Project description:BackgroundQuantitative relations between weather variables and malaria vector can enable pro-active control through meteorological monitoring. Such relations are also critical for reliable projections in a changing climate, especially since the vector abundance depends on a combination of weather variables, each in a given range. Further, such models need to be region-specific as vector population and exposure depend on regional characteristics.MethodsWe consider days of genesis based on daily temperature, rainfall and humidity in given ranges. We define a single model parameter based on estimates of exposure and transmission to calibrate the model; the model is applied to 12 districts of Arunachal Pradesh, a region endemic to malaria. The epidemiological data is taken as blood samples that test positive. The meteorological data is adopted from NCEP daily Reanalysis on a global grid; population data is used to estimate exposure and transmission coefficients.ResultsThe observed annual cycles (2006-2010) and the interannual variability (2002-2010) of epidemiology are well simulated for each of the 12 districts by the model. While no single weather variable like temperature can reproduce the observed epidemiology, a combination of temperature, rainfall and humidity provides an accurate description of the annual cycle as well as the inter annual variability over all the 12 districts.ConclusionInclusion of the three meteorological variables, along with the expressions for exposure and transmission, can quite accurately represent observed epidemiology over multiple locations and different years. The model is potentially useful for outbreak forecasts at short time scales through high resolution weather monitoring; however, validation with longer and independent epidemiological data is required for more robust estimation of realizable skill. While the model has been examined over a specific region, the basic algorithm is easily applicable to other regions; the model can account for shifting vulnerability due to regional climate change.

Project description:Recent studies in experimental mice have shown that mild deficiency of methylenetetrahydrofolate reductase (MTHFR) enzyme confers protection against malaria, thus providing an important basis for the hypothesis that MTHFR polymorphism, i.e. C677T, might have been subjected to selection pressure against malaria. The present study was undertaken in a malaria endemic region in North East India to assess whether a similar selection advantage exists for other genes in folate metabolism pathway.A total of 401 subjects including 131 symptomatic malaria, 97 asymptomatic malaria and 173 normal healthy controls were analysed for nine polymorphisms (single-nucleotide polymorphisms [SNPs] in eight genes and insertion/deletion in one gene): MTHFR C677T, methionine synthase reductase (MTRR) A66G, glutamate carboxypeptidase II (GCPII) C1561T, cystathionine beta-synthase (CBS) 844ins68, reduced folate carrier-1 (RFC-1) G80A, serine hydroxymethyltransferase (SHMT) C1420T, methionine synthase (MTR) A2756G, MTHFR G1793A (D 919G), glycine N-methyltransferase (GNMT) 1289 by PCR-RFLP technique. Differences in frequencies of genotype distribution of each polymorphic marker between these groups were evaluated.MTRR A2756G, SHMT C1420T, GCPII C1561T, MTRR A2756G and GNMT C1289T and RFC1 G80A polymorphisms showed significantly different prevalence between different groups analyzed. No significant differences were seen in the distribution of other polymorphisms.The study gives a clue for the possible selection of specific polymorphisms in the genes involved in the folate metabolism pathway by malaria parasite.

Project description:BACKGROUND:Anopheles baimaii is a primary vector of human malaria in the forest settings of Southeast Asia including the north-eastern region of India. Here, the genetic population structure and the basic population genetic parameters of An. baimaii in north-east India were estimated using DNA sequences of the mitochondrial cytochrome oxidase sub unit II (COII) gene. METHODS:Anopheles baimaii were collected from 26 geo-referenced locations across the seven north-east Indian states and the COII gene was sequenced from 176 individuals across these sites. Fifty-seven COII sequences of An. baimaii from six locations in Bangladesh, Myanmar and Thailand from a previous study were added to this dataset. Altogether, 233 sequences were grouped into eight population groups, to facilitate analyses of genetic diversity, population structure and population history. RESULTS:A star-shaped median joining haplotype network, unimodal mismatch distribution and significantly negative neutrality tests indicated population expansion in An. baimaii with the start of expansion estimated to be ~0.243 million years before present (MYBP) in north-east India. The populations of An. baimaii from north-east India had the highest haplotype and nucleotide diversity with all other populations having a subset of this diversity, likely as the result of range expansion from north-east India. The north-east Indian populations were genetically distinct from those in Bangladesh, Myanmar and Thailand, indicating that mountains, such as the Arakan mountain range between north-east India and Myanmar, are a significant barrier to gene flow. Within north-east India, there was no genetic differentiation among populations with the exception of the Central 2 population in the Barail hills area that was significantly differentiated from other populations. CONCLUSIONS:The high genetic distinctiveness of the Central 2 population in the Barail hills area of the north-east India should be confirmed and its epidemiological significance further investigated. The lack of genetic population structure in the other north-east Indian populations likely reflects large population sizes of An. baimaii that, historically, were able to disperse through continuous forest habitats in the north-east India. Additional markers and analytical approaches are required to determine if recent deforestation is now preventing ongoing gene flow. Until such information is acquired, An. baimaii in north-east India should be treated as a single unit for the implementation of vector control measures.

Project description:BackgroundThe western districts of Mizoram (Lunglei, Mamit, and Lawngtlai) are malaria hotspots. Understanding the knowledge, attitude, and practices of the tribal communities in Mizoram's western districts will aid the development of specific interventions.MethodsAn explanatory sequential mixed-method study was conducted from April to November 2023 in the Lunglei district. In a community-based cross-sectional survey of 353 participants, the knowledge, attitude, practices, and care-seeking behaviour toward malaria were assessed using a semi-structured questionnaire. Data was analysed using SPSS version 29 software; univariate variables were presented in percentage, and bivariate and multivariate variables were analysed using the chi-square test and logistic regression, respectively. This was followed by in-depth telephonic interviews of twelve participants, and the data was analysed using NVivo.ResultsOut of the 353 respondents, 77.9%, 82.7%, 55.5%, and 63.2% of the participants had good knowledge, attitude, practices, and care-seeking behaviour, respectively. The in-depth qualitative interviews highlighted the villagers' good knowledge of the various aspects of malaria transmission, treatment, and prevention practices (indoor residual spraying and use of insecticide-treated nets).ConclusionHigh disease endemicity, awareness programmes and vector control interventions might be contributing to the overall good knowledge, attitude, and practices toward malaria among the villagers. In addition to vector control measures, active parasite surveillance is key to malaria control in this region.

