ABSTRACT: This clinical trial is focused in the development of a screening test for the people at risk of colo-rectal cancer (aged more than 50 years old), valid and safe, improving the screening prognosis increasing the sensitivity and sensitive as compared with the current method, fecal occult blood.

Project description:To explore the effects of gut microbiota of young (8 weeks) or old mice (18～20 months) on stroke, feces of young (Y1-Y9) and old mice (O6-O16) were collected and analyzed by 16s rRNA sequencing. Then stroke model was established on young mouse receive feces from old mouse (DOT1-15) and young mouse receive feces from young mouse (DYT1-15). 16s rRNA sequencing were also performed for those young mice received feces from young and old mice.

Syringaresinol delays immunosenescence in aged mice: rejuvenating the immune system and reconstituting the gut microbiota

Project description:Improving immune function during aging contributes to the extension of healthspan. However, little is known about interventions for overcoming immunosenescence. Here, we investigated whether syringaresinol (SYR), an activator of FOXO3, overcomes immunosenescence and the factors associated with this effect. To address this, we administered SYR to 42-week-old mice for 10 weeks and analyzed immunological parameters and the gut microbiota. Compared to control mice, SYR-treated mice exhibited reversal of the age-related changes in lymphocyte subsets—such as CD3+ T, CD19+ B and Foxp3+ regulatory T (Treg) cells—and T-cell function in vitro. SYR induced the expression of Bim as well as the activation of FOXO3 in Tregs, which probably regulated Treg homeostasis. Furthermore, SYR reduced the serum level of lipopolysaccharide-binding protein, an inflammatory marker, and enriched the gut microbiota with beneficial bacteria, Lactobacillus and Bifidobacterium, effects that were closely associated with the changes in lymphocyte subsets. Finally, SYR enhanced humoral immunity against influenza vaccination to the level of young control mice. Collectively, these findings indicate that SYR may rejuvenate immunosenescence by enhancing the immune response and modulating the gut microbiota, possibly affecting systemic inflammation, although the precise mechanism awaits further study, and suggest SYR to be a potent candidate for anti-immunosenescence intervention.

Project description:The gut microbiota is a complex, densely populated community, home to many different species that collectively provide huge benefits for host health. Disruptions to this community, as can result from recurrent antibiotic exposure, alter the existing network of interactions between bacteria and can render this community susceptible to invading pathogens. Recent findings show that direct antagonistic and metabolic interactions play a critical role in shaping the microbiota. However, the part played by quorum sensing, a means of regulating bacterial behavior through secreted chemical signals, remains largely unknown. We have recently shown that the interspecies signal, autoinducer-2 (AI-2), can modulate the structure of the gut microbiota by using Escherichia coli to manipulate signal levels. Here, we discuss how AI-2 could influence bacterial behaviors to restore the balance between the 2 major bacteria phyla, the Bacteroidetes and Firmicutes, following antibiotic treatment. We explore how this may impact on host physiology, community susceptibility or resistance to pathogens, and the broader potential of AI-2 as a means to redress the imbalances in microbiota composition that feature in many infectious and non-infectious diseases.

Analysis of gut microbiota by pyrosequencing of 16S rRNA gene in F344 rats

Project description:Chronic acid suppression by proton pump inhibitor (PPI) has been hypothesized to alter the gut microbiota via a change in intestinal pH. To evaluate the changes in gut microbiota composition by long-term PPI treatment. Twenty-four week old F344 rats were fed with (n = 5) or without (n = 6) lansoprazole (PPI) for 50 weeks. Then, profiles of luminal microbiota in the terminal ileum were analyzed. Pyrosequencing for 16S rRNA gene was performed by genome sequencer FLX (454 Life Sciences/Roche) and analyzed by metagenomic bioinformatics.

Gut microbiota and IBD: causation or correlation?

Project description:A general consensus exists that IBD is associated with compositional and metabolic changes in the intestinal microbiota (dysbiosis). However, a direct causal relationship between dysbiosis and IBD has not been definitively established in humans. Findings from animal models have revealed diverse and context-specific roles of the gut microbiota in health and disease, ranging from protective to pro-inflammatory actions. Moreover, evidence from these experimental models suggest that although gut bacteria often drive immune activation, chronic inflammation in turn shapes the gut microbiota and contributes to dysbiosis. The purpose of this Review is to summarize current associations between IBD and dysbiosis, describe the role of the gut microbiota in the context of specific animal models of colitis, and discuss the potential role of microbiota-focused interventions in the treatment of human IBD. Ultimately, more studies will be needed to define host-microbial relationships relevant to human disease and amenable to therapeutic interventions.
