The complete mitochondrial genome of Aeolothrips indicus Bhatti, 1964 (Thysanoptera: Thripidae).

ABSTRACT: Here, we have generated the complete mitochondrial sequence of Aeolothrips indicus Bhatti, 1964. So far, this is the first largest mitogenome with 17,042 bp length in order Thysanoptera. It includes 13 protein-coding genes, 22 transfer RNA genes, and two ribosomal RNA genes along with three non-coding regions. AT composition of A. indicus is 72.5% (37.7% A and 34.8% T) and GC 27.5% (15.6% C and 11.9% G). The constructed phylogeny revealed the monophyly of family Aeolothripidae in the order Thysanoptera. The data would provide further insight into the evolution and phylogeny of the order Thysanoptera.

Project description:Thrips hawaiiensis (Morgan) (Thysanoptera: Thripidae) is a common Thysanoptera insect widely distributed in Asia and the Pacific, it damages various plants. In this study the complete mitochondrial genome of T. hawaiiensis was sequenced and characterized by using next-generation sequencing technique. The total length of the complete genome is 15,357 bp and A + T content of 77.8% (GeneBank accession No. MW582621). The T. hawaiiensis mitochondrial genome consists of 13 protein-coding genes (PCGs), 2 ribosomal RNA genes, 22 transfer RNA genes (tRNAs) and 2 non-coding control regions (Dloop region). According to previous studies, only a few complete mitochondrial genomes from Order Thysanoptera have been reported. Thus, T. hawaiiensis complete mitochondrial genome sequence reported will provide molecular information for mitochondrial genome research on Thysanoptera.

The complete mitochondrial genome of Odontothrips loti (Haliday, 1852) (Thysanoptera: Thripidae).

Project description:Odontothrips loti (Haliday, 1852) is an important insect pest of Medicago sativa. In this study, the complete mitochondrial genome of O. loti (GenBank Accession number: MN584901) was sequenced using Illumina HiSeq X Ten. The genome is 15,584 bp long and comprises 13 protein-coding genes (PCGs), two ribosomal RNA genes (rRNAs), 22 transfer RNA genes (tRNAs), and putative control region (CR). The nucleotide composition of O. loti mitochondrial genome is 41.18% of A, 34.45% of T, 12.72% of C, and 11.64% of G. The CR, located between tRNA-Thr and ND5, is 651 bp long. The length of 22 tRNAs range from 56 bp to 69 bp. The two rRNAs were located on the H-strand of which one was 1127 bp (16S rRNA) long and another was 732 bp (12S rRNA) long. Phylogenetic analysis showed that O. loti is closely related to Frankliniella intonsa and Frankliniella occidentalis. These new mitochondrial genome data are more complete and can be better used to provide a basis for studies of the mitochondrial evolution of Odontothrips.

The complete mitochondrial genome of Taeniothrips tigris Bhatti, 1995 (Thysanoptera: Thripidae).

Project description:In this study, we sequenced complete mitogenome of Taeniothrips tigris Bhatti 1995. It was 15,501 bp in length containing 13 protein-coding genes, 22 transfer RNA genes, and 2 ribosomal RNA genes along with two non-coding regions. The overall base composition of Ta. tigris is 43.66% A, 35.20% T, 11.46% C, and 9.68% G, with a high AT bias of 78.86%. The constructed phylogeny using 19 mitogenomes revealed that the genus Taeniothrips is in close relationship with genus Thrips. This mitogenome data would help in deducing phylogenetic relationships studies in the order Thysanoptera.

The complete mitochondrial genome of the stick tea thrips Dendrothrips minowai (Thysanoptera: Thripidae).

Project description:The stick tea thrips, Dendrothrips minowai Priesner (Thysanoptera: Thripidae), is a major pest of tea plantation and poses a considerable economic threat to tea industry. The mitochondrial genome of D. minowai have been sequenced and annotated completely. The entire genome is 14,631 bp in length with an A + T content of 78.53% (GenBank accession No. MF582634). The stick tea thrips mt genome encodes all 37 genes that are typically found in animal mt genomes, consists of 13 protein-coding genes, two ribosomal RNA genes, and 22 transfer RNA genes. The gene order is unique and different from that of the other thrips. The A + T-rich region is 149 bp long and contains two poly-T stretchs. Phylogenetic analysis was performed using 13 protein-coding genes with six thrips showed that D. minowai and other five Thripidae species were clustered into a branch, which is formed a sister clade to H. aculeatus (family Phlaeothripidae).

