Project description:The functional success of a biomedical implant critically depends on its stable bonding with the host tissue. Aseptic implant loosening accounts for more than half of all joint replacement failures. Various materials, including metals and plastic, confer mechanical integrity to the device, but often these materials are not suitable for direct integration with the host tissue, which leads to implant loosening and patient morbidity. We describe a self-assembled, osteogenic, polymer-based conformal coating that promotes stable mechanical fixation of an implant in a surrogate rodent model. A single modular, polymer-based multilayered coating was deposited using a water-based layer-by-layer approach, by which each element was introduced on the surface in nanoscale layers. Osteoconductive hydroxyapatite (HAP) and osteoinductive bone morphogenetic protein-2 (BMP-2) contained within the nanostructured coating acted synergistically to induce osteoblastic differentiation of endogenous progenitor cells within the bone marrow, without indications of a foreign body response. The tuned release of BMP-2, controlled by a hydrolytically degradable poly(?-amino ester), was essential for tissue regeneration, and in the presence of HAP, the modular coating encouraged the direct deposition of highly cohesive trabecular bone on the implant surface. In vivo, the bone-implant interfacial tensile strength was significantly higher than standard bioactive bone cement, did not fracture at the interface, and had long-term stability. Collectively, these results suggest that the multilayered coating system promotes biological fixation of orthopedic and dental implants to improve surgical outcomes by preventing loosening and premature failure.

Project description:Infections associated with orthopedic implants cause increased morbidity and significant healthcare cost. A prolonged and expensive two-stage procedure requiring two surgical steps and a 6-8 week period of joint immobilization exists as today's gold standard for the revision arthroplasty of an infected prosthesis. Because infection is much more common in implant replacement surgeries, these issues greatly impact long-term patient care for a continually growing part of the population. Here, we demonstrate that a single-stage revision using prostheses coated with self-assembled, hydrolytically degradable multilayers that sequentially deliver the antibiotic (gentamicin) and the osteoinductive growth factor (BMP-2) in a time-staggered manner enables both eradication of established biofilms and complete and rapid bone tissue repair around the implant in rats with induced osteomyelitis. The nanolayered construct allows precise independent control of release kinetics and loading for each therapeutic agent in an infected implant environment. Antibiotics contained in top layers can be tuned to provide a rapid release at early times sufficient to eliminate infection, followed by sustained release for several weeks, and the underlying BMP-2 component enables a long-term sustained release of BMP-2, which induced more significant and mechanically competent bone formation than a short-term burst release. The successful growth factor-mediated osteointegration of the multilayered implants with the host tissue improved bone-implant interfacial strength 15-fold when compared with the uncoated one. These findings demonstrate the potential of this layered release strategy to introduce a durable next-generation implant solution, ultimately an important step forward to future large animal models toward the clinic.

Project description:Discrete block co-oligomers (BCOs) are gaining considerable attention due to their potential to form highly ordered ultrasmall nanostructures suitable for lithographic templates. However, laborious synthetic routes present a major hurdle to the practical application. Herein, we report a readily available discrete BCO system that is capable of forming various self-assembled nanostructures with ultrasmall periodicity. Click coupling of propargyl-functionalized sugars (containing 1-7 glucose units) and azido-functionalized terpenoids (containing 3, 4, and 9 isoprene units) afforded the discrete and monodisperse BCOs with a desired total degree of polymerization and block ratio. These BCOs microphase separated into lamellar, gyroid, and cylindrical morphologies with the domain spacing (d) of 4.2-7.5 nm. Considering easy synthesis and rich phase behavior, presented BCO systems could be highly promising for application to diverse ~4-nm nanofabrications.