Project description:Microrefugia at high altitudes or high latitudes are thought to play an important role in the post-glacial colonization of species. However, how populations in such microrefugia have responded to climate changes in alternating cold glacial and warm interglacial stages remain unclear. Here we present evidence to indicate the Rongbuk Valley of the Mt. Qomolangma (Mt. Everest) area, the highest region on earth, had microrefugia for Hippophae tibetana and discuss how this low shrub was adapted to the extreme climate fluctuations of the last 25,000 years by shifts. By integrating geological, glaciological, meteorological, and genetic information, we found that the Rongbuk Valley was not only a glacial microrefugium but also an interglacial microrefugium for H. tibetana: the former was located on the riverbank below 4800 m above sea level (asl) or lower area and the latter at ? 5000 m asl. Our results show that after the Last Glacial Maximum (LGM), H. tibetana in the valley has undergone upward and downward migrations around ? 5000 m driven by climate fluctuations and the population in the glacial microrefugium has suffered extinction or extreme contraction. Moreover, with the rise of temperature in the last four decades, the upper limit of H. tibetana has shifted at least 30 m upward. Combining population history and recent range shift of this species is important in predicting the fate of this endemic species to future climate changes.

Project description:Background: The European North-East of Russia is the territory which includes the Nenets Autonomous District, represented by the East European tundra (from Kanin Peninsula to Vaigach Island), Komi Republic with its taiga ecosystems and the Urals (Northern, SubPolar and Polar). Over 20 years of systematic studies of soil fauna in the studied region has resulted in a huge amount of data being accumulated that can be analysed from different positions. Considering that the representation of Russian soil biota data, especially from European North-East of Russia in the GBIF database is not large, our data are of great interest to the scientific world community. The accumulation of such data will solve questions on national and global scales using large arrays.This study produced a dataset containing information on occurrences on soil invertebrates (Lumbricidae, Chilopoda, Diplopda, Collembola, Elateridae and Staphylinidae) in the European North-East of Russia. The dataset summarises occurrences noted in natural and disturbed forests, tundra and mountain ecosystems.New information: Data from 196 geo-referenced localities of European North-East of Russia (tundra, taiga and mountains ecosystems) have been collated. A total of 5412 occurrences are included in the resource. The current project surveys 13 species of earthworms, 20 species of millipedes, 246 species of springtails, 446 species of rove beetles and 60 species of click beetles. The diversity of soil invertebrates in the European North-East of Russia has not been fully explored and further exploration will lead to more taxa.

Project description:Purpose:The Middle East and North Africa (MENA) region registers some of the lowest serum 25?hydroxyvitamin D [25(OH)D] concentrations, worldwide. We describe the prevalence and the risk factors for hypovitaminosis D, completed and ongoing clinical trials, and available guidelines for vitamin D supplementation in this region. Methods:This review is an update of previous reviews published by our group in 2013 for observational studies, and in 2015 for randomized controlled trials (RCTs) from the region. We conducted a comprehensive search in Medline, PubMed, and Embase, and the Cochrane Library, using MeSH terms and keywords relevant to vitamin D, vitamin D deficiency, and the MENA region, for the period 2012-2017 for observational studies, and 2015-2017 for RCTs. We included large cross-sectional studies with at least 100 subjects/study, and RCTs with at least 50 participants per arm. Results:We identified 41 observational studies. The prevalence of hypovitaminosis D, defined as a 25?hydroxyvitamin D [25(OH)D] level below the desirable level of 20?ng/ml, ranged between 12-96% in children and adolescents, and 54-90% in pregnant women. In adults, it ranged between 44 and 96%, and the mean 25(OH)D varied between 11 and 20?ng/ml. In general, significant predictors of low 25(OH)D levels were female gender, increasing age and body mass index, veiling, winter season, use of sun screens, lower socioeconomic status, and higher latitude.We retrieved 14 RCTs comparing supplementation to control or placebo, published during the period 2015-2017: 2 in children, 8 in adults, and 4 in pregnant women. In children and adolescents, a vitamin D dose of 1000-2000?IU/d was needed to maintain serum 25(OH)D level at target. In adults and pregnant women, the increment in 25(OH)D level was inversely proportional to the dose, ranging between 0.9 and 3?ng/ml per 100?IU/d for doses ?2000?IU/d, and between 0.1 and 0.6?ng/ml per 100?IU/d for doses ?3000?IU/d. While the effect of vitamin D supplementation on glycemic indices is still controversial in adults, vitamin D supplementation may be protective against gestational diabetes mellitus in pregnant women. In the only identified study in the elderly, there was no significant difference between 600?IU/day and 3750?IU/day doses on bone mineral density. We did not identify any fracture studies.The available vitamin D guidelines in the region are based on expert opinion, with recommended doses between 400 and 2000?IU/d, depending on the age category, and country. Conclusion:Hypovitaminosis D is prevalent in the MENA region, and doses of 1000-2000?IU/d may be necessary to reach a desirable 25(OH)D level of 20?ng/ml. Studies assessing the effect of such doses of vitamin D on major outcomes, and confirming their long term safety, are needed.
