Analysis of the Structural Aspects of Tannin-Based Adhesives by 2D-NMR.

ABSTRACT: We developed non-toxic, harmless adhesives composed of all-natural and renewable resources, of which one was composed of tannin and gelatin, which unfortunately was lacking water resistance, and the other of tannin and ε-poly-l-lysine. In this study, we analyzed the chemical structures of these adhesives by two-dimensional nuclear magnetic resonance (2D-NMR) to explain the difference in water-resistance of the two glues. The results showed that only one proton was left in the benzene ring of tannin after mixing. This suggests that the amino group of the protein was directly attached to the benzene ring by a Michael addition-type reaction, and not to the hydroxyl group. In addition, the heteronuclear multiple bond correlation spectrum of the tannin-poly-l-lysine compound indicated that the hydroxyl groups of the tannin oxidized, suggesting the improvement of its water resistance.

Structural Analysis of Non-native Peptide-Based Catalysts Using 2D NMR-Guided MD Simulations.

Project description:Proteins and enzymes generally achieve their functions by creating well-defined 3D architectures that pre-organize reactive functionalities. Mimicking this approach to supramolecular pre-organization is leading to the development of highly versatile artificial chemical environments, including new biomaterials, medicines, artificial enzymes, and enzyme-like catalysts. The use of β-turn and α-helical motifs is one approach that enables the precise placement of reactive functional groups to enable selective substrate activation and reactivity/selectivity that approaches natural enzymes. Our recent work has demonstrated that helical peptides can serve as scaffolds for pre-organizing two reactive groups to achieve enzyme-like catalysis. In this study, we used CYANA and AmberTools to develop a computational approach for determining how the structure of our peptide catalysts can lead to enhancements in reactivity. These results support our hypothesis that the bifunctional nature of the peptide enables catalysis by pre-organizing the two catalysts in reactive conformations that accelerate catalysis by proximity. We also present evidence that the low reactivity of monofunctional peptides can be attributed to interactions between the peptide-bound catalyst and the helical backbone, which are not observed in the bifunctional peptide.

Highly Branched Tannin-Tris(2-aminoethyl)amine-Urea Wood Adhesives.

Project description:Condensed tannin copolymerized with hyperbranched tris(2-aminoethyl)amine-urea formed by amine-amido deamination yields a particleboard thermosetting adhesive without any aldehydes satisfying the requirements of relevant standards for the particleboard internal bond strength. The tannin-triamine-urea cures well at 180 °C, a relatively low temperature for today's particleboard hot pressing. As aldehydes were not used, the formaldehyde emission was found to be zero, not even in traces due to the heating of wood. The effect is ascribed to the presence of many reactive sites, such as amide, amino, and phenolic groups belonging to the three reagents used. The tannin appears to function as an additional cross-linking agent, almost a nucleating agent, for the triamine-urea hyperbranched oligomers. Chemical analysis by MALDI ToF and 13C NMR has shown that the predominant cross-linking reaction is that of the substitution of the tannin phenolic hydroxyls by the amino groups of the triamine. The reaction of tannin with the still-free amide groups of urea is rather rare, but it may occur with the rarer tannin flavonoid units in which the heterocyclic ring is opened. Due to the temperature gradient between the surfaces and the board core in the particleboard during hot pressing, the type and the relative balance of covalent and ionic bonds in the resin structure may differ in the surfaces and the board core.

Project description:The higher order structure (HOS) of protein therapeutics is essential for drug safety and efficacy and can be evaluated by two-dimensional (2D) nuclear magnetic resonance (NMR) spectroscopy at atomic resolution. 1Hn-15N amide correlated and 1H-13C methyl correlated NMR spectroscopies at natural isotopic abundance have been demonstrated as feasible on protein therapeutics as large as monoclonal antibodies and show great promise for use in establishing drug substance structural consistency across manufacturing changes and in comparing a biosimilar to an originator reference product. Spectral fingerprints from 1Hn-1Hα correlations acquired using 2D homonuclear proton-proton J-correlated NMR experiments provide a complementary approach for high-resolution assessment of the HOS of lower molecular weight (<25 kDa) protein therapeutics. Here, we evaluate different pulse sequences (COSY, TOCSY and TACSY) used to generate proton-proton J-correlated NMR spectral fingerprints and appraise the performance of each method for application to protein therapeutic HOS assessment and comparability.

