Project description:The incorporation of functionalised carbon nanodots within a novel low molecular weight salt hydrogel derived from 5-aminosalicylic acid is reported. The carbon dots result in markedly enhanced gelation properties, while inclusion within the hydrophobic gel results in a dramatic fluorescence enhancement for the carbon nanomaterials. The resulting hybrid CD gels exhibit a useful sensor response for heavy metal ions, particularly Pb2+.

Project description:Fluorescent carbon dots have received considerable attention as a result of their accessibility and potential applications. Although several prior studies have demonstrated that nearly any organic compound can be converted into carbon dots by chemical carbonization processes, mechanisms explaining the formation of carbon dots still remain unclear. Herein, we propose a seed-growth mechanism of carbon dot formation facilitated by ferulic acid, a widespread and naturally occurring phenolic compound in the seeds of Ocimum basilicum (basil). Ferulic acid triggers the local condensation of polysaccharide chains and forms catalytic core regions resulting in nanoscale carbonization. Our study indicates that carbon dots generated from natural sources might share the similar mechanism of phenolic compound mediated nanoscale condensation followed by core carbonization.

Project description:Contamination of groundwater with radioactive substances comprising actinides and lanthanides is a significant environmental hazard and thus the development of selective, sensitive, and easy-to-apply sensors for water-soluble actinide and lanthanide ions is highly sought. We constructed a new selective fluorescent sensor for UO2 2+, Sm3+, and Eu3+ based on a carbon dot (C-dot)-aerogel hybrid prepared through in situ carbonization of 2-thenoyltrifluoroacetone (TTA), a high-affinity heavy metal chelator. The TTA-C-dot-aerogel enabled the detection of UO2 2+ ions, which induced a significant red fluorescence shift, whereas Eu3+ and particularly Sm3+ ions gave rise to pronounced fluorescence quenching. Importantly, the lanthanide/actinide ion-selective TTA-C-dots could be synthesized only in situ inside the aerogel pores, indicating the crucial role of the aerogel host matrix both in enabling the formation of the C-dots and in promoting the adsorption and interactions of the lanthanide and actinide metal ions with the embedded C-dots.

Project description:This paper presents a simple and highly efficient method for dopamine detection using water-soluble carbon dot nanoparticles. The ZnO-associated carbon dots (CDZs) were synthesized using a green chemical strategy. An examination of the effects of biomolecules on the fluorescence of CDZs revealed selective dopamine-induced quenching. In a phosphate buffer (pH = 7.4) medium, a detection limit of 1.06 nM was obtained. This "turn off" phenomenon was attributed to the electronic interaction between CDZs and dopamine, during the oxidation of dopamine. At lower pH, however, the effects of dopamine on the fluorescence of CDZs were insignificant as the oxidation of dopamine was hindered when the proton concentration was increased. This method was found to be free from the interference of coexisting molecules, that is, ascorbic acid and uric acid. This sensing platform was applied successfully in biological fluids to confirm the practical significance of the as-designed sensor.

Project description:Biotemplated nanomaterials offer versatile functionality for multimodal imaging, biosensing, and drug delivery. There remains an unmet need for traceable and biocompatible nanomaterials that can be synthesized in a precisely controllable manner. Here, we report self-assembled quantum dot DNA hydrogels that exhibit both size and spectral tunability. We successfully incorporate DNA-templated quantum dots with high quantum yield, long-term photostability, and low cytotoxicity into a hydrogel network in a single step. By leveraging DNA-guided interactions, we introduce multifunctionality for a variety of applications, including enzyme-responsive drug delivery and cell-specific targeting. We report that quantum dot DNA hydrogels can be used for delivery of doxorubicin, an anticancer drug, to increase potency 9-fold against cancer cells. This approach also demonstrated high biocompatibility, trackability, and in vivo therapeutic efficacy in mice bearing xenografted breast cancer tumors. This work paves the way for the development of new tunable biotemplated nanomaterials with multiple synergistic functionalities for biomedical applications.The development of nanomaterials for imaging and drug delivery has been of great interest to the field. Here, the authors synthesized multifunctional enzyme-responsive hydrogels with self-assembling quantum dots for nucleic acid and drug delivery as well as having imaging capability.

