Health Related Quality of Life Following Hepatectomy for Colorectal Liver Metastasis: Global and Disease Specific Changes Over Time

ABSTRACT: This study is being done to learn more about health related quality of life factors in people having surgery for colorectal liver metastasis. The investigators will look at how these factors may change over time. The information gained from this study will help the investigators to understand the long-term effects that cancer treatments have on the health related quality of life of patients. This information is of high value and will help doctors talk to patients about the possible effects of their operations. While many patients live a long time after such operations, the studies that have been done do not tell the full story of what patients go through after surgery. This study will help us to understand cancer treatment from the patient’s perspective. It will also help cancer patients make better decisions about their treatment options and will help them know what to expect after the operation.

Project description:We identify and correlate chromosomal alterations, methylation patterns and gene expression in pediatric pineal germinomas. Due the low incidence of pineal germinoma, there is insufficient molecular data related to chromosomal alterations, methylation patterns and genetic expression that may help understand the changes that influence tumor development. In addition, at present, the diagnosis of this tumor in childhood is challenging because of its location and cellular origin, which is why our aim was to understand and to identify possible molecular biomarkers to provide information on the diagnosis and tumor development. Tumor biopsies were obtained from pediatric patients of 0 to 16 years of age, they were subsequently evaluated by histopathological analysis. Sample selection was based on the percentage of tumor cells present in the tumor samples, processing the samples containing the lowest 70% and 80% tumor cells, respectively.

Quantitative Proteome Analysis Reveals Increased Content of Basement Membrane Proteins in Arteries from Patients with Type 2 Diabetes and Lower Levels among Metformin Users

Project description:We analyzed non-atherosclerotic repair arteries gathered at coronary by-pass operations from 30 patients with type 2 diabetes, as well as from 30 age- and gender-matched non-diabetic individuals. Quantitative proteome analysis was done by iTRAQ-labelling and LC-MS/MS analysis on individual arterial samples. The amounts of the basement membrane (BM) components, alpha-1- and alpha-2- type IV collagen, gamma-1- and beta-2-laminin were significantly increased in patients with diabetes. Moreover, the expressions of basement membrane components and other vascular proteins were significantly lower among metformin users, as compared to non-users. Patients treated with or without metformin had similar levels of HbA1c, cholesterol and blood pressure. In addition, quantitative histomorphometry showed increased area fractions of collagen-stainable material in tunica intima and media among patients with diabetes.

Information gathered from this study may help researchers better understand if ctDNA found in the blood can predict whether colorectal cancer may come back or spread.

A Retrospective and Cross-Sectional Analysis of Patients Treated for SCID Since January 1, 1968

Project description:<p>Individuals with a past diagnosis of severe combined immune deficiency (including many cases of "leaky SCID", Omenn syndrome, and reticular dysgenesis) who have undergone blood and marrow transplant, gene therapy, or enzyme replacement in the past may be eligible for 6902. The purpose of 6902 is very similar to 6901, except 6902 is looking backwards at what has already been done in the past (compared to 6901 which is looking into the future). Over 800 patients with SCID are expected to be enrolled on 6902. This makes 6902 the largest study ever to describe outcomes for patients with SCID treated at many different hospitals around North America. </p> <p>One of the most important components of the 6902 study is the "cross sectional" study. Patients who have received their treatments (BMT, gene therapy, enzyme replacement) many years ago are asked to come back to the hospital where they were treated. During this visit, additional research blood work is drawn and information is gathered regarding long-term transplant outcomes such as infections, graft-versus-host disease, autoimmune diseases, and quality of life. This will allow PIDTC researchers to better understand long-term outcomes from procedures that occurred many years ago (sometimes over 30 years ago) - something that is not possible at the present time with 6901. This will help researchers to best design new treatments and clinical trials in the future for children with SCID.</p>

Blood transcriptional profiles distinguish different clinical stages of cutaneous leishmaniasis in humans

Project description:Cutaneous leishmaniasis (CL) is prevalent in many low-income countries. Although CL is considered non-fatal, it has a major impact on public health systems of affected nations, causing social and economic problems due to disfiguration, morbidity, loss of productivity and medical costs for the public sector. Since no effective human vaccine against CL exists, it is crucial to understand better factors that may affect the outcome of the disease, including the impact of the pathogen on the host immune system. While transcriptomics analyses of the skin lesion in CL patients were recently reported, there is a dearth of information on the expression of immune-related genes in the blood of CL patients. We herein sought to profile the expression of immune-related genes in the blood of healed, asymptomatic and active CL patients.

Global profiling of microRNAs in peripheral blood and synovial fluid mononuclear cells of patients with Enthesitis Related Arthritis

Project description:Enthesitis related arthritis (ERA) is the most common category of juvenile idiopathic arthritis in Asia. MicroRNA dysregulation has been shown in chronic arthritides however there is no data in ERA. MicroRNA dysregulation in ERA may help understand the pathogenesis of disease. Thus we determined the difference in global miRNA expression in peripheral blood mononuclear cells (PBMCs) of patients with ERA (n=8) as compared to healthy controls (HC) (n=8) and further to understand the changes at the site of inflammation, the PBMCs profile was compared with the synovial fluid mononuclear cells (SFMCs) (n=8) of patients with ERA.
