Project description:Transcription activator-like effector nuclease (TALEN) is a programmable genome editing tool with wide applications. Since TALENs perform cleavage of DNA as heterodimers, a pair of TALENs must be synthesized for each target genome locus. Conventionally, TALEN pairs are either expressed on separate vectors or synthesized separately and then subcloned to the same vector. Neither approach allows high-throughput construction of TALEN libraries for large-scale applications. Here we present a single-step assembly scheme to synthesize and express a pair of TALENs in a single-transcript format with the help of a P2A self-cleavage sequence. Furthermore, we developed a fully automated platform to custom manufacture TALENs in a versatile biological foundry. 400 pairs of TALENs can be synthesized with over 96.2% success rate at a material cost of $2.1/pair. This platform opens the door to TALEN-based genome-wide studies.

Project description:Fully conjugated ladder copolymers have attracted considerable attention due to their unique fused-ring structure and optoelectronic properties. In this study, two fully conjugated ladder naphthalene diimide (NDI) copolymers, P(NDI-CZL) and P(NDI-TTL) with imine-bridged structures are presented in high yields. Both of the two copolymers have good solubility and high thermal stability. The corresponding compounds with the same structure as the copolymers were synthesized as model system. The yields for each step of the synthesis of the model compounds are higher than 95%. These results suggest that P(NDI-CZL) and P(NDI-TTL) can be synthesized successfully with fewer structural defects. The structures and optoelectronic properties of compounds and copolymers are investigated by NMR, fourier transform infrared spectroscopy (FTIR), ultraviolet-visible spectroscopy (UV-vis), and cyclic voltammetry (CV). Both in solution and as a thin film, the two copolymers show two UV-vis absorption bands (around 300⁻400 nm and 400⁻750 nm) and a very weak fluorescence. The collective results suggest that the two fully conjugated ladder copolymers can be used as potential acceptor materials.

Project description:The crystallization and morphology of PLA-mb-PBS copolymers and their corresponding stereocomplexes were studied. The effect of flexible blocks (i.e., polybutylene succinate, PBS) on the crystallization of the copolymers and stereocomplex formation were investigated using polarized light optical microscopy (PLOM), differential scanning calorimetry (DSC), infrared spectroscopy (FTIR), and carbon-13 nuclear magnetic resonance spectroscopy (13C-NMR). The PLA and PBS multiple blocks were miscible in the melt and in the glassy state. When the PLA-mb-PBS copolymers are cooled from the melt, the PLA component crystallizes first creating superstructures, such as spherulites or axialites, which constitute a template within which the PBS component has to crystallize when the sample is further cooled down. The Avrami theory was able to fit the overall crystallization kinetics of both semi-crystalline components, and the n values for both blocks in all the samples had a correspondence with the superstructural morphology observed by PLOM. Solution mixtures of PLLA-mb-PBS and PLDA-mb-PBS copolymers were prepared, as well as copolymer/homopolymer blends with the aim to study the stereocomplexation of PLLA and PDLA chain segments. A lower amount of stereocomplex formation was observed in copolymer mixtures as compared to neat L100/D100 stereocomplexes. The results show that PBS chain segments perturb the formation of stereocomplexes and this perturbation increases with the amount of PBS in the samples. However, when relatively low amounts of PBS in the copolymer blends are present, the rate of stereocomplex formation is enhanced. This effect dissappears when higher amounts of PBS are present. The stereocomplexation was confirmed by FTIR and solid state 13C-NMR analyses.

Project description:Recent advances in the fundamental understanding of the ordered phases of multi-block copolymers (MBCPs) at the molecular level have attracted considerable scientific interest in recent years. Herein, by employing molecular dynamics simulation, we focus on the four typical systems: linear alternating, branch-like, star-like AB-type MBCPs and linear copolymers filled with nanoparticles (NPs). First, we establish the phase diagram for the linear tetrablock copolymers (ABAB) as a function of the composition ratio between A- and B-block, exhibiting six typical phase states. Furthermore, increasing the mutual repulsive interaction strength, the temperature and the periodic dynamic shearing cycle result in the merging of spheres, presenting a clear beginning of the order-to-order transition (OOT) behavior. Second, we examine the branch-like and star-like copolymers and find that increasing branch density significantly leads to the occurrence of phase transition. Particularly, we illustrate that the sphere configurations of the MBCPs can be described in terms of tail, loop and bridge conformations. Increasing the number of distinct blocks in linear alternating copolymers results in an enhancement of the bridge conformation, in which case some spheres are separated to smaller ones. Furthermore, for the tail conformation, we present a unified theoretical framework to rationalize the topological state of the chain arrangements of spheres and infer that the entanglements within the internal reaction layer between different A-blocks result in the inhomogeneous distribution of the spheres sizes even with controlled molecular weight and composition ratio between each block. Finally, we find that the ABAB tetrablock copolymers filled with moderate spherical NPs exhibit a clear OOT from spheres to double gyroid or cylinders. We infer that the maximum amount of the B-block within the second and/or third layers for the filled spherical NPs connects different NPs effectively, leading to the complicated OOT behavior. Generally, this fundamental study could provide some guidelines for designing and fabricating high performance BCPs by manipulating the formation of the ordered phases.

Project description:Fullerene derivatives have been widely used for conventional acceptor materials in organic photovoltaics (OPVs) because of their high electron mobility. However, there are also considerable drawbacks for use in OPVs, such as negligible light absorption in the visible-near-IR regions, less compatibility with donor polymeric materials and high cost for synthesis and purification. Therefore, the investigation of non-fullerene acceptor materials that can potentially replace fullerene derivatives in OPVs is increasingly necessary, which gives rise to the possibility of fabricating all-polymer (polymer/polymer) solar cells that can deliver higher performance and that are potentially cheaper than fullerene-based OPVs. Recently, considerable attention has been paid to donor-acceptor (D-A) block copolymers, because of their promising applications as fullerene alternative materials in all-polymer solar cells. However, the synthesis of D-A block copolymers is still a challenge, and therefore, the establishment of an efficient synthetic method is now essential. This review highlights the recent advances in D-A block copolymers synthesis and their applications in all-polymer solar cells.

