Cycloartane- and Lanostane-Type Triterpenoids from the Resin of Parthenium argentatum AZ-2, a Byproduct of Guayule Rubber Production.

ABSTRACT: A total of 12 new cycloartane- and lanostane-type triterpenoids including 16-deoxyargentatin A (1), 16-deoxyisoargentatin A (2), 7-oxoisoargentatin A (3), 24-epi-argentatin H (4), 24-O-p-anisoylargentatin C (5), 24-O-trans-cinnamoylargentatin C (6), 16-dehydroargentatin C (7), 16,17(20)-didehydroargentatin C (8), isoargentatin C (9), isoargentatin H (10), 3-epi-quisquagenin (11), and isoquisquagenin (12) together with 10 known triterpenoids (13-22) were isolated from the resin of Parthenium argentatum AZ-2 obtained as a byproduct of Bridgestone guayule rubber production. The structures of new triterpenoids 1-12 and argentatin H (13), which has previously been characterized as its diacetate (23), were elucidated by extensive analysis of their spectroscopic data and chemical conversions, and the known compounds 14-22 were identified by comparison of their spectroscopic data with those reported. Of these, 13, 14, and 18 exhibited weak cytotoxic activity for several cancer cell lines.

Transcriptome analysis of rubber biosynthesis in guayule (Parthenium argentatum gray).

Project description:BACKGROUND:Natural rubber is currently produced nearly exclusively from latex of the Para rubber tree, Hevea brasiliensis. The desire to reduce the environmental cost of rubber production, fears of pathogen susceptibility in clonal Hevea plantations, volatility in the price of natural rubber, and increasing labor costs have motivated efforts to diversify the supply of natural rubber by developing alternative rubber crops such as guayule (Parthenium argentatum Gray). In Hevea, latex is produced as an exudate following wounding while in guayule, rubber is deposited within the cortical parenchyma and its production is strongly influenced by environmental conditions. RESULTS:To better understand the enzymology and regulation of guayule rubber biosynthesis and to identify genes with potential uses in the improvement of rubber yields, we conducted de novo transcriptome assembly and differential gene expression analyses of this process in guayule. This analysis supports a role for rubber in the defense against pathogens, identified new enzymes potentially involved in the biosynthesis of rubber as well as transcription factors specifically expressed in rubber-producing tissues. CONCLUSIONS:Data presented here will be useful in the improvement of guayule as an alternative source of natural rubber and in better understanding the biosynthesis of this critical polymer. In particular, some of the candidate transcription factors are likely to control the rubber biosynthesis pathway and are good targets for molecular breeding or engineering of guayule plants with higher and more consistent production of rubber.

Molecular Studies of the Protein Complexes Involving Cis-Prenyltransferase in Guayule (Parthenium argentatum), an Alternative Rubber-Producing Plant.

Project description:Guayule (Parthenium argentatum) is a perennial shrub in the Asteraceae family and synthesizes a high quality, hypoallergenic cis-1,4-polyisoprene (or natural rubber; NR). Despite its potential to be an alternative NR supplier, the enzymes for cis-polyisoprene biosynthesis have not been comprehensively studied in guayule. Recently, implications of the protein complex involving cis-prenyltransferases (CPTs) and CPT-Binding Proteins (CBPs) in NR biosynthesis were shown in lettuce and dandelion, but such protein complexes have yet to be examined in guayule. Here, we identified four guayule genes - three PaCPTs (PaCPT1-3) and one PaCBP, whose protein products organize PaCPT/PaCBP complexes. Co-expression of both PaCBP and each of the PaCPTs could complemented the dolichol (a short cis-polyisoprene)-deficient yeast, whereas the individual expressions could not. Microsomes from the PaCPT/PaCBP-expressing yeast efficiently incorporated 14C-isopentenyl diphosphate into dehydrodolichyl diphosphates; however, NR with high molecular weight could not be synthesized in in vitro assays. Furthermore, co-immunoprecipitation and split-ubiquitin yeast 2-hybrid assays using PaCPTs and PaCBP confirmed the formation of protein complexes. Of the three PaCPTs, guayule transcriptomics analysis indicated that the PaCPT3 is predominantly expressed in stem and induced by cold-stress, suggesting its involvement in NR biosynthesis. The comprehensive analyses of these PaCPTs and PaCBP here provide the foundational knowledge to generate a high NR-yielding guayule.

Lanostane- and cycloartane-type triterpenoids from Abies balsamea oleoresin.

Project description:Phytochemical analysis of A. balsamea oleoresin led to the isolation of three new 3,4-seco-lanostane triterpenoids 1-3, one new cycloartane triterpenoid 4 along with fourteen known terpenoids. Structure determinations were based on extensive 1D/2D NMR, IR and MS spectroscopic analyses, and comparison with literature data. The isolated compounds were evaluated in vitro for their cytotoxicity against human cell lines (A549, DLD-1, WS1) and their antibacterial activity against E. coli and S. aureus. Abiesonic acid (6) exhibited weak cytotoxic activity against A549 (IC50 = 22 µM) while compounds 1 and 4 were weakly active against S. aureus (MIC = 25 µM).

