Arctigenin: a lignan from Arctium lappa.

ABSTRACT: The title compound {systematic name: (3R-trans)-4-[(3,4-dimethoxy-phen-yl)meth-yl]-3-[(4-hydr-oxy-3-methoxy-phen-yl)meth-yl]-4,5-dihydrofuran-2(3H)-one}, C(21)H(24)O(6), has a dibenz-yl-butyrolactone skeleton. The two aromatic rings are inclined at a dihedral angle of 68.75?(7)° with respect to each other. The lactone ring adopts an envelope conformation. A series of O-H?O and C-H?O hydrogen bonds contribute to the stabilization of the crystal packing. The absolute configuration was assigned on the basis of the published literature.

Project description:Four new lignan glycosides; urenalignosides A-D (1-4), along with 12 known ones (5-16) were isolated from Urena lobata. Their structures were determined on the basis of extensive spectroscopic and spectrometric data (1D and 2D NMR; IR; CD; and HRESIMS). Compounds 2-4; 6; 7; 10; and 11 showed inhibition of nitric oxide production in lipopolysaccharide-induced RAW 264.7 macrophage cells with IC50 values in the range of 25.5-98.4 μM (positive control; quercetin; IC50 = 7.2 ± 0.2 μM).

Schizandrol A: a lignan from Schisandra chinensis.

Project description:The title compound (systematic name1,2,3,10,11,12-hexa-meth-oxy-6,7-dimethyl-5,6,7,8-tetra-hydro-dibenzo[a,c]cyclo-oc-ten-6-ol), C(24)H(32)O(7), has a dibenzocyclo-octa-diene skeleton. There are three mol-ecules in the asymmetric unit, which are related by a pseudo-translation in the direction of the c axis. Nevertheless, the three mol-ecules differ in the torsion angle of one of the meth-oxy groups. The dihedral angles between the two aromatic rings are 62.39?(10), 62.65?(10) and 61.84?(10)° for the three mol-ecules. The crystal packing is stabilized by a series of O-H?O and C-H?O hydrogen bonds, as well as C-H?? inter-actions.

Potent cytotoxic arylnaphthalene lignan lactones from Phyllanthus poilanei.

Project description:Two new (1 and 2) and four known arylnaphthalene lignan lactones (3-6) were isolated from different plant parts of Phyllanthus poilanei collected in Vietnam, with two further known analogues (7 and 8) being prepared from phyllanthusmin C (4). The structures of the new compounds were determined by interpretation of their spectroscopic data and by chemical methods, and the structure of phyllanthusmin D (1) was confirmed by single-crystal X-ray diffraction analysis. Several of these arylnaphthalene lignan lactones were cytotoxic toward HT-29 human colon cancer cells, with compounds 1 and 7-O-[(2,3,4-tri-O-acetyl)-?-L-arabinopyranosyl)]diphyllin (7) found to be the most potent, exhibiting IC50 values of 170 and 110 nM, respectively. Compound 1 showed activity when tested in an in vivo hollow fiber assay using HT-29 cells implanted in immunodeficient NCr nu/nu mice. Mechanistic studies showed that this compound mediated its cytotoxic effects by inducing tumor cell apoptosis through activation of caspase-3, but it did not inhibit DNA topoisomerase II? activity.

Lignan Glucosides from the Stem Barks of Illicium difengpi.

Project description:In this study, four new lignan glucosides, named difengpiosides A-D (1-4), were isolated from the stem barks of Illicium difengpi, together with seven known compounds 5-11. Their structures were identified on the basis of spectroscopic analyses (1D and 2D NMR, HRESIMS, CD) and a comparison with literature data. All the compounds were evaluated for their inhibitory effects on lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW264.7 cells.

Kalshiolin A, new lignan from <i>Kalimeris shimadai</i>.

Project description:The Asian plant Kalimeris shimadai has been used as food and ethnologic medicine for over a thousand years. In this study, we isolated and identified one new lignan, kalshiolin A (1), and 12 known lignans (2-13). The structures were characterized by the comprehensive analyses of spectroscopic data (HR-ESI-MS, IR, 1D, and 2D-NMR) and the absolute configuration of 1 was determined from ECD calculations. The new compound 1 was also screened for cytotoxic activity but did not show significant potency (IC50 35.9-43.3 μM) against A549, MDA-MB-231, MCF7, KB, and KB-VIN cell lines.

(+)-Sesamin-oxidising CYP92B14 shapes specialised lignan metabolism in sesame.

Project description:Sesamum spp. (sesame) are known to accumulate a variety of lignans in a lineage-specific manner. In cultivated sesame (Sesamum indicum), (+)-sesamin, (+)-sesamolin and (+)-sesaminol triglucoside are the three major lignans found richly in the seeds. A recent study demonstrated that SiCYP92B14 is a pivotal enzyme that allocates the substrate (+)-sesamin to two products, (+)-sesamolin and (+)-sesaminol, through multiple reaction schemes including oxidative rearrangement of α-oxy-substituted aryl groups (ORA). In contrast, it remains unclear whether (+)-sesamin in wild sesame undergoes oxidation reactions as in S. indicum and how, if at all, the ratio of the co-products is tailored at the molecular level. Here, we functionally characterised SrCYP92B14 as a SiCYP92B14 orthologue from a wild sesame, Sesamum radiatum, in which we revealed accumulation of the (+)-sesaminol derivatives (+)-sesangolin and its novel structural isomer (+)-7´-episesantalin. Intriguingly, SrCYP92B14 predominantly produced (+)-sesaminol either through ORA or direct oxidation on the aromatic ring, while a relatively low but detectable level of (+)-sesamolin was produced. Amino acid substitution analysis suggested that residues in the putative distal helix and the neighbouring heme propionate of CYP92B14 affect the ratios of its co-products. These data collectively show that the bimodal oxidation mechanism of (+)-sesamin might be widespread across Sesamum spp., and that CYP92B14 is likely to be a key enzyme in shaping the ratio of (+)-sesaminol- and (+)-sesamolin-derived lignans from the biochemical and evolutionary perspectives.

Three new lignan glucosides from the roots of Scutellaria baicalensis.

Project description:Three new lignan glucosides, baicalensinosides A-C (1-3), were isolated from the roots of Scutellaria baicalensis. The structural elucidation was achieved by in-depth spectroscopic examinations and qualitative chemical test. Structurally, these compounds belong to the 3,4-dibenzyltetrahydrofuran-type lignan glycoside with a mono-hydroxyl substitution at the 7'-position of benzylidene group on the numbering system of lignans being one of their shared critical features. The anti-osteoporotic activity of the isolated compounds was assessed in an in vitro osteoprotegerin (OPG) transcriptional activity assay using dual luciferase reporter detection. At 10 μmol/L, compounds 1-3 increased the relative activating ratio of OPG transcription to 1.83, 0.84 and 0.98 times that of the control group, respectively.
