The complete mitochondrial genome of a parthenogenetic ant Monomorium triviale (Hymenoptera: Formicidae).

ABSTRACT: Monomorium is one of the most species-rich yet taxonomically problematic ant genus. An East Asian species, M. triviale Wheeler, W.M., 1906, is reproduced by obligate thelytokous parthenogenesis and performs strict reproductive division of labor. We sequenced the M. triviale mitogenome using next-generation sequencing methods. The circular mitogenome of M. triviale was 16,290 bp in length, consisting of 13 protein-coding genes, two ribosomal RNA genes, 22 transfer RNAs, and a single non-coding region of 568 bp. The base composition was AT-biased (82%). Gene order rearrangements were detected and likely to be unique to the genus Monomorium. We announce the M. triviale mitogenome as additional genomic resources for elucidating phylogenetic and taxonomic problems of Monomorium and comparative genomics of parthenogenetic ant species.

Project description:The yellow crazy ant Anoplolepis gracilipes is an invasive species that threatens biodiversity in introduced ecosystems. We sequenced the A. gracilipes mitogenome using next-generation sequencing methods. The circular mitogenome of A. gracilipes was 16,943 bp included 13 protein-coding genes, two ribosomal RNA genes, 22 transfer RNAs, and a single large non-coding region of 893 bp. The base composition was AT-biased (72%). Three genomic rearrangements compared to ancestral insects were found. Phylogenetic analysis based on the concatenated nucleotide sequences of the 13 protein-coding genes supports A. gracilipes belonging to the Formicinae subfamily. We announce the A. gracilipes mitogenome as a DNA reference for further population genetic, phylogenetic, and evolutionary analyses.

Complete mitochondrial genome of the acrobat ant Crematogaster teranishii Santschi, 1930 (Formicidae; Hymenoptera).

Project description:The genus Crematogaster is a diverse group of ants found around the world. We have completed the mitochondrial genome of Crematogaster teranishii, which is the first mitochondrial genome of the genus. The mitochondrial genome is 17,442 bp long and 20.3% in GC ratio, which is similar to those of other ants. It contains 13 protein-coding genes, two ribosomal RNAs, 22 transfer RNAs, and a control region with same gene order to other myrmicine species. The intergenic region between nad3 and trnA was unusually long compared to other ant species. Phylogenetic analysis showed that C. teranishii was closely related to other members of tribe Crematogastrini.

Project description:The genus Lasius is a conspicuous and popular genus of ants found in the Holarctic regions. We have completed the mitochondrial genome of Lasius spathepus as the first mitochondrial genome of Lasius. The mitochondrial genome is 18,951 bp long, which is the sixth longest ant mitochondrial genome known to science. It contains 13 protein-coding genes, 2 ribosomal RNAs, 22 transfer RNAs, and a control region in a gene order shared with other species of subfamily Formicinae. The control region is 2,147 bp long, longest of all ants. Phylogenetic analysis shows L. spathepus groups with Nylanderia flavipes of the same tribe Lasiini.

Complete mitochondrial genome of the H3 haplotype Argentine ant Linepithema humile (Mayr, 1868) (Formicidae; Hymenoptera).

Project description:The argentine ant, Linepithema humile (Mayr, 1867), is an invasive ant species that has spread across the world. We have determined the mitochondrial genome of L. humile collected in South Korea, which is 15,934 bp containing 10 SNPs and 5 INDELs compared to the previous mitogenome. Most SNPs were found in cox3, followed by cytb. From SNPs our mitogenome was identified as a H3 haplotype, which was previously recorded in Japan and the U.S. while the previous mitogenome was H1 haplotype. Phylogenetic analysis was congruent to previous study within the tribe Leptomyrmecini but not between other tribes of subfamily Dolichoderinae.

Project description:Colobopsis ants are unique ants known for their phragmotic behavior. We have completed the mitochondrial genome of Colobopsis nipponica (Wheeler, W.M., 1928) as the first mitochondrial genome of the genus. The mitogenome is 17,431 bp long and 19.4% in GC ratio, which is the third longest mitochondrial genome in subfamily Formicinae. It contains 13 protein-coding genes, 2 rRNA genes, 22 tRNA genes and a large 1534 bp long control region. Both gene order and phylogenetic analysis agree with the recent elevation of Colobopsis from subgenus to genus.

Monomorium dryhimi sp. n., a new ant species (Hymenoptera, Formicidae) of the M. monomorium group from Saudi Arabia, with a key to the Arabian Monomorium monomorium-group.

