Isolation and identification of carotenoids produced by a green alga (Dictyococcus cinnabarinus) in submerged culture.

ABSTRACT: 1. Six carotenoid pigments were produced by and isolated from the green alga Dictyococcus cinnabarinus grown in submerged culture in the presence of glucose. 2. The first, second and fourth pigments were identified respectively as beta-carotene, echinenone and canthaxanthin; the physicochemical properties of the other three are described and their similarity to other oxo-carotenoids is shown. 3. Culture techniques, isolation and identification procedures are described.

Four carotenoid pigments were isolated and identified as beta-carotene, gamma-carotene, rhodoxanthin and torularhodin. 2. For the first time rhodoxanthin was isolated as a fungal metabolite. 3. Linoleic acid was the major fatty acid in the total lipids. 4. Ergosterol was present in the lipids in the non-esterified form.

Project description:The halotolerant green alga Dunaliella tertiolecta accumulates starch and triacylglycerol (TAG) amounting to 70% and 10-15% of total cellular carbon, respectively, when exposed to nitrogen (N) deprivation. The purpose of this study was to clarify the inter-relationships between the biosynthesis of TAG, starch, and polar lipids (PLs) in this alga. Pulse labeling with [14C]bicarbonate was utilized to label starch and [14C]palmitic acid (PlA) to label lipids. Transfer of 14C into TAG was measured and used to calculate rates of synthesis. About two-thirds of the carbon in TAG originates from starch, and one-third is made de novo by direct CO2 assimilation. The level made from degradation of pre-formed PLs is estimated to be very small. Most of the de novo synthesis involves fatty acid transfer through PLs made during the first day of N deprivation. The results suggest that starch made by photosynthetic carbon assimilation at the early stages of N deprivation is utilized for synthesis of TAG. Trans-acylation from PLs is the second major contributor to TAG biosynthesis. The utilization of starch for TAG biosynthesis may have biotechnological applications to optimize TAG biosynthesis in algae.

High production of carotenoids by the green microalga Asterarcys quadricellulare PUMCC 5.1.1 under optimized culture conditions.

Project description:Since carotenoids are important as natural colorants, antioxidants, neutraceutics and pharmaceutics, the aim of the present study was to find a new good source of these pigments. We hereby report a green microalga Asterarcys quadricellulare PUMCC 5.1.1 as a new and good producer of carotenoids. The organism produced 35±1.75 ?g carotenoids mg-1 dry biomass during stationary phase in control cultures. The growth and carotenoids production by the test microalga were optimized by varying nutrient growth media, pH, nitrogen and phosphate source, salinity, light quality, intensity and duration. The optimized conditions for carotenoid production were: Bold basal (BB) medium with pH 8.5, containing with10 mM nitrate, 3.5 mM phosphate and 0.17 mM salinity and illuminated with blue light with 60 ?mol m-2 s-1 photon flux light intensity. Cultivation of cultures in the above mentioned optimized conditions resulted in nearly 3.0 fold increase in carotenoid production compared to the control cultures grown in unmodified BB medium. Using HPTLC, four carotenoids have been identified as ?-carotene, lutein, astaxanthin and canthaxanthin. Further, carotenoids were also separated and purified by flash chromatography and the amounts of purified carotenoids were determined by HPLC. The organism produced 47.0, 28.7, 15.5 and 14.0 ?g ?-carotene, lutein, astaxanthin and canthaxanthin mg-1 dry biomass, respectively, under optimized conditions. The amount of total carotenoids (118 ?g mg-1 dry biomass) produced by Asterarcys quadricellulare PUMCC 5.1.1 under optimized culture conditions was significantly higher than control cultures. Thus, this microalgal strain is a promising candidate for carotenoid production at commercial level.

Bioactivity assessment of exopolysaccharides produced by <i>Pleurotus pulmonarius</i> in submerged culture with different agro-waste residues.

