From SNPs our mitogenome was identified as a H3 haplotype, which was previously recorded in Japan and the U.S. while the previous mitogenome was H1 haplotype. Phylogenetic analysis was congruent to previous study within the tribe Leptomyrmecini but not between other tribes of subfamily Dolichoderinae.

Project description:Colobopsis ants are unique ants known for their phragmotic behavior. We have completed the mitochondrial genome of Colobopsis nipponica (Wheeler, W.M., 1928) as the first mitochondrial genome of the genus. The mitogenome is 17,431 bp long and 19.4% in GC ratio, which is the third longest mitochondrial genome in subfamily Formicinae. It contains 13 protein-coding genes, 2 rRNA genes, 22 tRNA genes and a large 1534 bp long control region. Both gene order and phylogenetic analysis agree with the recent elevation of Colobopsis from subgenus to genus.

Congruence of microsatellite and mitochondrial DNA variation in acrobat ants (Crematogaster subgenus Decacrema, Formicidae: Myrmicinae) inhabiting Macaranga (Euphorbiaceae) myrmecophytes.

Project description:A previously reported mitochondrial DNA (mtDNA) phylogeny of Crematogaster (subgenus Decacrema) ants inhabiting Macaranga myrmecophytes indicated that the partners diversified synchronously and their specific association has been maintained for 20 million years. However, the mtDNA clades did not exactly match morphological species, probably owing to introgressive hybridization among younger species. In this study, we determined the congruence between nuclear simple sequence repeat (SSR, also called microsatellite) genotyping and mtDNA phylogeny to confirm the suitability of the mtDNA phylogeny for inferring the evolutionary history of Decacrema ants. Analyses of ant samples from Lambir Hills National park, northeastern Borneo, showed overall congruence between the SSR and mtDNA groupings, indicating that mtDNA markers are useful for delimiting species, at least at the local level. We also found overall high host-plant specificity of the SSR genotypes of Decacrema ants, consistent with the specificity based on the mtDNA phylogeny. Further, we detected cryptic genetic assemblages exhibiting high specificity toward particular plant species within a single mtDNA clade. This finding, which may be evidence for rapid ecological and genetic differentiation following a host shift, is a new insight into the previously suggested long-term codiversification of Decacrema ants and Macaranga plants.

Complete mitochondrial genome sequence of Tapinoma melanocephalum (Hymenoptera: Formicidae).

Project description:Tapinoma melanocephalum is a ubiquitous invasive species and widely distributed in subtropical and tropical regions around the world. Here, we sequenced and annotated the complete mitochondrial genome (mitogenome) of T. melanocephalum. This mitogenome was 15,499 bp long and encoded 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and 2 ribosomal RNA unit genes (rRNAs). Compared to other Formicidae species, gene order of T. melanocephalum was not conserved and one tRNA cluster trnW-trnC-trnY converted to trnW-trnY-trnC. The whole mitogenome exhibited heavy AT nucleotide bias (79.5%). All PCGs started with the standard ATN codons. Except for cox1 and nad5 end with the incomplete codon T-, all PCGs terminated with the stop codon TAA. Phylogenetic analysis showed that T. melanocephalum got together with three same subfamily Dolichoderinae species and one Dorylinae species, indicating the close relationship of Dolichoderinae and Dorylinae.

This sequence data would play an important role in the investigation of mitochondrial evolution of the subfamily Myrmicinae.