Project description:Phenolic-matrix composites possess excellent synergistic effects on mechanical and tribological properties and can be used in the aerospace, medical, and automobile industries. In this work, a series of phenol-formaldehyde resin/hexagonal boron nitride nanocomposites (PF/BNs) were in situ synthesized using an easy method. PF/BN coatings (PF/BNCs) on 316L steels were prepared through a spin-casting method. The wear behaviors of these PF/BNCs were investigated by dry sliding with steel balls. The percentage of BN, the thickness of the coating, and the heat treatment temperature affected the coefficients of friction (COFs) and wear rates of these coatings. After heat treatment at 100 °C, the tribological properties of the PF/BNCs were remarkably improved, which might be attributed to both the transformation of carbon on the worn surfaces from C-O/C=O into C=N, carbide, and other chemical bonds and the cross-linking of the prepolymers.

Project description:This study reports a systematic investigation of fine-tuning the filtration performance of nanofiltration membranes with biophenol coatings to produce solvent-resistant membranes with 390-1550 g mol-1 molecular weight cutoff (MWCO) and 0.5-40 L m-2 h-1 bar-1 permeance. Six kinds of inexpensive, commercial biophenols (dopamine, tannic acid, vanillyl alcohol, eugenol, morin, and quercetin) were subjected to identical oxidant-promoted polymerization to coat six kinds of loose asymmetric membrane supports: polyimide (PI), polyacrylonitrile (PAN), polysulfone (PSf), polyvinylidene difluoride (PVDF), polybenzimidazole (PBI), and polydimethylsiloxane (PDMS). The coatings were characterized by Fourier-transform infrared spectroscopy (FTIR), and the morphologies were characterized by scanning electron microscopy (SEM) and atomic force microscopy (AFM). The long-term stability of 42 membranes were tested in 12 organic solvents, including emerging green solvents MeTHF and Cyrene. The biophenol coatings led to tighter membranes with a decrease in MWCO of 12-79% at a penalty of a 22-92% permeance decrease in acetone.

Project description:The dual threats posed by the COVID-19 pandemic and hospital-acquired infections (HAIs) have emphasized the urgent need for self-disinfecting materials for infection control. Despite their highly potent antimicrobial activity, the adoption of photoactive materials to reduce infection transmission in hospitals and related healthcare facilities has been severely hampered by the lack of scalable and cost-effective manufacturing, in which case high-volume production methods for fabricating aPDI-based materials are needed. To address this issue here, we examined the antimicrobial efficacy of a simple bicomponent spray coating composed of the commercially-available UV-photocrosslinkable polymer N-methyl-4(4'-formyl-styryl)pyridinium methosulfate acetal poly(vinyl alcohol) (SbQ-PVA) and one of three aPDI photosensitizers (PSs): zinc-tetra(4-N-methylpyridyl)porphine (ZnTMPyP4+), methylene blue (MB), and Rose Bengal (RB). We applied these photodynamic coatings, collectively termed SbQ-PVA/PS, to a variety of commercially available materials. Scanning electron microscopy (SEM) and time-of-flight secondary ion mass spectrometry (ToF-SIMS) confirmed the successful application of the coatings, while inductively coupled plasma-optical emission spectroscopy (ICP-OES) revealed a photosensitizer loading of 0.09-0.78 nmol PS/mg material. The antimicrobial efficacy of the coated materials was evaluated against methicillin-susceptible Staphylococcus aureus ATCC-29213 and human coronavirus strain HCoV-229E. Upon illumination with visible light (60 min, 400-700 nm, 65 ± 5 mW/cm2), the coated materials inactivated S. aureus by 97-99.999% and HCoV-229E by 92-99.999%, depending on the material and PS employed. Photobleaching studies employing HCoV-229E demonstrated detection limit inactivation (99.999%) even after exposure for 4 weeks to indoor ambient room lighting. Taken together, these results demonstrate the potential for photodynamic SbQ-PVA/PS coatings to be universally applied to a wide range of materials for effectively reducing pathogen transmission.