Project description:Megalurothrips usitatus is a serious pest on Vigna unguiculata. In this study, the complete mitochondrial genome sequence of M. usitatus was characterized and its phylogenetic relationship within the Order Thysanoptera was determined. The mitochondrial genome of M. usitatus was a circular molecule of 15426 bp in length, containing 13 protein-coding genes, 2 rRNA genes, 22 tRNA genes, and the control region. It showed the typical insect mitochondrial genome arrangement. The AT content of the whole genome was 77.69% and the length of the control region was 567 bp with 78.66% AT content. The Maximum likelihood (ML) phylogenetic analysis based on mitochondrial protein-coding genes of 17 insect speciesshowed that M. usitatus is closest to Frankliniella occidentalis.

Characterizing the complete mitogenome of Odontothrips phaseoli (Thysanoptera: Thripidae) and its mitochondrial phylogeny.

Project description:Described originally from Heilongjiang, China, Odontothrips phaseoli is a potential pest of threatening bean plant in northern China. The complete mitochondrial genome of O. phaseoli was sequenced and assembled, with a total length of 15,540 bp. Within this genome, 37 genes have been identified: 13 PCGs, 22 tRNAs, two rRNAs, and two putative control regions. Most PCGs terminate with TAA, while four genes (atp8, nad1, nad2 and nad4) use an incomplete 'T' and nad6 employs TAG as the stop codon. Compared to the mitogenome of the ancestral insect, O. phaseoli displays significant gene rearrangement. However, it retains three conserved gene blocks in common with its related species, Megalurothrips usitatus, both of which belong to the Megalurothrips genus-group. The phylogenetic tree, constructed based on the entire mitogenome dataset of all thrips species available in NCBI, shows that the two species cluster closely together. This alignment might underscore the close link between gene arrangements and the phylogeny relationships.

The complete mitochondrial genome of Megalurothrips usitatus has been sequenced and annotated in this study, which is 17,209 bp long and contains 13 protein-coding genes (PCGs), two rRNAs, and 22 tRNA genes. Most of the protein-coding genes (PCGs) start with ATG except ND4 using TTG. Meanwhile, eight PCGs stop with TAA, four PCGs have an incomplete stop codon, and the gene Cytb ends with TAG. Phylogenetic analysis showed that M. usitatus is closely related to Frankliniella intonsa and F. occidentalis, providing a basis for the study of the mitochondrial evolution of Thripinae.

Complete mitochondrial genome of Gynaikothrips ficorum (Thysanoptera: Phlaeothripidae).

Project description:Gynaikothrips ficorum (Marchal 1908) is a major pest of bonsai ficus and poses a considerable economic threat to gardening industry. The mitochondrial genome of G. ficorum was sequenced and annotated in this study. Its whole mitogenome was 15,313 bp in length, including 37 typical genes in animal mitogenomes. ATN was used as start codon in most of the PCGs except for nad4l, which used TTG. All PCGs used TAA as termination codon except atp8 and atp6 which were ended with an incomplete T and TAG, respectively. A phylogenetic tree based on complete mitochondrial genomes of 17 species (15 Thysanoptera species and two outgroups) showed that the monophyly of Phlaeothripidae was supported and G. ficorum and G. uzeli formed a sister group.

Complete mitochondrial genome of Macromia manchurica Asahina, 1964 (Odonata: Macromiidae).

Project description:We describe the first time sequencing and assembly of the complete mitochondrial genome of Macromia manchurica Asahina, 1964 (Odonata; Macromiidae; Macromia). The mitochondrial genome of M. manchurica was found to be 15,560 bp. It contains thirteen protein-coding genes (PCGs), 22 transfer RNAs (tRNAs), two ribosomal RNAs (rRNAs), and AT-rich region. The overall base composition of A. japonicus is A-38.6%, C-17.0%, G-12.5%, and T-31.9%. A phylogenetic analysis of 14 species within the order Odonata and order Ephemeroptera suggested that Macromia amphigena is most closely related to M. manchurica.

Chromosome-level genome assembly of bean flower thrips Megalurothrips usitatus (Thysanoptera: Thripidae).

Project description:Bean flower thrips Megalurothrips usitatus is a staple pest of cowpea and other legumes and causes dramatic economic losses. Its small size allows for easy concealment, and large reproductive capacity easily leads to infestations. Despite the importance of a genome in developing novel management strategies, genetic studies on M. usitatus remain limited. Thus, we generated a chromosome-level M. usitatus genome using a combination of PacBio long read and Hi-C technologies. The assembled genome was 238.14 Mb with a scaffold N50 of 13.85 Mb. The final genome was anchored into 16 pseudo-chromosomes containing 14,000 genes, of which 91.74% were functionally annotated. Comparative genomic analyses revealed that expanded gene families were enriched in fatty acid metabolism and detoxification metabolism (ABC transporters), and contracted gene families were strongly associated with chitin-based cuticle development and sensory perception of taste. In conclusion, this high-quality genome provides an invaluable resource for us to understand the thrips' ecology and genetics, contributing to pest management.
