Project description:Undoped and manganese doped zinc sulfide nanoparticles were produced by a fast, one-step and two-component microwave-assisted synthesis method. The solid phase retains around 78% of the initial Mn concentration, as revealed by Particle Induced X-ray Emission analysis. X-ray diffraction patterns confirmed zinc blende structure and in the transmission electron microscopy images, nanoparticles with triangular prism and cube shapes were observed, respectively with an average particle size around 7 nm and 13 nm. Dried powders of zinc sulfide nanoparticles, doped with 0.1 mol% and 0.7 mol% of Mn ions, show highest brilliance of luminescence under UV light. Increasing dopant levels resulted in a diminishing emission that vanishes above 4% of dopant concentration. The synthesis of ZnS was monitored and two main events were detected, one at 145 °C corresponding to the sol-gel phase formation and another after ~3 min at 300 °C where the precipitation of the zinc sulfide nanoparticles occurs.

Project description:Presenilin-mediated endoproteolysis of transmembrane proteins plays a key role in physiological signaling and in the pathogenesis of Alzheimer disease and some cancers. Numerous inhibitors have been found via library screens, but their structural mechanisms remain unknown. We used several biophysical techniques to investigate the structure of human presenilin complexes and the effects of peptidomimetic γ-secretase inhibitors. The complexes are bilobed. The head contains nicastrin ectodomain. The membrane-embedded base has a central channel and a lateral cleft, which may represent the initial substrate docking site. Inhibitor binding induces widespread structural changes, including rotation of the head and closure of the lateral cleft. These changes block substrate access to the catalytic pocket and inhibit the enzyme. Intriguingly, peptide substrate docking has reciprocal effects on the inhibitor binding site. Similar reciprocal shifts may underlie the mechanisms of other inhibitors and of the "lateral gate" through which substrates access to the catalytic site.

Project description:We report a simple, fast and green phosphine-free colloidal chemistry to synthesize high-quality wurtzite-type Mn-doped ZnS quantum rods (QRs) with tunable diameters (1.6-5.6 nm), high aspect ratios (up to 50), variable Mn doping levels (0.18-1.60%), and high quantum yields (up to 45%). The electron paramagnetic resonance spectra with modeling reveal the successful doping of paramagnetic Mn(2+) ions in the host ZnS QRs. The Mn-doped ZnS QRs demonstrate tunable dual-color (orange and blue) emissions by tuning the doping levels and UV excitation wavelengths. The orange emission with long decay lifetime (3.3 ms) originates from the doped Mn(2+) states, while the blue emission with fast decay lifetime (0.31 ns) is attributed to the QR surface states. The bright two- and three-photon excitation upconversion luminescence from the Mn-doped ZnS QRs have been observed using tunable near-infrared femtosecond laser. Our strategy provides a versatile route to programmably control the optical properties of anisotropic semiconductor nanomaterials, which may create new opportunities for photonic devices and bioimaging applications.

Project description:Zinc sulfide (ZnS) nanowires represent a promising candidate in many fields, including optoelectronics and photocatalysis because of their advantages such as excellent optical properties, chemical stability and an easy-scalable simple synthesis method. In this study, an energy-friendly microwave radiation process was used to develop the single-step, solvothermal process for the growth of manganese-doped zinc sulfide (ZnS) and undoped nanocrystals (NCs) in the forms of nanowires using two short amines as a stabilizer, e.g. ethylenediamine and hydrazine, respectively. ZnS nanowires doped with Mn atoms show absorbance in UV and in the visible region of the spectrum. The photocatalytic degradation of rhodamine B in the presence of Mn-doped and undoped ZnS nanocrystals illuminated with only a 6-W UV lamp has been comprehensively studied. The effect of Mn doping and the presence of a nanocrystal stabilizer on the degradation process was determined. It was found that the efficiency of a photocatalytic degradation process was strongly affected by both factors: the doping process of nanowires with Mn2+ atoms and the attachment of ligands to the nanocrystal surface.

Project description:Bacteria naturally alter the redox state of many compounds and perform atom-by-atom nanomaterial synthesis to create many inorganic materials. Recent advancements in synthetic biology have spurred interest in using biological systems to manufacture nanomaterials, implementing biological strategies to specify the nanomaterial characteristics such as size, shape, and optical properties. Here, we combine the natural synthetic capabilities of microbes with engineered genetic control circuits toward biogenically synthesized semiconductor nanomaterials. Using an engineered strain of Shewanella oneindensis with inducible expression of the cytochrome complex MtrCAB, we control the reduction of manganese (IV) oxide. Cytochrome expression levels were regulated using an inducer molecule, which enabled precise modulation of dopant incorporation into manganese doped zinc sulfide nanoparticles (Mn:ZnS). Thereby, a synthetic gene circuit controlled the optical properties of biogenic quantum dots. These biogenically assembled nanomaterials have similar physical and optoelectronic properties to chemically synthesized particles. Our results demonstrate the promise of implementing synthetic gene circuits for tunable control of nanomaterials made by biological systems.