A Study of Concept to Prepare Totally Biosourced Wood Adhesives from Only Soy Protein and Tannin.

Project description:This is a study of concept on the initial application for wood adhesives totally biosourced from the covalent reaction between soy protein isolate (SPI) and a commercial flavonoid tannin, namely quebracho tannin. The adhesive is composed exclusively of the two vegetable biomaterials mentioned and thus is totally biosourced and non-toxic, as tannin has been classified as being not at all toxic by the European Commission REACH program. The pre-reaction between the two yielded the best plywood bonding results when limited to a temperature of 40 °C, final cross-linking being achieved during the plywood higher temperature hot pressing procedure, as for any other thermosetting adhesive. Pre-reaction at higher temperatures, namely 60 °C and 80 °C, achieved extensive premature cross-linking that lost any activity to cross-link further when hot pressed for preparing plywood. The reaction was followed by thermomechanical analysis, by matrix assisted laser desorption ionization time of flight (MALDI ToF) mass spectrometry, and by plywood shear strength tested dry, after a 24 h cold water soak and 1 h in boiling water. The adhesive of this approach lends itself to be further reinforced by the multitude of approaches on soy resins already developed by several other research groups.

Debondable phenoxy-based structural adhesives with β-amino amide containing reversible crosslinkers.

Project description:This study introduces dynamic phenoxy-based adhesives using β-aminoamide exchange chemistry, designed for durability, reprocessability and sustainability. Synthesized through a two-step process, the adhesive features linear poly-aminoamides and tailored amine formulations to optimize adhesion, flexibility, and the glass transition. The corresponding phenoxy-based adhesives demonstrated effective crosslinking, high thermal stability (T d5% ∼340-350 °C), and temperature-responsive viscoelastic properties. Notably, the materials with 5 mol% of TETA (E-BAAT5) exhibited ideal activation energy for stress relaxation, exceptional creep resistance, and retained up to 98% lap shear strength after recycling, with controlled debonding at elevated temperatures, making it ideal for high-performance, recyclable adhesive applications.

2D NMR Analysis as a Sensitive Tool for Evaluating the Higher-Order Structural Integrity of Monoclonal Antibody against COVID-19.

Project description:The higher-order structure (HOS) of protein therapeutics has been confirmed as a critical quality parameter. In this study, we compared 2D 1H-13C ALSOFAST-HMQC NMR spectra with immunochemical ELISA-based analysis to evaluate their sensitivity in assessing the HOS of a potent human monoclonal antibody (mAb) for the treatment of coronavirus disease 2019 (COVID-19). The study confirmed that the methyl region of the 2D 1H-13C NMR spectrum is sensitive to changes in the secondary and tertiary structure of the mAb, more than ELISA immunoassay. Because of its highly detailed level of characterization (i.e., many 1H-13C cross-peaks are used for statistical comparability), the NMR technique also provided a more informative outcome for the product characterization of biopharmaceuticals. This NMR approach represents a powerful tool in assessing the overall higher-order structural integrity of mAb as an alternative to conventional immunoassays.

NMR-based Structural Analysis of Threonylcarbamoyl-AMP Synthase and Its Substrate Interactions.

Project description:The hypermodified nucleoside N(6)-threonylcarbamoyladenosine (t(6)A37) is present in many distinct tRNA species and has been found in organisms in all domains of life. This post-transcriptional modification enhances translation fidelity by stabilizing the anticodon/codon interaction in the ribosomal decoding site. The biosynthetic pathway of t(6)A37 is complex and not well understood. In bacteria, the following four proteins have been discovered to be both required and sufficient for t(6)A37 modification: TsaC, TsaD, TsaB, and TsaE. Of these, TsaC and TsaD are members of universally conserved protein families. Although TsaC has been shown to catalyze the formation of L-threonylcarbamoyl-AMP, a key intermediate in the biosynthesis of t(6)A37, the details of the enzymatic mechanism remain unsolved. Therefore, the solution structure of Escherichia coli TsaC was characterized by NMR to further study the interactions with ATP and L-threonine, both substrates of TsaC in the biosynthesis of L-threonylcarbamoyl-AMP. Several conserved amino acids were identified that create a hydrophobic binding pocket for the adenine of ATP. Additionally, two residues were found to interact with L-threonine. Both binding sites are located in a deep cavity at the center of the protein. Models derived from the NMR data and molecular modeling reveal several sites with considerable conformational flexibility in TsaC that may be important for L-threonine recognition, ATP activation, and/or protein/protein interactions. These observations further the understanding of the enzymatic reaction catalyzed by TsaC, a threonylcarbamoyl-AMP synthase, and provide structure-based insight into the mechanism of t(6)A37 biosynthesis.

