Electronic Staining increases the Detection of Tumors of the Large Intestine and Rectum

ABSTRACT: Interventions: Group 1:151 patients will be submitted to colonoscopy with use of linked color (LCI); Group 2: 150 patients will be submitted to colonoscopy with use of blue laser brght-mode (BLI-bright) and; Group 3: 150 patients will undergo colonoscopy with white light (WLI). The electronic chromoscopy technique used at each colonoscopy (in each patient) will be determined according to the randomization.;Device;E01.370.372.250.250.200 Primary outcome(s): Adenoma detection rate, evaluated by the techniques of linked color imaging (LCI), white light imaging (WLI) and blue laser imaging (BLI-bright) and confirmed by a blinded pathologist within 7 days of the colonoscopy. Project description:Interventions: 200 patients will undergo diagnostic colonoscopy, prior to the examination all patients must ingest 1 liter of 10% mannitol solution on the day of the examination, preceded by a day with a diet of clear liquids without fibers, for intestinal cleaning. Using image enhancement in colonoscopy to increase detection of colonic polyps

Project description:Interventions: In this study, we aim to compare the adenoma detection rate with Linked Colour Imaging (LCI) in patients undergoing elective colonoscopy at Gold Coast University Hospital. Our overall objective is to demonstrate whether LCI increases the adenoma detection rate compared with high-definition white light (WL; current standard of care). All procedures will be performed by experienced colonoscopists or fellows already familiar with the Fujifilm colonoscopy system which has LCI in-built (ELUXEO, FUJIFILM Co., Tokyo, Japan) and accessible with the simple push of a button. The endoscopes are currently used for routine practice and procedures in our endoscopy unit (including both high-definition white light and LCI). Prior to enrolment of patients, study endoscopists’ baseline adenoma detection rates will be recorded based on data from 100 prior consecutive colonoscopies. Endoscopists will be classified as baseline expert or non-expert adenoma detectors and adenoma detection assessed both according to group and on an individual basis. Each participant will undergo a single colonoscopy. Participants will be randomised once to examination either under LCI or high-definition WL, with both insertion and withdrawal performed using the same modality. All patients will be sedated by anaesthetist-led propofol sedation as is our current practice. Polyps will be evaluated under Blue Light Imaging (BLI) for characteristic features. They will be removed as they are identified (via standard polypectomy techniques) and submitted for pathological examination. Pathology will be utilised to confirm the presence of polyp tissue. Adenoma Detection Rate of 3D Colonoscopy

Project description:Adenoma detection rate (ADR) has been the most important quality indicator on colonoscopy because ADR was reversely related with CRC incidence and mortality. Several image enhanced technologies, such as narrow-band imaging (NBI) or linked colour imaging (LCI) had been proved to have ability to increase the ADR. 3D techonology, however, has not been validated on colonoscopy performance. Therefore, current study was to compare the ADR between new 3D colonoscopy and conventional 2D colonoscopy.

Differentially expressed proteins wereassociated withtwo color morphs of Asian citrus psyllid, Diaphorina citri

Project description:Diaphorina citriis a vector of ‘CandidatusLiberibacter asiaticus,’(CLas), associated with citrus greening or Huanglongbing (HLB) disease in citrus. D. citriexhibits two group three different color morph variants, blueand non-blue (includes gray and yellow). Blue morphs have a greater capacity for long-distance flight while has lower efficiency in CLas transmission as compared to non-blue morphswhich influences disease epidemiology. In this study, wecompare the protein profilesof two-color morphsand evaluate the effect of CLas infection on abundance of pre-identified proteins in each color morphs. Our results showed that blue morphs have higher abundance of the immunity-associated proteins, while their abundance were upregulated dramatically in the non-blue morphcompared to blue morphin result of CLas infection. This observation proposes blue morph has higher immunity and CLas trigger the immunity in both morph while in non-blue’s response is late and non-effective.Further, challenging both color morphs byentomopathogenic fungi Beauveria bassiana, were showed significantly lower mortality in blue morph vs non-blue. Also, to test the effect of delayed response in CLas acquisition, we did CLas acquisition assay after priming immunity by feeding heat-killed Liberibacter crescens to both color-morphs. The obtained results were showed that priming immunity in non-blue morph can significantly decrease CLas acquisition as close as the level of CLas acquisition in blue morph. Taking all these in account propose that higher immunity in blue morph suppress CLas acquisition in a way that CLas acts as a pathogenand costly forACP. While in non-blue ACP, CLas triggers the immunity but the delayed immunity is not effective to block CLas from acquisition.

Gene expression analysis on Hydrangea macrophylla sepal pigmentation due to aluminum accumulation

Project description:Most blue color in flowers is due to anthocyanin, and considerable proportion of blue coloration can be attributed to metal-complexed anthocyanins. Recently, we reported vacuolar localized iron-transporter in blue petal cells of Tulipa gesneriana. However the mechanism of another metal ion transporters and subsequent flower color development has yet to be fully explored. In Hydrangea macrophylla, Al3+ is involved in blue coloration and the anthocyanin is formed Al3+-complex in vacuoles. To identify the molecular mechanism of blue coloration in hydrangea flowers, we tried to isolate the related genes transporting metal ion into vacuoles. From the sepal cDNA library we read the sequences of ca. 12000 genes, then a microarray analysis was carried out. From the sequences information, we chose several genes that might localize vacuolar membrane and transport Al3+. By using Al3+-sensitive yeast strain, we could identify the gene transporting Al3+ into vacuole. From the functional similarity and predicted localization, we could also identify the gene transporting Al3+ into cytosol. We will report the Al3+ mobilization from out of cell into vacuole in the sepal of Hydrangea macrophylla.

Project description:In this study, we collected the inner epidermis of umbrella from blue and brown R. esculenta for comparative metabolomic and proteomic analyses to reveal the blue body color formation. In summary, these results explored our knowledge of the blue body color formation of R. esculenta and provided valuable guidance to future studies of color breeding and blue pigment application in jellyfish.

Phototoxic effects of nonlinear optical microscopy on cell cycle, oxidative states, and gene expression

Project description:Nonlinear optical imaging modalities, such as stimulated Raman scattering (SRS) microscopy, use pulsed-laser excitation with high peak intensity that can perturb the native state of cells. In this study, we used bulk RNA sequencing, quantitative measurement of cell proliferation, and fluorescent measurement of the generation of reactive oxygen species (ROS) to assess phototoxic effects of near-IR pulsed laser radiation, at different time scales, for laser excitation settings relevant to SRS imaging. We define a range of laser excitation settings for which there was no significant change in mouse Neuro2A cells after laser exposure. This study provides guidance for imaging parameters that minimize photo-induced perturbations in SRS microscopy to ensure accurate interpretation of experiments with time-lapse imaging or with paired measurements of imaging and sequencing on the same cells.
