A Clinical Study of Regorafenib in Participants Who Have Been Treated in Previous Bayer-sponsored Regorafenib Studies That Have Been Completed

ABSTRACT: The purpose of this study is to enable participants, currently receiving regorafenib in a Bayer-sponsored clinical trial, to continue treatment after their respective study has been closed. Patients participating in this study will be observed to collect information on how safe the drugs are and how this treatment is tolerated.

Project description:Here, we describe mutants of Mycoplasma pulmonis that were obtained using a minitransposon, Tn4001TF1, which actively transposes but is then unable to undergo subsequent excision events. Using Tn4001TF1, we disrupted 39 genes previously thought to be essential for growth. Thus, the number of genes required for growth has been overestimated. This study also revealed evidence of gene duplications in M. pulmonis and identified chromosome segregation proteins that are dispensable in mycoplasmas but essential in Bacillus subtilis.

Treating Parkinson's Disease with Antibodies: Previous Studies and Future Directions.

Project description:Parkinson's disease is a neurodegenerative disorder mainly characterized by the degeneration of dopaminergic neurons in the substantia nigra. Degenerating neurons contain abnormal aggregates called Lewy bodies, that are predominantly composed of the misfolded and/or mutated alpha-synuclein protein. Post-translational modifications, cellular stress, inflammation and gene mutations are thought to trigger its pathological misfolding and aggregation. With alpha-synuclein pathology being strongly associated with dopaminergic neuronal toxicity, strategies aimed to reduce its burden are expected to be beneficial in slowing disease progression. Moreover, multiple sources of evidence suggest a cell-to-cell transmission of pathological alpha-synuclein in a prion-like manner. Therefore, antibodies targeting extra- or intracellular alpha-synuclein could be efficient in limiting the aggregation and transmission. Several active and passive immunization strategies have been explored to target alpha-synuclein. Here, we summarize immunotherapeutic approaches that were tested in pre-clinical or clinical studies in the last two decades in an attempt to treat Parkinson's disease.

Transcription profiling of human samples from the MILE (Microarray Innovations In LEukemia) study pre-phase to investigate cross site reproducibility

Project description:An international standardization program towards the application of gene expression profiling in routine leukaemia diagnostics: The MILE study pre-phase. A total of 11 laboratories from three continents performed 204 analyses using a whole-genome expression microarray. In an effort to standardize the microarray procedure, each participating centre was equipped with identical hardware, software, and reagents. Designated study operators received intensive individual training on the sample preparation protocol. Experiment Overall Design: This study is part of the MILE Study (Microarray Innovations In LEukemia) program, headed by the European Leukaemia Network (ELN) and sponsored by Roche Molecular Systems, Inc. Experiment Overall Design: Each laboratory prepared two separate cell line total RNA samples, as well as three replicate leukaemia patient lysates in two distinct project stages: (i) a five-day course of protocol training, and (ii) independent proficiency testing. Experiment Overall Design: Using unsupervised, supervised, and rÂ² correlation analyses we demonstrated that microarray analysis can be performed not only with remarkably high intra-laboratory reproducibility but also with comparable quality and reliability between eleven distinct laboratories.
