Effect of a new light and contrast mechanism on the detection of lesions in Endoscopic examination of the colon in Cancer screening procedures

ABSTRACT: Interventions: 200 patients will undergo diagnostic colonoscopy, prior to the examination all patients must ingest 1 liter of 10% mannitol solution on the day of the examination, preceded by a day with a diet of clear liquids without fibers, for intestinal cleaning. Patients will be randomized to one of the following groups: (White light image-WLI, blue light-BLI-bright, and Linked color Imaging-LCI). Project description:Interventions: 0.5 ml ICG was injected the mucosa into the submucosal space of the bowel wall at just the distal side of the tumor preoperative endoscopically. We identified the colonic tumor site in the PINPOINT Endoscopic Fluorescence Imaging System, in which NIR fluorescence is superimposed in pseudo-color (green) on a white light image. Gene expression analysis during time dependent Cry toxin exposure in Achaea janata larva.

Project description: Bacillus thuringiensis Cry toxin is used for insect pest management. When susceptible insect larvae ingest the Bt toxin it shows various physiological and cellular changes. We have employed Agilent One Color matrix based microarray to check the mid-gut gene expression upon Cry toxin exposure in Achaea janata (Castor semilooper). Microarray analysis shows significant variation in gene expression upon Cry toxin exposure.

Rapid identification of prognostic imaging biomarkers for non-small cell lung cancer by leveraging public gene expression microarray data

Project description:To rapidly identify new prognostic imaging biomarkers, we propose a bioinformatics approach that integrates gene expression and image data and leverages public gene expression data. We demonstrate our approach in non-small cell lung carcinoma patients for whom CT, PET/CT and gene expression data are available but without clinical follow-up. We extracted 180 image features and 56 high quality gene expression clusters, represented by metagenes. 115 image features were expressed in terms of metagenes, using sparse linear regression and cross-validation, with an accuracy of 65-86%. After mapping the signatures to a public gene expression dataset, 26 image features were significantly associated with recurrence-free survival and 22 with overall survival. A multivariate analysis identified multiple image features that were prognostic, independent of clinical covariates. Identifying prognostic imaging biomarkers by linking images and gene expression with outcomes in public gene expression datasets promises to accelerate the role of imaging in personalized medicine.

Transcription profiling of Streptococcus pneumoniae after incubation with host respiratory tract epithelial cells

Project description:In order to study pneumococcal gene expression changes after incubation with host epithelial cells, logarithmic-phase grown S. pneumoniae strains (82 and R6) were incubated with host epithelial cells (A549 and ETEC) in T75 cell culture flasks at an MOI range of 10:1 ~ 50:1. After incubation for 1 h or 3 h, bacterial solution was removed and 7 ml of host cell lysis buffer which can remove host cells and stablize bacterial RNA was added immediately. Bacteria incubated with antibiotic-free media (MEM or DMEM) with 1% (v/v) FBS for the same time periods were used as medium controls. The RNA samples were isolated with RiboPure Bacteria kit (Ambion) or RNeasy Minikit (Qiagen), and purified with DNase I treatment. cDNA samples were synthesized and labeled with biotin-dUTP (samples) and fluorescein-dUTP (Enzo) (medium controls), respectively, by using a LabelStar Array kit (Qiagen). Divergent labeled samples were hybridized to the same microarray slide by using a Two-Color Nucleic Acid Microarray Toolkit (Invitrogen). The scattered light signals of Resonance Light Scattering (RLS) particles Ag and Au were captured with a GSD-501 RLS imager using an ICS-501 (version 2.3) Image Capture System (Invitrogen). The gridding of spots was performed with ArrayVision 8.3 (Imaging Research).

Proteomic analysis of cerebrospinal fluid in epileptic and healthy dogs

Project description:This study included four dog groups (group A: 10 healthy dogs, group B: 9 dogs with idiopathic epilepsy receiving antiepileptic medication-AEM, group C: 8 dogs with idiopathic epilepsy without AEM administration and group D: 7 dogs with seizures due to structural brain abnormalities). The epileptic dogs were allocated into these 3 groups after a diagnostic investigation that included laboratory testing, thoracic radiographic and abdominal ultrasonographic examination, brain imaging, CSF routine and proteomic analysis. The purpose of the current study was to investigate and compare the proteomic profile among the four groups of dogs.
