Matrix Isolation FTIR and Theoretical Study of Weakly Bound Complexes of Isocyanic Acid with Nitrogen.

ABSTRACT: Weak complexes of isocyanic acid (HNCO) with nitrogen were studied computationally employing MP2, B2PLYPD3 and B3LYPD3 methods and experimentally by FTIR matrix isolation technique. The results show that HNCO interacts specifically with N2. For the 1:1 stoichiometry, three stable minima were located on the potential energy surface. The most stable of them involves a weak, almost linear hydrogen bond from the NH group of the acid molecule to nitrogen molecule lone pair. Two other structures are bound by van der Waals interactions of N⋯N and C⋯N types. The 1:2 and 2:1 HNCO complexes with nitrogen were computationally tracked as well. Similar types of interactions as in the 1:1 complexes were found in the case of the higher stoichiometry complexes. Analysis of the HNCO/N2/Ar spectra after deposition indicates that the 1:1 hydrogen-bonded complex is prevalent in argon matrices with a small amount of the van der Waals structures also present. Upon annealing, complexes of the 1:2 and 2:1 stoichiometry were detected as well.

Project description:Fumaric and maleic acids ((E)- and (Z)-HOOC-CH═CH-COOH, FA and MA) were studied experimentally by infrared spectroscopy in nitrogen matrixes and theoretically by quantum chemical calculations. The calculations, carried out at the DFT(B3LYP) and MP2 levels of theory, predicted the existence of at least 5 conformers of maleic acid and 10 conformers of fumaric acid. After the deposition of the matrixes, two conformers of maleic acid (I and II) and three conformers (I-III) of fumaric acid were observed and characterized vibrationally. Selective narrowband near-infrared (NIR) excitation of the first OH stretching overtones of the different conformers of maleic and fumaric acids initially present in the matrixes allowed the generation of higher-energy forms, never before observed experimentally. In the case of maleic acid, conformers I (a cis-trans form, where cis and trans designate the conformation of the carboxylic groups of the molecule) and II (cis-cis) were found to generate the novel conformers VI (trans-trans) and VII (cis-trans), respectively. The conversion of conformer II into the most stable conformer I was also observed. For fumaric acid, the cis-cis conformers I-III were found to give rise to the new cis-trans conformers IV-VII, respectively. The tunneling decay of the new conformers produced upon NIR excitation of the lowest-energy conformers initially trapped in the matrixes was observed, and their lifetimes in solid N2 were determined. The increased stability of all of the observed high-energy conformers of the studied acids in the N2 matrix, compared to the argon matrix, where they could not be observed experimentally, demonstrates the stabilizing effect of the interaction between the OH groups of the acids with the matrix N2 molecules, in line with previous observations for other carboxylic acids. In addition, the photochemistry of matrix-isolated maleic and fumaric acids upon broad-band UV irradiation (λ > 235 nm) was also investigated. UV-induced isomerization of both acids around the C═C double bond was observed, together with their decarboxylation to acrylic acid.

Project description:Identification of matrix-bound nanovesicles (MBV) as ubiquitous components of the extracellular matrix (ECM) raises questions regarding their biologic functions and their potential theranostic application. Unlike liquid-phase extracellular vesicles (e.g., exosomes), MBV are tightly bound to the ECM, which makes their isolation and harvesting more challenging. The indiscriminate use of different methods to harvest MBV can alter or disrupt their structural and/or functional integrity. The objective of the present study was to compare the effect of various MBV harvesting methods upon yield, purity, and biologic activity. Combinations of four methods to solubilize the ECM (collagenase [COL], liberase [LIB], or proteinase K [PK] and nonenzymatic elution with potassium chloride) and four isolation methods (ultracentrifugation, ultrafiltration [UF], density barrier, and size exclusion chromatography [SEC]) were used to isolate MBV from urinary bladder-derived ECM. All combinations of solubilization and isolation methods allowed for the harvesting of MBV, however, distinct differences were noted. The highest yield, purity, cellular uptake, and biologic activity were seen with MBV isolated by a combination of liberase or collagenase followed by SEC. The combination of proteinase K and UF was shown to have detrimental effects on bioactivity. The results show the importance of selecting appropriate MBV harvesting methods for the characterization and evaluation of MBV and for analysis of their potential theranostic application. Impact statement Identification of matrix-bound nanovesicles (MBV) as ubiquitous components of the extracellular matrix (ECM) has raised questions regarding their biologic functions and their potential theranostic application. This study demonstrates that the harvesting methods used can result in samples with physical and biochemical properties that are unique to the isolation and solubilization methods used. Consequently, developing harvesting methods that minimize sample contamination with ECM remnants and/or solubilization agents will be essential in determining the theranostic potential of MBV in future studies.

Project description:Isocyanic acid, HNCO, mainly emitted by combustion processes, is doubted to be detrimental to human health if its concentration surpasses ∼1 ppbv. Very little information has been found regarding the HNCO loss in the gas phase. This study aims to close this knowledge gap by performing a theoretical kinetic study on the reaction of HNCO with the propargyl radical. The potential energy surface of the HNCO + C3H3 reaction was characterized utilizing high-level CCSD(T)/CBS(TQ5)//B3LYP/6-311++G(3df,2p) quantum-chemical approaches, followed by TST and RRKM/ME kinetic computations. The obtained results reveal that the reaction can proceed via H-abstraction, leading to the C3H4 + NCO bimolecular products with energy barriers of 23-25 kcal/mol, and/or addition, resulting in C4H4NO intermediates with 23-26 kcal/mol barrier heights. The C4H4NO adducts when formed can decompose to products and/or return to HNCO + C3H3 in which the reverse decompositions are found to be dominant with a branching ratio that accounts for nearly 100% at 300 K and 760 Torr. The calculated P-independent rate coefficients indicate that at low temperatures, the H-abstraction channels are insignificant. However, at high temperatures (T > 1500 K), the H-abstraction path leading to H3CCCH + NCO prevails with a branching ratio of ∼50-53% in the descending 1800-1500 K temperature range at 760 Torr, while the H-abstraction leading to H2CCCH2 + NCO is favorable at T > 1800 K, with the yield reaching above 50% at 760 Torr. In contrast to the H-abstraction rate constants, the calculated values for the additions and the C4H4NO decompositions show a positive pressure dependence. Both the total rate constants for the reactions HNCO + C3H3 → products and C4H4NO → products, which are, respectively, k _total_bimo(T) = 3.53 × 10-23 T 3.27 exp[(-21.35 ± 0.06 kcal/mol)/RT] cm3 molecule-1 s-1 and k _total_uni(T) = 1.13 × 1025 T -4.02 exp[(-11.77 ± 0.16 kcal/mol)/RT] s-1, increase with the increasing temperature in the 300-2000 K range at 760 Torr. The rate constant of HNCO + C3H3 → products is about 8 orders of magnitude smaller than the value of HCHO + C3H3 → products, showing that HCHO is more reactive toward the C3H3 free radicals than HNCO. The computed heats of formation for several species agree well with the available literature data with the deviation less than 1.0 kcal/mol, indicating that the methods used in this study are extremely reliable. With the given results, it is vigorously suggested that the predicted rate constants, together with the thermodynamic data of the species involved, can be confidently used for modeling HNCO-related systems under atmospheric and combustion conditions.
