Trial on Innovative Technologies in Colonoscopy

ABSTRACT: The trial will compare results of screening colonoscopy performed by means of conventional colonoscopy and using new visualisation techniques during endoscopic examination. * Electronic colonoscopes Olympus CF-HQ190F with following options: magnetic positioning (Scope Guide), responsive insertion technology (RIT), dual focus function, narrow band imaging (NBI) will be used for innovative colonoscopies * Electronic colonoscopes Olympus CF-H180DL with Scope Guide and NBI options will be used for conventional colonoscopies Endoscopists will archive all images and establish presumptive diagnosis based on the results of different visualisation techniques. All endoscopes will be attached to Olympus Evis Exera III system. Biopsy of all pathological lesions will be performed to establish final diagnosis. The main outcome measure is diagnostic accuracy of innovative colonoscopy in comparison with conventional technique.

Project description:The purpose of this study was to provide molecular evidence that surgical margin definition using narrow band imaging (NBI) resulted in more complete OSCC excision that conventional while light (WL) imaging. Samples were taken prospectively from 18 patients undergoing surgical resection for intraoral SCC (excluding lip, pharynx and hypopharynx). Biopsies were taken from the tumour and from 5mm beyond each of the abnormal tissue margins defined by WL and NBI. Transcriptional profiling was performed by microarray.

Colonoscope Insertion Trial Protocol

Project description:This research study is about determining how well a Technically Improved Colonoscope from Olympus works in helping the doctor in guiding the scope through the large bowel or colon. When a doctor performs a colonoscopy he guides or inserts the scope from the rectum to the appendix where the small bowel ends and the large bowel begins. There are points during this insertion where there are turns in the colon which can create a loop in the colonoscope. These loops can stretch the colon and create some discomfort and also require special maneuvers by the nurse or patient to help reduce or minimize the loop. Thus, reducing the looping will make it more comfortable for the patient and more efficient for screening for polyps. The purpose of the study is to examine this colonoscope and compare it to the traditional adult colonoscope (Olympus CF-H180). The investigators hypothesis is that this colonoscope will function better with regard to insertion of the scope from the rectum to the cecum.

Extraction of Entamoeba histolytica membrane proteins using three extraction methods

Project description:Entamoeba histolytica membrane proteins are important players in the parasite’s pathogenicity. However, most of the proteins have not been identified. This study reports the membrane proteins extracted using three extractions methods: two commercial kits (ProteoExtract® from Calbiochem and ProteoPrep® from Sigma), and a conventional laboratory method. The resulting membrane fractions (MF) and cytosolic fractions (CF)were analysed using LC-ESI-MS/MS. The proteins identified in at least two out of three biological replicates revealed a total of 490, 492, and 587 MF proteins extracted using the ProteoExtract® kit, ProteoPrep® kit and conventional method, respectively. Meanwhile, 487, 611 and 343 proteins were identified in the CF extracted using the ProteoExtract® kit, ProteoPrep® kit and conventional method, respectively. Analysis of the identified MF and CF proteins extracted by the respective extraction kits suggests that the ProteoPrep® extraction kit was the most selective in separating MF and CF among the three extraction methods.

A scalable multiplexed platform to accelerate protein interactome discovery

Project description:Chromatographic fractionation-mass spectrometry (CF-MS) allows for mapping native protein-protein interactions and macromolecular complexes on a proteome-scale but is tedious and resource intensive for comparative analysis. Here, we present a more parsimonious and reproducible multiplexing CF-MS platform for measuring multi-protein assemblies across different experimental samples simultaneously with an order-of-magnitude faster than previous approaches. We used this innovative strategy to elucidate protein networks in control and breast cancer cells models in parallel within two weeks of total MS instrument time.

Comparative Analysis of Neonatal versus Adult Brain in Unilateral Brain Injury Mice Model Revealing Genes Involved in High Compensatory Motor Functional Recovery Mechanism

Project description:Previously, while studying the development, especially of corticospinal neurons, it was concluded that the main compensatory mechanism after unilateral brain injury in rat at neonatal stage was due to in part by nonlesioned ipsilateral corticospinal neurons that escaped selection by axonal elimination or neuronal apoptosis (Yoshikawa et al., 2011, Dev Neuroscience). However, above result suggesting compensatory mechanism in neonate brain could not be correlated with high functional recovery. Therefore, we asked - what is the difference among neonate and adult in context of functional recovery and mechanism/s therein, especially focused on the corticospinal neurons? Toward this goal, in the present study, we have utilized a unilateral brain injury mice model and compared motor functional recovery mechanism post-neonatal brain injury (hereafter termed, NBI) with adult brain injury (hereafter termed, ABI). Three analyses were perfomed: i) quantitative behavioral analysis of the forelimb movements using the ladder walking test, where the corticospinal system has been suggested to play an important role ii) a neuroanatomical retrogradely tracing analysis of unlesioned side corticospinal neurons; and iii) differentially global gene expressions profiling in the unlesioned-side neocortex (rostral from bregma) in NBI and ABI using a high-throughput DNA microarray approach on a 8x60 K whole genome Agilent DNA chip using a two-color (Cy3/Cy5) dye-swap approach. Behavioral data confirmed the higher recovery ability in NBI over ABI is related to non-lesional frontal neocoetex including rostral caudal forelimb area. With a overall aim to unravel the underlying molecular events, in the present study a first inventory of the differential expressed gene expressions genome-wide in the NBI and ABI model mice has been provided.

In-depth proteomic investigation of isogenic radiation resistant prostate cancer cell lines

Project description:To model radioresistance, we generated radiation resistant cells using conventional (CF) and hypofractionated (HF) treatment schedules. We then evaluated the proteome of CF, HF and parental cells and discovered over 300 dysregulated proteins in the CF and HF cells relative to parental. Pathway analyses revealed upregulation of PI3K AkT mTOR signaling, oxidative phosphorylation, hypoxia, glycolysis, epithelial to mesenchymal transition (EMT), E2F targets and DNA repair in the radioresistant cells. We validated our proteomic findings that CD44 is enriched in CF and HF cells by western blot and flow cytometry. Finally, treatment with blocking anti-CD44 antibody promoted radiosensitization.

Quantitative characterization of protein N-linked core-fucosylation by an efficient glycan truncation strategy

Project description:We developed a truncation strategy which efficiently convert intact glycopeptides with hundreds of different glycans into two truncated forms, i.e. a monosaccharide HexNAc and a disaccharide HexNAc+core-Fucose. Further combination with data-independent analysis to form an integrated platform allowed the measurement of site-specific core-fucosylation abundances and the determination of CF occupancy ratio with high reproducibility. Notably, three times of CF sites were identified using this strategy compared to conventional methods based on intact glycopeptide.