Project description:Multifunctional nanocarrier-based theranostics is supposed to overcome some key problems in cancer treatment. In this work, a novel method for the preparation of a fluorescent mesoporous silica-carbon dot nanohybrid was developed. Carbon dots (CDs), from folic acid as the raw material, were prepared in situ and anchored on the surface of amino-modified mesoporous silica nanoparticles (MSNs-NH2) via a microwave-assisted solvothermal reaction. The as-prepared nanohybrid (designated MSNs-CDs) not only exhibited strong and stable yellow emission but also preserved the unique features of MSNs (e.g., mesoporous structure, large specific surface area, and good biocompatibility), demonstrating a potential capability for fluorescence imaging-guided drug delivery. More interestingly, the MSNs-CDs nanohybrid was able to selectively target folate receptor-overexpressing cancer cells (e.g., HeLa), indicating that folic acid still retained its function even after undergoing the solvothermal reaction. Benefited by these excellent properties, the fluorescent MSNs-CDs nanohybrid can be employed as a fluorescence-guided nanocarrier for the targeted delivery of anticancer drugs (e.g., doxorubicin), thereby enhancing chemotherapeutic efficacy and reducing side effects. Our studies may provide a facile strategy for the fabrication of multifunctional MSN-based theranostic platforms, which is beneficial in the diagnosis and therapy of cancers in future.

Project description:Fluorescent hydrogels (FH) have a variety of potential applications in the field of soft electronics. However, fabrication of mechanically stable and printable fluorescent hydrogels remains challenging. Here, we report a kind of fluorescent hydrogel based on the co-assembly of peptide motif and transition metal ions. The metal ions are captured in the hydrogel network at specific positions through covalently linked ligands on the peptide hydrogelators. This efficiently prevents the aggregation and self-quenching of organometallic chromophores. In addition, the formation of metal-ligand complexes introduces additional interactions to stabilize the hydrogel network, making the FH even more stable after the incorporation of metal ions. The FH is optically transparent but highly fluorescent. By using three different metal ions, the white light fluorescent supramolecular hydrogel has been achieved. As a proof-of-principle, we demonstrate the printability of the hydrogels to various patterns. We anticipate that with the improved fluorescent performance and stability, this kind of FH can find broad applications in extrusion-based 3D printing for the construction of soft electronics.

Project description:Nanometer-sized fragments of carbon in the form of multilayer graphene ("carbon dots") have been under highly active study for applications in imaging. While offering advantages of low toxicity and photostability, such nanomaterials are inhomogeneous and have limited wavelengths of emission. Here we address these issues by assembling luminescent aromatic C16-C38 hydrocarbons together on a DNA scaffold in homogeneous, soluble molecular compounds. Monomer deoxyribosides of five different aromatic hydrocarbons were synthesized and assembled into a library of 1296 different tetramer compounds on PEG-polystyrene beads. These were screened for photostability and a range of emission colors using 365 nm excitation, observing visible light (>400 nm) emission. We identified a set of six oligomers (DNA-carbon assemblies, DNA-CAs) with exceptional photostability that emit from 400 to 680 nm in water, with Stokes shifts of up to 110 nm, quantum yields ranging from 0.01 to 0.29, and fluorescence lifetimes from 3 to 42 ns. In addition, several of these DNA-CAs exhibited white emission in aqueous solution. The molecules were used in multispectral cell imaging experiments and were taken up into cells passively. The results expand the range of emission properties that can be achieved in water with all-hydrocarbon chromophores and establish the use of the DNA scaffold to arrange carbon layers in homogeneous, rapidly synthesized assemblies.

Project description:We report very fast, green, and large-scale synthesis of amino-functionalized carbon quantum dots (CQDs) using a domestic microwave to investigate CQD-Tb-based dual emission for visual detection of toxic Hg2+. Citric acid and p-phenylenediamine are used as precursor materials to synthesize the CQD, which shows excitation-independent blue luminescence. To achieve the dual emission, Tb-containing CQD is synthesized in a very easy and cost-effective way. These dual-emissive fluorescent materials have been successfully used as a fluorescent indicator for visual detection of toxic Hg2+ metal ions. An instant color change from blue to green in the presence of a very low amount of Hg2+ under a UV lamp (?365nm) is observed. The material is highly sensitive and selective toward detection of mercury ions in the presence of other metal ions. The photoluminescence quenching mechanism (photoinduced electron transfer process) has been explained using an electronic band diagram supported by zeta-potential and time-correlated single photon counting measurements.

Project description:Targeting visually identified neurons for electrophysiological recording is a fundamental neuroscience technique; however, its potential is hampered by poor visualization of pipette tips in deep brain tissue. We describe quantum dot-coated glass pipettes that provide strong two-photon contrast at deeper penetration depths than those achievable with current methods. We demonstrated the pipettes' utility in targeted patch-clamp recording experiments and single-cell electroporation of identified rat and mouse neurons in vitro and in vivo.