Project description:Superhydrophobic materials rely on both chemical apolarity and surface roughness to achieve the high contact angles and the low roll-off angles that lead to self-cleaning and antibacterial properties. Current superhydrophobic coatings tend to be delicate and lose their properties easily when subjected to droplet impact. Such impact deteriorates these coatings through hydrodynamic wear; changing structure, eroding hydrophobic chemistry, and quickly leading to full wet out of the substrate. In fact, hydrodynamic wear is more detrimental to coatings than seemingly more aggressive mechanical wear including scratching with sandpaper - a common approach used to claim both self-similarity of a material and extreme robustness against wear. What makes certain coatings more robust against hydrodynamic wear? To understand this answer, we systematically study ten disparate self-similar superhydrophobic coating approaches from academia to industry by subjecting them to hydrodynamic wear with rapid droplet impacts. We offer an iteration of a spinning disk methodology that enables parallel testing of multiple coatings simultaneously. We have developed an analytical model that accurately estimates the average size and velocity of droplets created from the spinning disk. We find rapid droplet impacts that simulate a medium rain can deteriorate most coatings within seconds or minutes, with certain exceptions lasting up to 22 days. The more resilient coatings share common attributes including robust apolar chemistry, hierarchal topography, and a slow loss of sacrificial material. The best performing coatings can be characterized using power-law relationships that parallel mechanical fatigue functions and provide a predictive quantitative metric for the performance of hydrophobic coatings. Overall, this paper offers a quantitative approach to hydrodynamic wear of self-similar superhydrophobic coatings.

Project description:Heart failure (HF) is a modern epidemic and a heterogeneous disorder with many therapeutic options. While the average response to each individual treatment is favorable, significant interindividual variation exists in the response to HF therapeutics. As a result, the optimal regimen for an individual patient or subgroup of patients is elusive, with current treatment being mainly empirical. Pharmacogenetic customization of HF therapy may provide an important opportunity to improve the treatment of HF. Common genetic variations exist in genes related to most classes of HF drugs, many of which have known functional consequences for or established relationships with drug response. This review summarizes the current understanding of the pharmacogenetics of HF therapeutics, including angiotensin-converting enzyme inhibitors and beta-blockers, and focuses on recent advances and medium-term expectations for the field.

Project description:Friction and wear cause energy wastage and system failure. Usually, thicker overcoats serve to combat such tribological concerns, but in many contact sliding systems, their large thickness hinders active components of the systems, degrades functionality, and constitutes a major barrier for technological developments. While sub-10-nm overcoats are of key interest, traditional overcoats suffer from rapid wear and degradation at this thickness regime. Using an enhanced atomic intermixing approach, we develop a ~7- to 8-nm-thick carbon/silicon nitride (C/SiN x ) multilayer overcoat demonstrating extremely high wear resistance and low friction at all tribological length scales, yielding ~2 to 10 times better macroscale wear durability than previously reported thicker (~20 to 100 nm) overcoats on tape drive heads. We report the discovery of many fundamental parameters that govern contact sliding and reveal how tuning atomic intermixing at interfaces and varying carbon and SiN x thicknesses strongly affect friction and wear, which are crucial for advancing numerous technologies.

Project description:Stress-induced reactions at the sliding interface during relative movement are known to cause structural or chemical modifications in contacting materials. The nature of these modifications at the atomic level and formation of byproducts in an oil-free environment, however, remain poorly understood and pose uncertainties in predicting the tribological performance of the complete tribosystem. Here, we demonstrate that tribochemical reactions occur even in dry conditions when hydrogenated diamond-like carbon (H-DLC) surface is slid against two-dimensional (2D) molybdenum disulfide along with nanodiamonds in dry nitrogen atmosphere. Detailed experimental studies coupled with reactive molecular dynamics simulations reveal that at high contact pressures, diffusion of sulfur from the dissociated molybdenum disulfide led to amorphization of nanodiamond and subsequent transformation to onion-like carbon structures (OLCs). The in situ formation of OLCs at the sliding interface provide reduced contact area as well as incommensurate contact with respect to the H-DLC surface, thus enabling successful demonstration of superlubricity.