Project description:Global trials are grappling toward identifying prosperous remediation against the ever-emerging and re-emerging pathogenic respiratory viruses. Battling coronavirus, as a model respiratory virus, via repurposing existing therapeutic agents could be a welcome move. Motivated by its well-demonstrated curative use in herpes simplex and influenza viruses, utilization of the nanoscale zinc oxide (ZnO) would be an auspicious approach. In this direction, ZnO nanoparticles (NPs) were fabricated herein and relevant aspects related to the formulation such as optimization, structure, purity, and morphology were elucidated. In silico molecular docking was conducted to speculate the possible interaction between ZnO NPs and COVID-19 targets including the ACE2 receptor, COVID-19 RNA-dependent RNA polymerase, and main protease. The cellular internalization of ZnO NPs using human lung fibroblast cells was also assessed. Optimized hexagonal and spherical ZnO nanostructures of a crystallite size of 11.50 ± 0.71 nm and positive charge were attained. The pure and characteristic hexagonal wurtzite P63mc crystal structure was also observed. Interestingly, felicitous binding of ZnO NPs with the three tested COVID-19 targets, via hydrogen bond formation, was detected. Furthermore, an enhanced dose-dependent cellular uptake was demonstrated. The obtained results infer a rationale, awaiting validation from further biological and therapeutic studies.

Project description:This study reports the phase transformation behaviour associated with electrolytic manganese dioxide (EMD) utilized as the positive electrode active material for aqueous zinc-ion batteries. Electrochemical techniques, including galvanostatic charge-discharge and rotating ring-disk electrode measurements, and microstructural techniques, using X-ray powder diffraction, scanning electron microscopy, and transmission/scanning transmission electron microscopy, were utilized to characterize the positive electrode at different stages of discharge and charge of zinc-ion cells. The results indicate that, during discharge, a fraction of EMD undergoes a transformation to ZnMn2O4 (spinel-type) and Zn2+ is intercalated into the tunnels of the γ- and ε-MnO2 phases, forming ZnxMnO2 (tunnel-type). When a critical concentration of Mn3+ in the intercalated ZnxMnO2 species is reached, a disproportionation/dissolution reaction is triggered leading to the formation of soluble Mn2+ and hydroxide (OH-) ions; the latter precipitates as zinc hydroxide sulfate (ZHS, Zn4(OH)6(SO4)·5H2O) by combination with the ZnSO4/H2O electrolyte. During charge, Zn2+ is reversibly deintercalated from the intergrown tunneled phases (γ-/ε-ZnxMnO2), Mn2+ is redeposited as layered chalcophanite (ZnMn3O7·3H2O), and ZHS is decomposed by protons (H+) formed during the electrochemical deposition of chalcophanite.

Project description:Farmland soil typical for the Polish rural environment was used in pot experiment to estimate the impact of cadmium and zinc on the manganese, lead and copper uptake by lemon balm (Melissa officinalis L). Bioavailable and total forms of investigated metals in soil and metal concentrations in plants were determined by atomic absorption spectrometry. The plant photosynthesis indicators were also examined. Intensification of photosynthesis upon the high zinc and cadmium soil supplementation was observed. This effect was not detected at low metal concentrations. ANOVA proved that cadmium and zinc treatments influenced manganese, lead and copper transfer from soil and their concentration in plants. Zinc uptake and accumulation in either roots or above-ground parts in plant was inversely proportional to cadmium concentration in soil. Manganese concentration in roots decreased upon the soil supplementation with either zinc or cadmium. It suggests that the latter ions are transported via symplastic pathways and compete with manganese for similar transporters. The opposite situation was observed for lead and copper. Soil supplementation with cadmium and zinc affects manganese, lead and copper concentrations and photosynthesis intensity in lemon balm plant. The following combined interactions in either normal or stress conditions are important indicators of the migration pathways.

Project description:Birnessite is a layered MnO2 mineral capable of intercalating nanometric water films in its bulk. With its variable distributions of Mn oxidation states (MnIV, MnIII, and MnII), cationic vacancies, and interlayer cationic populations, birnessite plays key roles in catalysis, energy storage solutions, and environmental (geo)chemistry. We here report the molecular controls driving the nanoscale intercalation of water in potassium-exchanged birnessite nanoparticles. From microgravimetry, vibrational spectroscopy, and X-ray diffraction, we find that birnessite intercalates no more than one monolayer of water per interlayer when exposed to water vapor at 25 °C, even near the dew point. Molecular dynamics showed that a single monolayer is an energetically favorable hydration state that consists of 1.33 water molecules per unit cell. This monolayer is stabilized by concerted potassium-water and direct water-birnessite interactions, and involves negligible water-water interactions. Using our composite adsorption-condensation-intercalation model, we predicted humidity-dependent water loadings in terms of water intercalated in the internal and adsorbed at external basal faces, the proportions of which vary with particle size. The model also accounts for additional populations condensed on and between particles. By describing the nanoscale hydration of birnessite, our work secures a path for understanding the water-driven catalytic chemistry that this important layered manganese oxide mineral can host in natural and technological settings.

Project description:High dose manganese (Mn) exposure can result in changes in tissue concentrations of other essential metals due to Mn-induced alterations in metal absorption and competition for metal transporters and regulatory proteins. We evaluated responses in mice with a Parkin gene defect (parkin mice) and a wildtype strain (C57BL/6J) following neonatal Mn exposure. Neonatal parkin and C57BL/6J littermates were randomly assigned to 0, 11, or 25 mg Mn/kg-day dose groups with oral exposures occurring from postnatal day (PND) 1 through PND 28. We report liver, femur, olfactory bulb, striatum, and frontal cortex iron, copper, and zinc concentrations and changes in hepatic gene expression of different metal transporters in PND 29 parkin and C57BL/6J mice. A companion manuscript (Foster et al., 2017) [1] describes the primary study findings. This data provides insights into strain differences in the way Mn interacts with other trace metals in mice.
