Project description:The development of cross-reactive sensor arrays for volatile organics (electronic noses, e-noses) is an active area of research. In this manuscript, we present a new format for barcoded polymer sensor arrays based on porous polymer beads. An array of nine self-encoded polymers was analyzed by Raman spectroscopy before and after exposure to a series of volatile organic compounds, and the changes in the vibrational fingerprints of their polymers was recorded before and after exposure. Our results show that the spectroscopic changes experienced by the porous spectroscopically encoded beads after exposure to an analyte can be used to identify and classify the target analytes. To expedite this analysis, analyte-specific changes induced in the sensor arrays were transformed into a response pattern using multivariate data analysis. These studies established the barcoded bead array format as a potentially effective sensing element in e-nose devices. Devices such as these have the potential to advance personalized medicine, providing a platform for non-invasive, real-time volatile metabolite detection.

Project description:Earthquakes are lethal natural disasters frequently burying people alive under collapsed buildings. Tracking entrapped humans from their unique volatile chemical signature with hand-held devices would accelerate urban search and rescue (USaR) efforts. Here, a pilot study is presented with compact and orthogonal sensor arrays to detect the breath- and skin-emitted metabolic tracers acetone, ammonia, isoprene, CO2, and relative humidity (RH), all together serving as sign of life. It consists of three nanostructured metal-oxide sensors (Si-doped WO3, Si-doped MoO3, and Ti-doped ZnO), each specifically tailored at the nanoscale for highly sensitive and selective tracer detection along with commercial CO2 and humidity sensors. When tested on humans enclosed in plethysmography chambers to simulate entrapment, this sensor array rapidly detected sub-ppm acetone, ammonia, and isoprene concentrations with high accuracies (19, 21, and 3 ppb, respectively) and precision, unprecedented by portable sensors but required for USaR. These results were in good agreement (Pearson's correlation coefficients ?0.9) with benchtop selective reagent ionization time-of-flight mass spectrometry (SRI-TOF-MS). As a result, an inexpensive sensor array is presented that can be integrated readily into hand-held or even drone-carried detectors for first responders to rapidly screen affected terrain.

Project description:Living systems employ cilia to control and to sense the flow of fluids for many purposes, such as pumping, locomotion, feeding, and tissue morphogenesis. Beyond their use in biology, functional arrays of artificial cilia have been envisaged as a potential biomimetic strategy for inducing fluid flow and mixing in lab-on-a-chip devices. Here we report on fluid transport produced by magnetically actuated arrays of biomimetic cilia whose size approaches that of their biological counterparts, a scale at which advection and diffusion compete to determine mass transport. Our biomimetic cilia recreate the beat shape of embryonic nodal cilia, simultaneously generating two sharply segregated regimes of fluid flow: Above the cilia tips their motion causes directed, long-range fluid transport, whereas below the tips we show that the cilia beat generates an enhanced diffusivity capable of producing increased mixing rates. These two distinct types of flow occur simultaneously and are separated in space by less than 5 microm, approximately 20% of the biomimetic cilium length. While this suggests that our system may have applications as a versatile microfluidics device, we also focus on the biological implications of our findings. Our statistical analysis of particle transport identifying an enhanced diffusion regime provides novel evidence for the existence of mixing in ciliated systems, and we demonstrate that the directed transport regime is Poiseuille-Couette flow, the first analytical model consistent with biological measurements of fluid flow in the embryonic node.

Project description:Due to the sophisticated hierarchical structure and limited reparability of articular cartilage (AC), the ideal regeneration of AC defects has been a major challenge in the field of regenerative medicine. As defects progress, they often extend from the cartilage layer to the subchondral bone and ultimately lead to osteoarthritis. Tissue engineering techniques bring new hope for AC regeneration. To meet the regenerative requirements of the heterogeneous and layered structure of native AC tissue, a substantial number of multilayered biomimetic scaffolds have been studied. Ideal multilayered scaffolds should generate zone-specific functional tissue similar to native AC tissue. This review focuses on the current status of multilayered scaffolds developed for AC defect repair, including design strategies based on the degree of defect severity and the zone-specific characteristics of AC tissue, the selection and composition of biomaterials, and techniques for design and manufacturing. The challenges and future perspectives of biomimetic multilayered scaffold strategies for AC regeneration are also discussed.

