ABSTRACT: The primary objective is to estimate the effect of the human homolog of the Kirsten rat sarcoma-2 virus oncogene (KRAS) mutation status (wild type versus mutant) from tumor tissue on efficacy endpoints in patients with metastatic colorectal cancer (mCRC) receiving second-line chemotherapy with panitumumab after failing first-line treatment.

Expression data from tumors generated from genome-engineered mice

Project description:Kirsten rat sarcoma viral oncogene homolog (KRAS) mutations are found in 30–40% of human colorectal cancers (CRC). However, specific therapeutics against KRAS-mutated CRC have not been established. We have previously reported mouse models for colon cancer with and without Kras mutations (CDX2P-G22Cre;Apcflox/flox; KrasG12D and CDX2P-G22Cre;Apcflox/flox mice, respectively). Herein, we aimed to identify candidate genes as novel therapeutic targets or biomarkers for KRAS-mutated CRC by comparing the gene expression profiles of these two mouse models.

Primary endpoints: Progression Free Survival (PFS)PFS time is defined as the time from the date of randomization to the first observation of disease progression (as classified by modified RECIST based on the investigator assessment of radiographic scans, or clinical progression whichever occurs first,) or death due to any cause. If a subject’s disease has not progressed and the subject is alive by the analysis data cut-off date, PFS will be censored at the last date they are known to be progression-free (ie, the last evaluable disease assessment date).

Triple SILAC exosomal quantitative proteomic analysis

Project description:Exosomes are 30-100 nm sized membrane vesicles released by cells into extracellular space and mediate the intercellular communication via transfer of proteins and RNAs. To better understand the function of these microvesicles in lung carcinogenesis, we employed a Triple SILAC quantitative proteomic strategy to examine the differential protein abundance between exosomes derived from an immortalized normal bronchial epithelial cell line and two non-small cell lung cancer (NSCLC) cell lines harboring distinct activating mutations in the cell signaling molecules Kirsten rat sarcoma viral oncogene homolog (KRAS) or epidermal growth factor receptor (EGFR). We were able to quantify 727 exosomal proteins derived from the three cell lines. Proteins associated with signal transduction are enriched in NSCLC exosomes which are functionally active in regulating cell proliferation. The study is the first to investigate protein abundance differences in exosomes derived from NSCLC cells and their normal counterparts, and reveals the role of exosomes in NSCLC cancer progression, which may have clinical implications in biomarker development for patients with NSCLC.