Lignosulfonate-Based Polyurethane Adhesives.

Project description:The feasibility of using lignosulfonate (LS) from acid sulphite pulping of eucalyptus wood as an unmodified polyol in the formulation of polyurethane (PU) adhesives was evaluated. Purified LS was dissolved in water to simulate its concentration in sulphite spent liquor and then reacted with 4,4'-diphenylmethane diisocyanate (pMDI) in the presence or absence of poly(ethylene glycol) with Mw 200 (PEG200) as soft crosslinking segment. The ensuing LS-based PU adhesives were characterized by infrared spectroscopy and thermal analysis techniques. The adhesion strength of new adhesives was assessed using Automated Bonding Evaluation System (ABES) employing wood strips as a testing material. The results showed that the addition of PEG200 contributed positively both to the homogenization of the reaction mixture and better crosslinking of the polymeric network, as well as to the interface interactions and adhesive strength. The latter was comparable to the adhesive strength recorded for a commercial white glue with shear stress values of almost 3 MPa. The optimized LS-based PU adhesive formulation was examined for the curing kinetics following the Kissinger and the Ozawa methods by non-isothermal differential scanning calorimetry, which revealed the curing activation energy of about 70 kJ·mol-1.

Positive GABA(A) receptor modulators from Acorus calamus and structural analysis of (+)-dioxosarcoguaiacol by 1D and 2D NMR and molecular modeling.

Project description:In a two-microelectrode voltage clamp with Xenopus laevis oocytes, a petroleum ether extract of Acorus calamus rhizomes enhanced the GABA-induced chloride current through GABA(A) receptors of the ?????(2S) subtype by 277% ± 9.7% (100 ?g/mL). ?-Asarone (1), (+)-dioxosarcoguaiacol (2), (+)-shyobunone (3), and (+)-preisocalamenediol (4) were subsequently identified as main active principles through HPLC-based activity profiling and targeted isolation. The compounds induced maximum potentiation of the chloride current ranging from 588% ± 126% (EC??: 65.3 ± 21.6 ?M) (2) to 1200% ± 163% (EC(50): 171.5 ± 34.6 ?M) (1), whereas (-)-isoshyobunone (5) and (-)-acorenone (6) exhibited weak GABA(A) modulating properties (5: 164% ± 42.9%; EC??: 109.4 ± 46.6 ?M and 6: 241% ± 23.1%; EC??: 34.0 ± 6.7 ?M). The relative configuration of 2 was established as 4R*8S*10R* by NOESY experiments and conformational analysis.

A facile strategy for synthesizing isosorbide-based polyurethane structural adhesives and core-shell rubber.

Project description:A structural adhesive series of biomass-based polyurethane (Biomass-PU) is synthesized using polypropylene glycol (PPG2000), isosorbide-based polyol (RPO300) as polyols, isophorone diisocyanate (IPDI) as an isocyanate and 4-tert-butylphenol (BP) as a capping agent. Three different equivalent ratios of PPG2000/RPO300, 9/1 (Biomass-PU1), 7/3 (Biomass-PU2), and 1/1 (Biomass-PU3), are evaluated to determine the effect of isosorbide-based polyol content on the properties and the optimizing formulation of biomass-PU structure adhesive. The 9/1 ratio of PPG2000/RPO300 substantially leads to the improvement of impact strength by up to 35 MPa, and the PPG2000/RPO300 = 9/1 ratio exhibits better thermal properties and impact strength than those of other ratios. To achieve more compatibility between biomass-PU structure adhesive and core-shell rubber (CSR) toughener, novel CSRs are successfully synthesized using acryl-PU as a shell and biomass-based PU as a core. The chemical structure of biomass-PU structure adhesives is analyzed through FT-IR Spectroscopy and NCO% titration. Thermal properties are evaluated using TGA and DSC analysis. Their molecular weights are measured by GPC. Also, the core-shell rubber (CSR) with a polyurethane shell is prepared to reinforce the impact strength of Biomass-PU structure adhesive.
