Project description:Polyurea elastomers are utilized for a myriad of applications ranging from coatings and foams to dielectric materials for capacitors and actuators. However, current synthetic methods for polyureas rely on highly reactive isocyanates, solvents, and catalysts, which collectively pose serious safety considerations. This report details the synthesis and characterization of melt processible, poly(tetramethylene oxide) (PTMO)-based segmented polyurea elastomers utilizing an isocyanate-, solvent-, and catalyst-free approach. Dynamic mechanical analysis and differential scanning calorimetry suggested microphase separation between the hard and soft segments. Tensile analysis revealed high strain at break for all segmented copolymers between 340 and 770%, and tunable modulus between 0.76 and 29.5 MPa. Dielectric spectroscopy revealed that the composition containing 20 wt% hard segment offered the highest permittivity at 10.6 (1 kHz, 300 K) of the segmented copolymers, indicating potential as a dielectric elastomer.

Project description:A regio- and stereoselective silver-catalyzed formal carbene insertion into 1,3-dicarbonyls has been developed, using N-nosylhydrazones as diazo surrogates. Two new C-C bonds are constructed at the carbenic carbon center through the selective cleavage of the C-C(=O) σ-bond of acyclic 1,3-dicarbonyls, enabling the preparation of various synthetically useful polysubstituted γ-diketones, γ-ketoesters, and γ-ketoamides in high yields. The in situ formation of a donor-acceptor cyclopropane, via reaction of the enolate of the 1,3-dicarbonyl with an electrophilic silver carbenoid, is proposed as a key process in the catalytic cycle.

Project description:We report here "sandwich"-diimine copper complex-catalyzed trifluoroethylation and pentafluoropropylation of unactivated C(sp3 )-H bonds in alkyl esters, halides, and protected amines by employing CF3 CHN2 and CF3 CF2 CHN2 reagents. Reactions proceed in dichloromethane solvent at room temperature. Identical C-H functionalization conditions and stoichiometries are employed for generality and convenience. Selectivities for C-H insertions are higher for compounds possessing stronger electron-withdrawing substituents. Preliminary mechanistic studies point to a mechanism involving a pre-equilibrium forming a "sandwich"-diimine copper-CF3 CHN2 complex followed by rate-determining loss of nitrogen affording the reactive copper carbene. It reacts with trifluoromethyldiazomethane about 6.5 times faster than with 1-fluoroadamantane explaining the need for slow addition of the diazo compound.

Project description:Hemoproteins have recently emerged as promising biocatalysts for promoting a variety of carbene transfer reactions including cyclopropanation and Y-H insertion (Y = N, S, Si, B). For these and synthetic carbene transfer catalysts alike, achieving high chemoselectivity toward cyclopropanation in olefin substrates bearing unprotected Y-H groups has proven remarkably challenging due to competition from the more facile carbene Y-H insertion reaction. In this report, we describe the development of a novel artificial metalloenzyme based on an engineered myoglobin incorporating a serine-ligated Co-porphyrin cofactor that is capable of offering high selectivity toward olefin cyclopropanation over N-H and Si-H insertion. Intramolecular competition experiments revealed a distinct and dramatically altered chemoselectivity of the Mb(H64V,V68A,H93S)[Co(ppIX)] variant in carbene transfer reactions compared to myoglobin-based variants containing the native histidine-ligated heme cofactor or other metal/proximal ligand substitutions. These studies highlight the functional plasticity of myoglobin as a "carbene transferase" and illustrate how modulation of the cofactor environment within this metalloprotein scaffold represents a valuable strategy for accessing carbene transfer reactivity not exhibited by naturally occurring hemoproteins or transition metal catalysts.

Project description:Previous work has demonstrated that variants of a heme protein, Rhodothermus marinus cytochrome c (Rma cyt c), catalyze abiological carbene boron-hydrogen (B-H) bond insertion with high efficiency and selectivity. Here we investigated this carbon-boron bondforming chemistry with cyclic, lactone-based carbenes. Using directed evolution, we obtained a Rma cyt c variant BOR LAC that shows high selectivity and efficiency for B-H insertion of 5- and 6-membered lactone carbenes (up to 24,500 total turnovers and 97.1:2.9 enantiomeric ratio). The enzyme shows low activity with a 7-membered lactone carbene. Computational studies revealed a highly twisted geometry of the 7membered lactone carbene intermediate relative to 5- and 6-membered ones. Directed evolution of cytochrome c together with computational characterization of key iron-carbene intermediates has allowed us to expand the scope of enzymatic carbene B-H insertion to produce new lactone-based organoborons.

Project description:Expanding nature's catalytic repertoire to include reactions important in synthetic chemistry will open new opportunities for 'green' chemistry and biosynthesis. We demonstrate enzyme-catalyzed insertion of carbenoids into N-H bonds. This type of bond disconnection, which has no counterpart in nature, can be mediated by variants of the cytochrome P450 from Bacillus megaterium. The N-H insertion reaction takes place in water, provides the desired products in 26-83% yield, forms the single addition product exclusively, and does not require slow addition of the diazo component.

Project description:(2-((1-(4-chlorophenyl)-1H-pyrazol-3-yl)oxy)-N-(3,4-dichlorophenyl)-propanamide) is a new oil-soluble compound with good fungicidal activity against Rhizoctonia solani. Chitosan oligosaccharide (COS) is the depolymerization product of chitosan and can be developed into biological pesticides, growth regulators, and fertilizers due to its various bioactivities. COS is an oligomer of β- (1 → 4)-linked d -glucosamine and can be taken as a polyamine. In this study, microcapsules were prepared by interfacial polymerization of oil-soluble methylene diphenyl diisocyanate and water-soluble COS. The effects of several key preparation parameters, e.g., emulsifier dosage, agitation rate during emulsification, and core/shell ratio, on properties of the microcapsules such as the encapsulation efficiency, particle size, and size distribution were investigated. The microcapsules were characterized by infrared spectroscopy, thermogravimetric analysis, and scanning electron microscopy, etc., and the encapsulation efficiency and release behaviors were investigated. The results show that the microcapsules have a smooth surface and 93.3% of encapsulation efficiency. The microcapsules showed slow-release behavior following a first-order kinetic equation, and the accumulative release rates of the microcapsules with core/shell mass ratios of 8.0/4.0, 8.0/5.0, and 8.0/6.0, were 95.5%, 91.4%, and 90.1%, respectively, on day 30. Due to many high biological activities, biodegradability, and the pure nature of COS, microcapsules formed from COS are promising for applications in controlled release of pesticides, growth regulators, and fertilizer.

Project description:Engineered myoglobins have recently gained attention for their ability to catalyze a variety of abiological carbene transfer reactions including the functionalization of amines via carbene insertion into N-H bonds. However, the scope of myoglobin and other hemoprotein-based biocatalysts in the context of this transformation has been largely limited to aniline derivatives as the amine substrates and ethyl diazoacetate as the carbene donor reagent. In this report, we describe the development of an engineered myoglobin-based catalyst useful for promoting carbene N-H insertion reactions across a broad range of substituted benzylamines and α-diazo acetates with high efficiency (82-99% conversion), elevated catalytic turnovers (up to 7,000), and excellent chemoselectivity for the desired single insertion product (up to 99%). The scope of this transformation could be extended to cyclic aliphatic amines. These studies expand the biocatalytic toolbox available for the selective formation of C-N bonds, which are ubiquitous in many natural and synthetic bioactive compounds.