Project description:Pleurotus spp. are white-rot fungi that utilize different agro-wastes to produce useful biologically active compounds. In this study, exopolysaccharides (EPS) were produced by Pleurotus pulmonarius in submerged culture supplemented with different agro-wastes. Functional groups in EPS were revealed using Fourier Transform-Infrared (FT-IR) spectroscopy. Antimicrobial activity of EPS was tested against microorganisms using agar well diffusion. Scavenging potentials of EPS was tested against 1, 1- diphenyl-2-picryhydrazyl (DPPH), hydroxyl (OH), iron (Fe2+) and nitric oxide (NO) radicals. In vitro prebiotic activity of EPS was carried out. The highest yield (5.60 g/L) of EPS was produced by P. pulmonarius in submerged culture supplemented with groundnut shell (20.0 g/L). The functional groups in EPS were hydroxyl (-OH), methyl (-CH3), ketone (-RCOH) and carbonyl group (-C=O). EPS displayed zones of inhibition (5.00-14.00 mm) against tested microorganisms. Scavenging activity of EPS ranged from 65.70-81.80% against DPPH. EPS supported the growth of Lactobacillus delbrueckii and Streptococcus thermophiles with values ranged from 3.04 × 104-3.40 × 104 cfu/ml and 2.50 × 104-2.81 × 104 cfu/ml, respectively. Submerged culture of P. pulmonarius with addition of agro-wastes enhanced yield of EPS. The EPS exhibited bio-functional properties like antimicrobial, antioxidant and prebiotic activities. Hence, agrowastes can be recycled in submerged fermentation with fungi to produce promising biomaterials for biopharmaceutical applications.

MicroRNAs in a multicellular green alga Volvox carteri

Project description:We identified 174 miRNAs expressed in Volvox carteri. Some of Volvox miRNAs are highly enriched in gonidia or somatic cells. Subsequently, we predicted the targets of Volvox miRNAs and found many of target genes were regulated through mRNA degradation. Conservation analysis suggests the common origin of miRNA between Volvox and Chlamydomonas and high frequency of birth and death of Volvox miRNAs.

Revisiting the isolation source after half a century: <i>Emericellopsis mirabilis</i> on a yellow-green alga.

Project description:Fungi-algae interactions, such as lichen-forming fungi and parasitic chytrids on phytoplankton, are common in ecosystems. In contrast, interactions between filamentous fungi and soil algae that can be observed with the naked eye have been given little attention and remain unexplored. Here, we report a fungus that was associated with a visible symptom of dead algae on a soil surface in Sugadaira-kogen, Nagano, central Japan. Acremonium-like conidiophores were growing on vesicles and dead bodies of a yellow-green alga, Botrydium granulatum. The fungus was identified as Emericellopsis mirabilis based on its morphology by microscopic observation, phylogenetic analysis, and the similarity of the isolation substrate with the first description of the species. Co-culture experiments showed a filamentous cell differentiation of the alga by the fungus, but no harmful or beneficial effects on algal growth. Therefore, we speculate that E. mirabilis is a facultative parasite of B. granulatum under natural conditions.

Identification of Salt Stress Responding Genes Using Transcriptome Analysis in Green Alga Chlamydomonas reinhardtii.

Project description:Salinity is one of the most important abiotic stresses threatening plant growth and agricultural productivity worldwide. In green alga Chlamydomonas reinhardtii, physiological evidence indicates that saline stress increases intracellular peroxide levels and inhibits photosynthetic-electron flow. However, understanding the genetic underpinnings of salt-responding traits in plantae remains a daunting challenge. In this study, the transcriptome analysis of short-term acclimation to salt stress (200 mM NaCl for 24 h) was performed in C. reinhardtii. A total of 10,635 unigenes were identified as being differently expressed by RNA-seq, including 5920 up- and 4715 down-regulated unigenes. A series of molecular cues were screened for salt stress response, including maintaining the lipid homeostasis by regulating phosphatidic acid, acetate being used as an alternative source of energy for solving impairment of photosynthesis, and enhancement of glycolysis metabolism to decrease the carbohydrate accumulation in cells. Our results may help understand the molecular and genetic underpinnings of salt stress responses in green alga C. reinhardtii.