Project description:Multi-block glucans comprising permethylated and partially methylated blocks are compounds of interest. In order to monitor their formation by transglycosylation of corresponding starting glucans, a method has been developed and applied to model compounds. This method allows determining the average length of the blocks and the progress of incorporation of methyl blocks in partially methylated sequences with a random distribution. The method, comprising liquid chromatography mass spectrometry (LC-MS) and electrospray ionization collision-induced dissociation tandem mass spectrometry (ESI-CID-MSn) measurements of two types of peralkylated glucans representing derivatives of the target compounds, is comprehensively described and discussed. ESI-MSn allows looking into the sequences of oligomeric domains. In addition, transglycosylation is followed by attenuated total reflection FTIR and NMR spectroscopy. Graphical abstract.

Project description:A series of N-substituted polyether-block-amide (PEBA-X%) copolymers were prepared by melt polycondensation of nylon-6 prepolymer and polytetramethylene ether glycol at an elevated temperature using titanium isopropoxide as a catalyst. The structure, thermal properties, and crystallinity of PEBA-X% were investigated using nuclear magnetic resonance spectroscopy, Fourier-transform infrared spectroscopy, differential scanning calorimetry, wide angle X-ray diffraction, and thermogravimetric analysis. In general, the crystallinity, melting point, and thermal degradation temperature of PEBA-X% decreased as the incorporation of N-methyl functionalized groups increased, owing to the disruption caused to the structural regularity of the copolymer. However, in N-acetyl functionalized analogues, the crystallinity first dropped and then increased because of a new γ form arrangement that developed in the microstructure. After the cross-linking reaction of the N-methyl-substituted derivative, which has electron-donating characteristics, with poly(4,4'-methylenebis(phenyl isocyanate), the decomposition temperature of the resulting polymer significantly increased, whereas no such improvements could be observed in the case of the electro-withdrawing N-acetyl-substituted derivative, because of the incompleteness of its cross-linking reaction.

Project description:The introduction of chirality into aqueous self-assemblies by employing isotactic block copolymers (BCPs) is an emerging field of interest as it promises special membrane properties of polymersomes not accessible by atactic BCPs. However, isotactic BCPs typically exhibit crystalline behaviour, inducing high membrane stiffness and limiting their applicability in systems involving membrane proteins or sensitive cargo. In this study, an isotactic yet fully amorphous BCP is introduced which overcomes these limitations. Three BCPs composed of poly(butylene oxide)-block-poly(glycidol) (PBO-b-PG), differing solely in their tacticities (R/S, R and S), were synthesised and characterised regarding their structural, optical and thermal properties. Their self-assembly into homogenous phases of nanoscopic polymersomes (referred to as small unilamellar vesicles, SUVs) was analysed, revealing stability differences between SUVs composed of the different BCPs. Additionally, microscopic giant unilamellar vesicles (GUVs) were prepared by double emulsion microfluidics. Only the atactic BCP formed GUVs which were stable over several hours, whereas GUVs composed of isotactic BCPs ruptured within several minutes after formation. The ability of atactic PBO-b-PG to form microreactors was elucidated by reconstituting the membrane protein OmpF in the GUV membrane by microfluidics and performing an enzyme reaction inside its lumen. The system presented here serves as platform to design versatile vesicles with flexible membranes composed of atactic or isotactic BCPs. Hence, they allow for the introduction of chirality into nano- or microreactors which is a yet unstudied field and could enable special biotechonological applications.

Project description:Grubbs-catalyzed ring-opening metathesis polymerization (ROMP) of carborane- and phosphonate-containing monomers has been used for the generation of hybrid block copolymers. Molecular weights with Mn of 50,000 g/mol were readily obtained with polydispersity index values, ?, between 1.03?1.08. Reaction of the phospha ester and carborane substituted oxanorbornene block copolymer with trimethylsilyl bromide led to a new polymer with phosphonic acid functionalities. In application studies, the phospha-carborane functionalized block polymer was tested as heat resistance material. Thermal stability was investigated by thermal gravimetric analysis (TGA) and microscale combustion calorimetry (MCC) analysis. Thermal treatment and ceramic yield under air were directly correlated to the carborane content of the block copolymer. However, phosphorus content in the polymer was more crucial for the char residues when heated under nitrogen atmosphere. The peak heat release rate (PHRR) increased as the number of phosphonate functionalities increased. However, corresponding phosphonic acid derivatives featured a lower heat release rate and total heat release. Moreover, the phosphonic acid functionalities of the block copolymer offer efficient chelating capabilities for iron nanoparticles, which is of interest for applications in biomedicine in the future. The complexation with iron oxide nanoparticles was studied by transmission electron microscopy (TEM) and inductively coupled plasma mass spectrometry (ICP?MS).

Project description:Herein, we report a methodology for the synthesis of polyolefin containing block-copolymers using a catalytic postpolymerization modification strategy. The most common polyolefin grades are converted into macroinitiators using a cross-metathesis reaction. These functionalized polyolefins are then used to initiate living: coordinative ring opening polymerization of lactide, anionic ring opening polymerization of epoxide, and radical polymerization of styrene to yield the corresponding block copolymers. The high activity of the catalysts employed in the different steps offers improved practicality for scalable synthesis.