Project description:A new ant species, Monomorium dryhimi, is described based on workers from a single colony collected in Al Bahah, Asir Province, Saudi Arabia. This is the fourth species of the Monomorium monomorium-group collected from Arabian Peninsula, and appears to be closely related to Monomorium holothir Bolton, 1987, from Kenya. It can be distinguished by the following characters: head in profile with a weakly convex dorsal surface and a clearly convex ventral surface; eyes of moderate size with maximum diameter EL 0.19-0.25 × HW and with 6 ommatidia in the longest row; body colour yellow to light brownish yellow. In some individuals, head and gaster slightly but conspicuously darker than rest of body. Second halves of first and second gastral tergites with two characteristic brownish transverse bands. An identification key to the workers of the Arabian species of the Monomorium monomorium-group is presented. Scanning electron micrographs are given to illustrate the new species.

Complete mitochondrial genome sequence of Tapinoma melanocephalum (Hymenoptera: Formicidae).

Project description:Tapinoma melanocephalum is a ubiquitous invasive species and widely distributed in subtropical and tropical regions around the world. Here, we sequenced and annotated the complete mitochondrial genome (mitogenome) of T. melanocephalum. This mitogenome was 15,499 bp long and encoded 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and 2 ribosomal RNA unit genes (rRNAs). Compared to other Formicidae species, gene order of T. melanocephalum was not conserved and one tRNA cluster trnW-trnC-trnY converted to trnW-trnY-trnC. The whole mitogenome exhibited heavy AT nucleotide bias (79.5%). All PCGs started with the standard ATN codons. Except for cox1 and nad5 end with the incomplete codon T-, all PCGs terminated with the stop codon TAA. Phylogenetic analysis showed that T. melanocephalum got together with three same subfamily Dolichoderinae species and one Dorylinae species, indicating the close relationship of Dolichoderinae and Dorylinae.

This sequence data would play an important role in the investigation of mitochondrial evolution of the subfamily Myrmicinae.

Validation of Potential Reference Genes for Real-Time qPCR Analysis in Pharaoh Ant, Monomorium pharaonis (Hymenoptera: Formicidae).

Project description:Ants are highly diverse social insects living in colonies consisted of up to millions of individuals with reproductive division of labors. Due to the interests in disclosing the genetic and epigenetic regulation mechanisms underlying the distinct developmental trajectories between castes and division of labor in colonies, many ant species have recently been established as laboratory models for evolutionary development and social behavior studies. These functional studies often request a precise quantification of the relative gene expression level, which relies on a stably expressed reference genes for normalization. A core set of reliable reference genes for this purpose however has not been established yet in ants. In the present study, we tested the expression patterns and amplification efficiencies of 12 abundantly expressed candidate genes in Monomorium pharaonis, one of the few ant species that are suitable for laboratory rearing and experimentation. We quantified the expression levels of these genes by RT-qPCR in seven different conditions: embryo development, sexual development, worker development, adult phenotypes, tissues, and two abiotic manipulative treatments in pharaoh ant. Finally, five genes, elongation factor-1 alpha (EF1A), glyceraldehyde 3-phosphate dehydrogenase (GAPDH), TATA-box-binding protein (TATA), tubulin gamma-2 chain-like (TBLg2), heat shock protein 67B2-like (HSP67) were found to be the most stable reference genes across seven conditions. We also identified the most stable reference genes applicable for each distinct condition and the optimal number of reference genes entailed were evaluated. Our study validates reliable reference genes for RT-qPCR analysis which lays the foundation for future studies in pharaoh ant.

Linear Morphometry of Male Genitalia Distinguishes the Ant Genera Monomorium and Syllophopsis (Hymenoptera: Formicidae) in Madagascar.

Project description:Morphometric analyses of male genitalia are routinely used to distinguish genera and species in beetles, butterflies, and flies, but are rarely used in ants, where most morphometric analyses focus on the external morphology of the worker caste. In this work, we performed linear morphometric analysis of the male genitalia to distinguish Monomorium and Syllophopsis in Madagascar. For 80 specimens, we measured 10 morphometric characters, especially on the paramere, volsella, and penisvalvae. Three datasets were made from linear measurements: mean (raw data), the ratios of characters (ratio data), and the Removal of Allometric Variance (RAV data). The following quantitative methods were applied to these datasets: hierarchical clustering (Ward's method), unconstrained ordination methods including Principal Component Analysis (PCA), Non-Metric Multidimensional Scaling analyses (NMDS), Linear Discriminant Analysis (LDA), and Conditional Inference Trees (CITs). The results from statistical analysis show that the ratios proved to be the most effective approach for genus-level differentiation. However, the RAV method exhibited overlap between the genera. Meanwhile, the raw data facilitated more nuanced distinctions at the species level compared with the ratios and RAV approaches. The CITs revealed that the ratios of denticle length of the valviceps (SeL) to the paramere height (PaH) effectively distinguished between genera and identified key variables for species-level differentiation. Overall, this study shows that linear morphometric analysis of male genitalia is a useful data source for taxonomic delimitation.
