Project description:Mössbauer-effect studies of the super-reduced form of Chromatium high-potential iron-sulphur protein indicate that the iron atoms are in a similar valency state to those in reduced ferredoxin from Clostridium pasteurianum, with possibly some inequivalence between the iron atoms within the four-iron centre. Mössbauer spectroscopy also shows magnetic differences between the four-iron centres in the two proteins.

Project description:1. The previous Mössbauer work on Chromatium high-potential iron-sulphur protein by Moss et al. (1968) and Evans et al. (1970) was extended to high applied magnetic fields. 2. Measurements of the reduced protein confirm that it is non-magnetic. 3. Spectra of the oxidized protein in applied magnetic fields clearly indicate that some iron atoms have a positive hyperfine field, which is evidence for antiferromagnetic coupling. 4. The spectra can be interpreted in terms of two types of iron atom with positive and negative hyperfine fields of 9 and 12T respectively. 5. A consideration of the chemical shifts and other evidence suggests formal valences of two Fe(3+) and two Fe(2+) atoms in the non-magnetic reduced state, and three Fe(3+) atoms and one Fe(2+) atom in the oxidized state. 6. However, no separate Fe(3+) and Fe(2+) spectra are seen, suggesting that the d electrons are not localized on particular iron atoms.

Project description:1. Rubredoxin isolated from the green photosynthetic bacterium Chloropseudomonas ethylica was similar in composition to those from anaerobic fermentative bacteria. Amino acid analysis indicated a minimum molecular weight of 6352 with one iron atom per molecule. 2. The circular-dichroism and electron-paramagnetic-resonance spectra of Ch. ethylica rubredoxin showed many similarities to those of Clostridium pasteurianum, but suggested that there may be subtle differences in the protein conformation about the iron atom. 3. Mössbauer-effect measurements on rubredoxin from Cl. pasteurianum and Ch. ethylica showed that in the oxidized state the iron (high-spin Fe(3+)) has a hyperfine field of 370+/-3kG, whereas in the reduced state (high-spin Fe(2+)) the hyperfine field tensor is anisotropic with a component perpendicular to the symmetry axis of the ion of about -200kG. For the reduced protein the sign of the electric-field gradient is negative, i.e. the ground state of the Fe(2+) is a [unk] orbital. There is a large non-cubic ligand-field splitting (Delta/k=900 degrees K), and a small spin-orbit splitting (D~+4.4cm(-1)) of the Fe(2+) levels. 4. The contributions of core polarization to the hyperfine field in the Fe(3+) and Fe(2+) ions are estimated to be -370 and -300kG respectively. 5. The significance of these results in interpretation of the Mössbauer spectra of other iron-sulphur proteins is discussed.

Project description:Both the oxidized and reduced forms of Hipip (high-potential iron--sulphur protein) are reduced (approx. 30% yields) by eaq.- in a single-stage process, rate constants 1.7 x 10(10) and 1.8 x 10(10) M-1 . s-1 respectively, at 25 degrees C, pH 7.0 (5 mM-phosphate). Super-reduced Hipip, which is formed in the latter case, has a spectrum which closely resembles that of reduced ferredoxin, i.e. Fe4S4 (SR)4(3-) clusters. The spectrum is stable over 2 s periods investigated. Super-reduced Hipip is reoxidized with O2, rate constant 4.8 x 10(6) M-1 . s-1 at 25 degrees C.

Project description:Fur protein plays key roles in regulating numerous genes in bacteria and is essential for intracellular iron concentration regulation. However, atomic level pictures of the iron binding site and its functional mechanism remain to be established. Here we present results of the first quantum chemical investigation of various first- and second-shell models and experimental Mössbauer data of E. Coli Fur, including 1) the first robust evidence that site 2 is the Fe binding site with a 3His/2Glu ligand set, being the first case in non-heme proteins, with computed Mössbauer data in excellent accord with experiment; 2) the first discovery of a conservative hydrogen bonding interaction in the iron binding site based on X-ray and homology structures; 3) the first atomic level hypothesis of active site reorganization upon iron concentration increase, triggering the conformational change needed for its function. These results shall facilitate structural and functional studies of Fur family proteins.

Project description:Detailed circular dichroism (CD), steady-state and time-resolved tryptophan fluorescence studies on the holo- and apo- forms of high potential iron protein (HiPIP) from Chromatium vinosum and its mutant protein have been carried out to investigate conformational properties of the protein. CD studies showed that the protein does not have any significant secondary structure elements in the holo- or apo- HiPIP, indicating that the metal cluster does not have any effect on formation of secondary structure in the protein. Steady-state fluorescence quenching studies however, suggested that removal of the iron-sulfur ([Fe(4)S(4)](3+)) cluster from the protein leads to an increase in the solvent accessibility of tryptophans, indicating change in the tertiary structure of the protein. CD studies on the holo- and apo- HiPIP also showed that removal of the metal prosthetic group drastically affects the tertiary structure of the protein. Time-resolved fluorescence decay of the wild type protein was fitted to a four-exponentials model and that of the W80N mutant was fitted to a three-exponentials model. The time-resolved fluorescence decay was also analyzed by maximum entropy method (MEM). The results of the MEM analysis agreed with those obtained from discrete exponentials model analysis. Studies on the wild type and mutants helped to assign the fast picosecond lifetime component to the W80 residue, which exhibits fast fluorescence energy transfer to the [Fe(4)S(4)](3+) cluster of the protein. Decay-associated fluorescence spectra of each tryptophan residues were calculated from the time-resolved fluorescence results at different emission wavelengths. The results suggested that W80 is in the hydrophobic core of the protein, but W60 and W76 are partially or completely exposed to the solvent.