A high-throughput quantification of resin and rubber contents in Parthenium argentatum using near-infrared (NIR) spectroscopy.

Project description:Background:Guayule (Parthenium argentatum A. Gray), a plant native to semi-arid regions of northern Mexico and southern Texas in the United States, is an alternative source for natural rubber (NR). Rapid screening tools are needed to replace the current labor-intensive and cost-inefficient method for quantifying rubber and resin contents. Near-infrared (NIR) spectroscopy is a promising technique that simplifies and speeds up the quantification procedure without losing precision. In this study, two spectral instruments were used to rapidly quantify resin and rubber contents in 315 ground samples harvested from a guayule germplasm collection grown under different irrigation conditions at Maricopa, AZ. The effects of eight different pretreatment approaches on improving prediction models using partial least squares regression (PLSR) were investigated and compared. Important characteristic wavelengths that contribute to prominent absorbance peaks were identified. Results:Using two different NIR devices, ASD FieldSpec®3 performed better than Polychromix Phazir™ in improving R2 and residual predicative deviation (RPD) values of PLSR models. Compared to the models based on full-range spectra (750-2500 nm), using a subset of wavelengths (1100-2400 nm) with high sensitivity to guayule rubber and resin contents could lead to better prediction accuracy. The prediction power of the models for quantifying resin content was better than rubber content. Conclusions:In summary, the calibrated PLSR models for resin and rubber contents were successfully developed for a diverse guayule germplasm collection and were applied to roughly screen samples in a low-cost and efficient way. This improved efficiency could enable breeders to rapidly screen large guayule populations to identify cultivars that are high in rubber and resin contents.

Downregulation of a CYP74 Rubber Particle Protein Increases Natural Rubber Production in Parthenium argentatum.

Project description:We report functional genomics studies of a CYP74 rubber particle protein from Parthenium argentatum, commonly called guayule. Previously identified as an allene oxide synthase (AOS), this CYP74 constitutes the most abundant protein found in guayule rubber particles. Transgenic guayule lines with AOS gene expression down-regulated by RNAi (AOSi) exhibited strong phenotypes that included agricultural traits conducive to enhancing rubber yield. AOSi lines had higher leaf and stem biomass, thicker stembark tissues, increased stem branching and improved net photosynthetic rate. Importantly, the rubber content was significantly increased in AOSi lines compared to the wild-type (WT), vector control and AOS overexpressing (AOSoe) lines, when grown in controlled environments both in tissue-culture media and in greenhouse/growth chambers. Rubber particles from AOSi plants consistently had less AOS particle-associated protein, and lower activity (for conversion of 13-HPOT to allene oxide). Yet plants with downregulated AOS showed higher rubber transferase enzyme activity. The increase in biomass in AOSi lines was associated with not only increases in the rate of photosynthesis and non-photochemical quenching (NPQ), in the cold, but also in the content of the phytohormone SA, along with a decrease in JA, GAs, and ABA. The increase in biosynthetic activity and rubber content could further result from the negative regulation of AOS expression by high levels of salicylic acid in AOSi lines and when introduced exogenously. It is apparent that AOS in guayule plays a pivotal role in rubber production and plant growth.

Xuetonglactones A-F: Highly Oxidized Lanostane and Cycloartane Triterpenoids From Kadsura heteroclita Roxb. Craib.

Project description:Xuetonglactones A-F (1-6), six unreported highly oxidized lanostane- and cycloartane-type triterpenoids along with 22 known scaffolds (7-28) were isolated from the stems of Kadsura heteroclita (Roxb.) Craib. Compared with previous congeners, xuetonglactone A (1), possesses an unprecedented 20,21-α-epoxide, and xuetonglactone D (4) features an unusual 19-α-hydroperoxyl moiety. The structures and the absolute configurations of the compounds were established by extensive one- and two-dimensional NMR, and electronic circular dichroism (ECD) spectroscopic analysis, with those of 1 and 5 confirmed by single-crystal X-ray diffraction technique. Compounds 1 and 2 exhibited inhibition of iNOS activity in LPS-induced macrophages with IC50 values of 22.0, and 17.0 μg/mL, respectively. While compounds 6, 7, 8, and 24 showed potent cytotoxic activities against human cervical cancer cell lines (HeLa) with the IC50 values of 4.0, 5.8, 5.0, and 6.4 μM, and against human gastric cancer cells (BGC 823) with the IC50 values of 2.0, 5.0, 2.5, and 2.0 μM, respectively. Moreover, plausible biogenetic pathways of (1-6) were also proposed.