Project description:Biomimetic flexible tactile sensors endow prosthetics with the ability to manipulate objects, similar to human hands. However, it is still a great challenge to selectively respond to static and sliding friction forces, which is crucial tactile information relevant to the perception of weight and slippage during grasps. Here, inspired by the structure of fingerprints and the selective response of Ruffini endings to friction forces, we developed a biomimetic flexible capacitive sensor to selectively detect static and sliding friction forces. The sensor is designed as a novel plane-parallel capacitor, in which silver nanowire-3D polydimethylsiloxane (PDMS) electrodes are placed in a spiral configuration and set perpendicular to the substrate. Silver nanowires are uniformly distributed on the surfaces of 3D polydimethylsiloxane microcolumns, and silicon rubber (Ecoflex®) acts as the dielectric material. The capacitance of the sensor remains nearly constant under different applied normal forces but increases with the static friction force and decreases when sliding occurs. Furthermore, aiming at the slippage perception of neuroprosthetics, a custom-designed signal encoding circuit was designed to transform the capacitance signal into a bionic pulsed signal modulated by the applied sliding friction force. Test results demonstrate the great potential of the novel biomimetic flexible sensors with directional and dynamic sensitivity of haptic force for smart neuroprosthetics.

Project description:The combination of nanotechnology, biology, and bioengineering greatly improved the developments of nanomaterials with unique functions and properties. Biomolecules as the nanoscale building blocks play very important roles for the final formation of functional nanostructures. Many kinds of novel nanostructures have been created by using the bioinspired self-assembly and subsequent binding with various nanoparticles. In this review, we summarized the studies on the fabrications and sensor applications of biomimetic nanostructures. The strategies for creating different bottom-up nanostructures by using biomolecules like DNA, protein, peptide, and virus, as well as microorganisms like bacteria and plant leaf are introduced. In addition, the potential applications of the synthesized biomimetic nanostructures for colorimetry, fluorescence, surface plasmon resonance, surface-enhanced Raman scattering, electrical resistance, electrochemistry, and quartz crystal microbalance sensors are presented. This review will promote the understanding of relationships between biomolecules/microorganisms and functional nanomaterials in one way, and in another way it will guide the design and synthesis of biomimetic nanomaterials with unique properties in the future.

Project description:We outline an evolution process for tongue elements composed of poly(p-aryleneethynylene)s (PAE) and detergents, resulting in a chemical tongue (24 elements) that discerns antibiotics. Cross-breeding of this new tongue with tongue elements that consist of simple poly(p-phenyleneethynylene)s (PPE) at different pH-values leads to an enlarged sensor array, composed of 30 elements. This tongue was pruned, employing principal component analysis. We find that a filial tongue featuring three elements from each original array (i.e. a six element tongue) is superior to either of the prior tongues and the composite tongue in the discrimination of structurally different antibiotics. Such a selection process should be general and give an idea how to successfully generate powerful low-selectivity sensor elements and configure them into discriminative chemical tongues.

Project description:Electronic visual prostheses, or biomimetic eyes, have shown the feasibility of restoring functional vision in the blind through electrical pulses to initiate neural responses artificially. However, existing visual prostheses predominantly use wired connections or electromagnetic waves for powering and data telemetry, which raises safety concerns or couples inefficiently to miniaturized implant units. Here, we present a flexible ultrasound-induced retinal stimulating piezo-array that can offer an alternative wireless artificial retinal prosthesis approach for evoking visual percepts in blind individuals. The device integrates a two-dimensional piezo-array with 32-pixel stimulating electrodes in a flexible printed circuit board. Each piezo-element can be ultrasonically and individually activated, thus, spatially reconfigurable electronic patterns can be dynamically applied via programmable ultrasound beamlines. As a proof of concept, we demonstrate the ultrasound-induced pattern reconstruction in ex vivo murine retinal tissue, showing the potential of this approach to restore functional, life-enhancing vision in people living with blindness.

Project description:We address the problem of feature selection for classifying a diverse set of chemicals using an array of metal oxide sensors. Our aim is to evaluate a filter approach to feature selection with reference to previous work, which used a wrapper approach on the same data set, and established best features and upper bounds on classification performance. We selected feature sets that exhibit the maximal mutual information with the identity of the chemicals. The selected features closely match those found to perform well in the previous study using a wrapper approach to conduct an exhaustive search of all permitted feature combinations. By comparing the classification performance of support vector machines (using features selected by mutual information) with the performance observed in the previous study, we found that while our approach does not always give the maximum possible classification performance, it always selects features that achieve classification performance approaching the optimum obtained by exhaustive search. We performed further classification using the selected feature set with some common classifiers and found that, for the selected features, Bayesian Networks gave the best performance. Finally, we compared the observed classification performances with the performance of classifiers using randomly selected features. We found that the selected features consistently outperformed randomly selected features for all tested classifiers. The mutual information filter approach is therefore a computationally efficient method for selecting near optimal features for chemical sensor arrays.