Project description:The Mössbauer spectroscopy of iron carbides (α-Fe, γ'-FeC, η-Fe2C, ζ-Fe2C, χ-Fe5C2, h-Fe7C3, θ-Fe3C, o-Fe7C3, γ'-Fe4C, γ''-Fe4C, and α'-Fe16C2) is predicted utilizing the all electron full-potential linearized augmented plane wave (FLAPW) approach across various functionals from LDA to GGA (PBE, PBEsol, and GGA + U) to meta-GGA to hybrid functionals. To validate the predicted MES from different functionals, the single-phase χ-Fe5C2 and θ-Fe3C are synthesized in experiment and their experimental MES under different temperature (from 13 K to 298 K) are determined. The result indicates that the GGA functional (especially, the PBEsol) shows remarkable success on the prediction of Mössbauer spectroscopy of α-Fe, χ-Fe5C2 and θ-Fe3C with delocalized d electrons. From the reliable simulations, we propose a linear relationship between Bhf and μB with a slope of 12.81 T/μB for iron carbide systems and that the proportionality constant may vary from structure to structure.

Project description:Density Functional Theory (DFT) calculations on transition metal nitrosyls often reveal unusual spin density profiles, involving substantial spatial separation of majority and minority spin densities. Against this context, there is a significant lack of studies where DFT calculations have been quantitatively calibrated against experimental spectroscopic properties. Reported herein are DFT calculations of Mössbauer isomer shifts and quadrupole splittings for 21 nonheme iron complexes (26 distinct iron sites) including 9 iron nitrosyls. Low- (S = 1/2) and high-spin (S = 3/2) {FeNO}(7) complexes, S = 1/2 {Fe(NO)(2)}(9) species, and polynuclear iron nitrosyls are all represented within the set of compounds examined. The general conclusion with respect to isomer shifts is that DFT (OLYP/STO-TZP) performs comparably well for iron nitrosyls and for iron complexes in general. However, quadrupole splittings are less accurately reproduced for nitrosyl complexes.

Project description:The present study employs a suite of spectroscopic techniques to evaluate the electronic and bonding characteristics of the interstitial carbide in a set of iron-carbonyl-carbide clusters, one of which is substituted with a molybdenum atom. The M6C and M5C clusters are the dianions (Et4N)2[Fe6(μ6-C)(μ2-CO)2(CO)14] (1), [K(benzo-18-crown-6)]2[Fe5(μ5-C)(μ2-CO)1(CO)13] (2), and [K(benzo-18-crown-6)]2[Fe5Mo(μ6-C)(μ2-CO)2(CO)15] (3). Because 1 and 2 have the same overall cluster charge (2-) but different numbers of iron sites (1: 6 sites → 2: 5 sites), the metal atoms of 2 are formally oxidized compared to those in 1. Despite this, Mössbauer studies indicate that the iron sites in 2 possess significantly greater electron density (lower spectroscopic oxidation state) compared with those in 1. Iron K-edge X-ray absorption and valence-to-core X-ray emission spectroscopy measurements, paired with density functional theory spectral calculations, revealed the presence of significant metal-to-metal and carbide 2p-based character in the filled valence and low-lying unfilled electronic manifolds. In all of the above experiments, the presence of the molybdenum atom in 3 (Fe5Mo) results in somewhat unremarkable spectroscopic properties that are essentially a "hybrid" of 1 (Fe6) and 2 (Fe5). The overall electronic portrait that emerges illustrates that the central inorganic carbide ligand is essential for distributing charge and maximizing electronic communication throughout the cluster. It is evident that the carbide coordination environment is quite flexible and adaptive: it can drastically modify the covalency of individual Fe-C bonds based on local structural changes and redox manipulation of the clusters. In light of these findings, our data and calculations suggest a potential role for the central carbon atom in FeMoco, which likely performs a similar function in order to maintain cluster integrity through multiple redox and ligand binding events.

Project description:Vacuoles were isolated from fermenting yeast cells grown on minimal medium supplemented with 40 ?M (57)Fe. Absolute concentrations of Fe, Cu, Zn, Mn, Ca, and P in isolated vacuoles were determined by ICP-MS. Mössbauer spectra of isolated vacuoles were dominated by two spectral features: a mononuclear magnetically isolated high-spin (HS) Fe(III) species coordinated primarily by hard/ionic (mostly or exclusively oxygen) ligands and superparamagnetic Fe(III) oxyhydroxo nanoparticles. EPR spectra of isolated vacuoles exhibited a g(ave) ~ 4.3 signal typical of HS Fe(III) with E/D ~ 1/3. Chemical reduction of the HS Fe(III) species was possible, affording a Mössbauer quadrupole doublet with parameters consistent with O/N ligation. Vacuolar spectral features were present in whole fermenting yeast cells; however, quantitative comparisons indicated that Fe leaches out of vacuoles during isolation. The in vivo vacuolar Fe concentration was estimated to be ~1.2 mM while the Fe concentration of isolated vacuoles was ~220 ?M. Mössbauer analysis of Fe(III) polyphosphate exhibited properties similar to those of vacuolar Fe. At the vacuolar pH of 5, Fe(III) polyphosphate was magnetically isolated, while at pH 7, it formed nanoparticles. This pH-dependent conversion was reversible. Fe(III) polyphosphate could also be reduced to the Fe(II) state, affording similar Mössbauer parameters to that of reduced vacuolar Fe. These results are insufficient to identify the exact coordination environment of the Fe(III) species in vacuoles, but they suggest a complex closely related to Fe(III) polyphosphate. A model for Fe trafficking into/out of yeast vacuoles is proposed.
