A Trial of MB-6 for Reduction of Neutropenia Induced by Chemotherapy in Patients With Stage III Colorectal Cancer.

ABSTRACT: This is a randomized, double-blind, placebo-controlled multi-center phaseⅢstudy to evaluate efficacy and safety of oxaliplatin-based chemotherapy plus MB-6(320 mg/capsule, 6 capsules tid) in patients with stage 3 colorectal cancer who underwent surgical excision of their primary tumor.

Project description:Cryptococcus neoformans is a fungal pathogen responsible for hundreds of thousands of deaths per year. Its critical virulence factor is a polysacharride capsule which grows large upon entry into a mammalian host. We previously identified USV101 as a transcription factor whose deletion results in enlarged capsules. Here, we characterize strains lacking or overexpressing USV101 in terms of their virulence-related phenotypes, the altered course of infection by them and immune response to them in a mouse model, the relationship of Usv101 to other transcription factors involved in capsule regulation, and the changes in Usv101 activity during capsule induction. To study the function of Usv101, a key C. neoformans regulator of virulence, and its interactions with downstream capsule-regulating TFs GAT201 and SP1, expression profiles were generated by RNA-seq of a usv101 deletion mutant, a USV101 overexpression mutant, strains expressing GAT201 under various promoters, various double mutants, and wildtype cells grown in a capsule non-inducing condition, a capsule inducing condition for 90 minutes, or a capsule inducing condition for 24 hours.

Project description:Since estrogen is a major hormone involved in multiple gene expression pathways in the cell, we wanted to determine the corresponding changes in trigeminal ganglia transcriptome of the E2-treated rats. Adult ovariectomized female rats were subcutaneneously implanted with either 5% (physiological level comparable to diestrus; 15-20 pg/ml; one capsule), 20% (physiological level comparable to proestrus; 40-60 pg/ml; 2 capsules of 10%), 40% (supraphysiological level; 2 capsules of 20%) E2, or control (2 capsules of 100% cholesterol) capsules. One week after implantation, nocifensive behaviors were recorded and trigeminal ganglia tissue was extracted.

Differential gene expression in the posterior joint capsule for immobility in mice

Project description:To explore the factors involved in arthrofibrosis, we performed a genome-wide screening for mRNA expressed in the posterior capsule after immobilization in mice. A gene ontology term analysis for differentially expressed genes after 2 weeks immobilization indicated that the genes involved in the extracellular region were particularly enriched. A heatmap demonstrated that joint immobilization significantly altered the expressions of genes related to the extracellular region. The MA plot indicated that, compared to naïve mice, 1654 genes were upregulated more than 2-fold, and 985 genes were downregulated more than 2-fold in immobilized mice. The MA plot and heatmap visually revealed dynamic changes in the expression of genes in immobilized posterior capsules compared to those in naïve posterior capsules. Our study represents the first detailed analysis of mouse joint capsule transcriptomes, with biologic replicates, generated by RNA-seq technology. RNA-seq based transcriptome characterization would expedite genetic network analyses and clarify Unresolved pathophysiology of joint contractures.
